  • Integrative annotation of 21,037 human genes validated by full-length cDNA clones
  • 2004
  • Ingår i: PLoS biology. - : Public Library of Science (PLoS). - 1544-9173 .- 1545-7885. ; 2:6, s. 856-875
  • Tidskriftsartikel (refereegranskat)abstract
    • The human genome sequence defines our inherent biological potential; the realization of the biology encoded therein requires knowledge of the function of each gene. Currently, our knowledge in this area is still limited. Several lines of investigation have been used to elucidate the structure and function of the genes in the human genome. Even so, gene prediction remains a difficult task, as the varieties of transcripts of a gene may vary to a great extent. We thus performed an exhaustive integrative characterization of 41,118 full-length cDNAs that capture the gene transcripts as complete functional cassettes, providing an unequivocal report of structural and functional diversity at the gene level. Our international collaboration has validated 21,037 human gene candidates by analysis of high-quality full-length cDNA clones through curation using unified criteria. This led to the identification of 5,155 new gene candidates. It also manifested the most reliable way to control the quality of the cDNA clones. We have developed a human gene database, called the H-Invitational Database (H-InvDB; http://www.h-invitational.jp/). It provides the following: integrative annotation of human genes, description of gene structures, details of novel alternative splicing isoforms, non-protein-coding RNAs, functional domains, subcellular localizations, metabolic pathways, predictions of protein three-dimensional structure, mapping of known single nucleotide polymorphisms (SNPs), identification of polymorphic microsatellite repeats within human genes, and comparative results with mouse full-length cDNAs. The H-InvDB analysis has shown that up to 4% of the human genome sequence (National Center for Biotechnology Information build 34 assembly) may contain misassembled or missing regions. We found that 6.5% of the human gene candidates (1,377 loci) did not have a good protein-coding open reading frame, of which 296 loci are strong candidates for non-protein-coding RNA genes. In addition, among 72,027 uniquely mapped SNPs and insertions/deletions localized within human genes, 13,215 nonsynonymous SNPs, 315 nonsense SNPs, and 452 indels occurred in coding regions. Together with 25 polymorphic microsatellite repeats present in coding regions, they may alter protein structure, causing phenotypic effects or resulting in disease. The H-InvDB platform represents a substantial contribution to resources needed for the exploration of human biology and pathology.

  • Lifetime of the isomeric O-2(+) state in Be-12
  • 2007
  • Ingår i: Physics Letters. Section B: Nuclear, Elementary Particle and High-Energy Physics. - : Elsevier BV. - 0370-2693. ; 654:3-4, s. 87-91
  • Tidskriftsartikel (refereegranskat)abstract
    • Mean lifetime tau of the isomeric O-2(+) state in Be-12 has been determined by measuring decay spectra of delayed y-rays from stopped Be-12 2 nuclei produced by the projectile fragmentation of O-18 at 100 A MeV. A consistent value of tau = 331 +/- 12 ns was obtained from the time spectra of the E2 gamma decay to the 2(1)(+) state and the positron annihilation following the E0 decay to the ground state. Based on the observed branching ratio between the E2 and E0 decays, transition strengths of the two decay modes were deduced to be B(E2; O-2(+) -> 2(1)(+)) = 7.0 +/- 0.6 e(2) fm(4) and 2 1 1 (O-2(+) Sigma(i) e(i) r(i)(2) O-1(+)), = 0.87 +/- 0.03 e fM(2), respectively. • Study of high-spin states in 48Ca region induced by secondary fusion reactions
  • 2005
  • Konferensbidrag (refereegranskat)abstract
    • A new method of in-beam gamma-ray spectroscopy induced by secondary fusion reactions, 37P+9Be and 46Ar+9Be is presented. Low-energy secondary beams of 37P and 46Ar ions of ∼5 MeV/A were developed in order to induce fusion evaporation reactions. Excited states of nuclei in the vicinity of 48Ca, 49-52Ti and 46Ca, were studied by the method as well as β decay of the secondary beam 46Ar. Gamma-gamma coincidence and excitation function analysis were performed to study high-spin states of Ti isotopes.

  • Systematics of high-spin isomers in N=83 isotones and a high-spin isomer beam
  • 2002
  • Ingår i: European Physical Journal A. - 1434-6001 .- 1434-601X. ; 13:02-jan, s. 5-8
  • Tidskriftsartikel (refereegranskat)abstract
    • Isomers in N = 83 isotones of Z = 60 66 were studied systematically. Their spins and parities arc,49/2(+) and 27(+) for odd and odd-odd nuclei, respectively. Nearly constant excitation energies of these isomers indicated a decrease of a Z = 64 shell gap energy as Z decreases from 64 to 60 within the framework of a deformed independent-particle model (DIPM). Their configurations are [v(f(tau/2)h(9/2)i(13/2)), pi(h(11/2))(2)](49/2+) and [v(f(7/2)h(9/2)i(13/2)), pi(h(11/2))(2)(d(5/2))(-1)](27+) for odd and odd-odd nuclei, respectively. The shape of the yrast status changes suddenly at spin 49/2(odd) and 27(odd-odd) from a near spherical to an oblate shape. Transitions from isomers are highly hindered because of the shape changes. They may be categorized to be shape isomers. The development of a secondary beam produced by using these high-spin isomers is also described.

  • COPD exacerbations: the importance of a standard definition
  • 2004
  • Ingår i: Respiratory Medicine. - : Elsevier BV. - 1532-3064 .- 0954-6111. ; 98:2, s. 99-107
  • Tidskriftsartikel (refereegranskat)abstract
    • Efforts to assess the efficacy of new therapies in the treatment of acute exacerbations of chronic obstructive pulmonary disease (COPD) have been hampered by the lack of a widely agreed and consistently used definition. A variety of definitions have been used in clinical studies, based on changes in patient symptoms or the requirement for antibiotic therapy, oral steroids or hospitalisation. To date, none of these definitions have been assessed in detail for their reliability, responsiveness and validity determined. Considerable heterogeneity in the aetiology and manifestation of COPD exacerbations makes identification and quantification of defining symptoms extremely difficult. New approaches are therefore being sought with a view to identifying a serum or tissue marker that can be used as a valuable diagnostic tool Improvements in data recording will also contribute to the accuracy of data retrieval and assessment. if we are to progress to a level of sophistication seen in the diagnosis and management of other diseases, it is evident that considerable research efforts will be required to improve our understanding of COPD exacerbations and develop a standard definition for these events, thereby facilitating the assessment of therapeutic approaches.
