    • New variants in the SARS-CoV-2 pandemic are more contagious (Alpha/Delta), evade neutralizing antibodies (Beta), or both (Omicron). This poses a challenge in vaccine development according to WHO. We designed a more universal SARS-CoV-2 DNA vaccine containing receptor-binding domain loops from the huCoV-19/WH01, the Alpha, and the Beta variants, combined with the membrane and nucleoproteins. The vaccine induced spike antibodies crossreactive between huCoV-19/WH01, Beta, and Delta spike proteins that neutralized huCoV-19/WH01, Beta, Delta, and Omicron virus in vitro. The vaccine primed nucleoprotein-specific T cells, unlike spike-specific T cells, recognized Bat-CoV sequences. The vaccine protected mice carrying the human ACE2 receptor against lethal infection with the SARS-CoV-2 Beta variant. Interestingly, priming of cross-reactive nucleoprotein-specific T cells alone was 60% protective, verifying observations from humans that T cells protect against lethal disease. This SARS-CoV vaccine induces a uniquely broad and functional immunity that adds to currently used vaccines.

  • Design and Performance of Packet Retransmission Diversity Scheme for Wireless Networks

    • High data-rate wireless access systems are currently under discussion since the demand for wireless multimedia communication is rapidly increasing due to strong advances in wireless Internet services. Reliable high-speed data communications is one of the major challenges with regards to harsh conditions. With the necessity for high data rates, linear multi-level modulation schemes are becoming more and more important in wireless communication systems since they are bandwidth efficient. In this thesis protocols are designed and evaluated which improve system performance by combining ARQ-induced retransmissions through multipath channels in order to reduce the latency and improve the system throughput. The starting point is to show that the employment of simple packet combining schemes to wireless LANs such as IEEE 802.11, can achieve a considerable performance gain with the addition of only a small increase in complexity. A low-complexity method for enhancing and exploiting retransmission diversity by varying the bit-to-symbol mapping for each retransmission of a packet is evaluated. The selected mappings are chosen in order to maximize a bit log likelihood ratio (LLR) based metric. An ARQ scheme is also proposed, which combines retransmissions and bandwidth efficient multi-level modulation techniques with a change in the symbol mapping for every retransmission. The main idea behind this proposal is to take advantage of the extra dimension provided by a retransmission diversity scheme in improving the power efficiency if the used modulation without altering the diversity order of the system. Considering the overall scheme as one entity it is possible to obtain a transmission scheme able to perform very well in both additive Gaussian and fading channels without increase in receiver complexity. Finally, multiple antenna systems with ARQ functionality are evaluated. It has been established that using multiple antennas at both the transmitter and receiver along with intelligent signal design enables us to achieve better reliability as well as increase the transmission rate. In this thesis a space-time block coded hybrid ARQ scheme is considered which exploits both the spatial and time diversity of the MIMO channel. A bit-to-symbol mapping ARQ scheme suitable for multiple antenna systems is also considered.

    • BACKGROUND: Subendothelial deposited low-density lipoprotein particles are a known inflammatory factor in atherosclerosis. However, the causal components derived from low-density lipoprotein are still poorly defined. Apolipoprotein B100 (ApoB100) is the unexchangeable protein component of low-density lipoprotein, and the progression of atherosclerosis is associated with immune responses to ApoB100-derived peptides. In this study, we analyzed the proinflammatory activity of ApoB100 peptides in atherosclerosis. METHODS AND RESULTS: By screening a peptide library of ApoB100, we identified a distinct native peptide referred to as ApoB100 danger-associated signal 1 (ApoBDS-1), which shows sequence-specific bioactivity in stimulation of interleukin-8, CCL2, and interleukin-6. ApoBDS-1 activates mitogen-activated protein kinase and calcium signaling, thereby effecting the expression of interleukin-8 in innate immune cells. Ex vivo stimulation of carotid plaques with ApoBDS-1 enhances interleukin-8 and prostaglandin E release. Furthermore, we demonstrated that ApoBDS-1-positive peptide fragments are present in atherosclerotic lesions using immunoassays and that low-molecular-weight fractions isolated from plaque show ApoBDS-1 activity inducing interleukin-8 production. CONCLUSIONS: Our data show that ApoBDS-1 is a previously unrecognized peptide with robust proinflammatory activity, contributing to the disease-promoting effects of low-density lipoprotein in the pathogenesis of atherosclerosis.

  • Simple interference detection and classification for industrial Wireless Sensor Networks

    • Wireless Sensor Networks (WSNs) are increasingly deployed in office blocks, residential areas and also industrial locations, thanks to advantages in terms of flexibility and scalability. Nowadays available wireless fieldbuses are widely adopted for process monitoring and offer performance comparable with the wired counterparts, despite they still are more sensitive to interference from external sources. This work investigates the main sources of interference in the 2.4 GHz ISMband and evaluates the adoption of a simple algorithm to identify the interference. The proposed technique, called LPED, is based on bit error nature and forward error correction. The required computational effort is compatible with resources normally available in WSN nodes, as experimentally verified. In addition, performance in presence of IEEE802.11 and iWLAN is also verified; classification is correct in about 90% of cases.

    • The sudden infant death syndrome (SIDS) is a major diagnosis of post-neonatal mortality in the industrialised countries. The cause of death is, however, still elusive. The development of complementary diagnostic methods widening the possibility to analyse the postmortem findings is therefore important. The aim of this study was to histologically examine the enamel of teeth from children dying suddenly and unexpectedly in order to disclose if the presence of enamel disturbances corresponded to neo- or postnatal disorders. During enamel formation ameloblasts are susceptible to local and systemic disturbances which may be reflected in the fully formed enamel as hypoplasia, hypocalcification or accentuated incremental lines. Nineteen consecutive cases of deaths in infants between 1-19 months of age were investigated at the Department of Forensic Pathology, Stockholm, Sweden. The analyses of anamnestic and postmortem data and the analyses of incremental lines were done independently without knowledge of the conclusion of the other part. The results show a good agreement between enamel changes on one hand and the antemortem history and the autopsy findings on the other. This type of investigation may be a valuable complement to the ordinary postmortem investigations of a child who has suddenly and unexpectedly died.
