  • Novel genes and sex differences in COVID-19 severity

    • Here, we describe the results of a genome-wide study conducted in 11 939 coronavirus disease 2019 (COVID-19) positive cases with an extensive clinical information that were recruited from 34 hospitals across Spain (SCOURGE consortium). In sex-disaggregated genome-wide association studies for COVID-19 hospitalization, genome-wide significance (P < 5 × 10−8) was crossed for variants in 3p21.31 and 21q22.11 loci only among males (P = 1.3 × 10−22 and P = 8.1 × 10−12, respectively), and for variants in 9q21.32 near TLE1 only among females (P = 4.4 × 10−8). In a second phase, results were combined with an independent Spanish cohort (1598 COVID-19 cases and 1068 population controls), revealing in the overall analysis two novel risk loci in 9p13.3 and 19q13.12, with fine-mapping prioritized variants functionally associated with AQP3 (P = 2.7 × 10−8) and ARHGAP33 (P = 1.3 × 10−8), respectively. The meta-analysis of both phases with four European studies stratified by sex from the Host Genetics Initiative (HGI) confirmed the association of the 3p21.31 and 21q22.11 loci predominantly in males and replicated a recently reported variant in 11p13 (ELF5, P = 4.1 × 10−8). Six of the COVID-19 HGI discovered loci were replicated and an HGI-based genetic risk score predicted the severity strata in SCOURGE. We also found more SNP-heritability and larger heritability differences by age (<60 or ≥60 years) among males than among females. Parallel genome-wide screening of inbreeding depression in SCOURGE also showed an effect of homozygosity in COVID-19 hospitalization and severity and this effect was stronger among older males. In summary, new candidate genes for COVID-19 severity and evidence supporting genetic disparities among sexes are provided.

    • The drive to withstand environmental stresses and defend against invasion is a universal trait extant in all forms of life. While numerous canonical signaling cascades have been characterized in detail, it remains unclear how these pathways interface to generate coordinated responses to diverse stimuli. To dissect these connections, we followed heparanase (HPSE), a protein best known for its endoglycosidic activity at the extracellular matrix but recently recognized to drive various forms of late-stage disease through unknown mechanisms. Using herpes simplex virus-1 (HSV-1) infection as a model cellular perturbation, we demonstrate that HPSE acts beyond its established enzymatic role to restrict multiple forms of cell-intrinsic defense and facilitate host cell reprogramming by the invading pathogen. We reveal that cells devoid of HPSE are innately resistant to infection and counteract viral takeover through multiple amplified defense mechanisms. With a unique grasp of the fundamental processes of transcriptional regulation and cell death, HPSE represents a potent cellular intersection with broad therapeutic potential.

    • We present an analysis of the first space-based far-IR-submm observations of M33, which measure the emission from the cool dust and resolve the giant molecular cloud complexes. With roughly half-solar abundances, M33 is a first step towards young low-metallicity galaxies where the submm may be able to provide an alternative to CO mapping to measure their H-2 content. In this Letter, we measure the dust emission cross-section sigma using SPIRE and recent CO and HI observations; a variation in s is present from a near-solar neighborhood cross-section to about half-solar with the maximum being south of the nucleus. Calculating the total H column density from the measured dust temperature and cross-section, and then subtracting the HI column, yields a morphology similar to that observed in CO. The H-2/HI mass ratio decreases from about unity to well below 10% and is about 15% averaged over the optical disk. The single most important observation to reduce the potentially large systematic errors is to complete the CO mapping of M33.

    • Aims. We aim to better understand the emission of molecular tracers of the diffuse and dense gas in giant molecular clouds and the influence that metallicity, optical extinction, density, far-UV field, and star formation rate have on these tracers.Methods. Using the IRAM 30 m telescope, we detected HCN, HCO+, 12CO, and 13CO in six GMCs along the major axis of M 33 at a resolution of ~114 pc and out to a radial distance of 3.4 kpc. Optical, far-infrared, and submillimeter data from Herschel and other observatories complement these observations. To interpret the observed molecular line emission, we created two grids of models of photon-dominated regions, one for solar and one for M 33-type subsolar metallicity.Results. The observed HCO+/HCN line ratios range between 1.1 and 2.5. Similarly high ratios have been observed in the Large Magellanic Cloud. The HCN/CO ratio varies between 0.4% and 2.9% in the disk of M 33. The 12CO/13CO line ratio varies between 9 and 15 similar to variations found in the diffuse gas and the centers of GMCs of the Milky Way. Stacking of all spectra allowed HNC and C2H to be detected. The resulting HCO+/HNC and HCN/HNC ratios of ~8 and 6, respectively, lie at the high end of ratios observed in a large set of (ultra-)luminous infrared galaxies. HCN abundances are lower in the subsolar metallicity PDR models, while HCO+ abundances are enhanced. For HCN this effect is more pronounced at low optical extinctions. The observed HCO+/HCN and HCN/CO line ratios are naturally explained by subsolar PDR models of low optical extinctions between 4 and 10 mag and of moderate densities of n 3 × 103–3 × 104 cm-3, while the FUV field strength only has a small effect on the modeled line ratios. The line ratios are almost equally well reproduced by the solar-metallicity models, indicating that variations in metallicity only play a minor role in influencing these line ratios.

  • Biocomposites from Rice Straw Nanofibers: Morphology, Thermal and Mechanical Properties

    • Agricultural residues are major potential resources for biomass and for material production. In this work, rice straw residues were used to isolate cellulose nanofibers of different degree of oxidation. Firstly, bleached rice fibers were produced from the rice straw residues following chemical extraction and bleaching processes. Oxidation of rice fibers mediated by radical 2,2,6,6-tetramethylpiperidine 1-oxyl (TEMPO) at pH 10 was then applied to extract rice cellulose nanofibers, with diameters of 3-11 nm from morphological analysis. The strengthening capacity of rice nanofibers was tested by casting nanocomposite films with poly(vinyl alcohol) polymer. The same formulations with eucalyptus nanofibers were produced as comparison. Their thermal and mechanical performance was evaluated using thermogravimetry, differential scanning calorimetry, dynamic mechanical analysis and tensile testing. The glass transition of nanocomposites was shifted to higher temperatures with respect to the pure polymer by the addition of rice cellulose nanofibers. Rice nanofibers also acted as a nucleating agent for the polymer matrix. More flexible eucalyptus nanofibers did not show these two phenomena on the matrix. Instead, both types of nanofibers gave similar stiffening (as Young's modulus) to the matrix reinforced up to 5 wt.%. The ultimate tensile strength of nanocomposite films revealed significant enhancing capacity for rice nanofibers, although this effect was somehow higher for eucalyptus nanofibers.

    • The morphological properties of cellulose nanofibrils obtained from eucalyptus pulp fibres were assessed. Two samples were produced with the same chemical treatment (NaClO/NaBr/TEMPO (2,2,6,6-tetramethylpiperidine-1-oxyl radical) oxidation), but distinct mechanical treatment intensities during homogenization. It was shown that the nanofibrils production yield increases with the mechanical energy. The effect of mechanical treatment on the yield was confirmed by laser profilometry of air-dried nanocellulose films. However, no significant differences were detected regarding the nanofibrils width as measured by atomic force microscopy (AFM) of air-dried films. On the other hand, differences in size were found either by laser diffraction spectroscopy or by dynamic light scattering (DLS) of the cellulose nanofibrils suspensions as a consequence of the differences in the length distribution of both samples. The nanofibrils length of the more nanofibrillated sample was calculated based on the width measured by AFM and the hydrodynamic diameter obtained by DLS. A length value of ca. 600. nm was estimated. The DLS hydrodynamic diameter, as an equivalent spherical diameter, was used to estimate the nanofibrils length assuming a cylinder with the same volume and with the diameter (width) assessed by AFM. A simple method is thus proposed to evaluate the cellulose nanofibrils length combining microscopy and light scattering methods.

    • In the present work the evolution of physical and mechanical properties of papers and nanopapers is studied. Handsheets made of eucalyptus fibres reinforced with 0, 25, 50, 75 and 100 wt% of nanofibrillated cellulose (NFC) content were fabricated using a Rapid Köthen-like equipment. The obtained papers and nanopapers were physical- and mechanically-characterized. The results showed a significant increase in density and a reduction of porosity in the samples during their transition from paper to nanopaper; besides, nanopapers were more transparent and smoother than normal papers. These physical changes where more evident with increasing amounts of NFC. Regarding mechanical properties, nanopapers with a 100 wt% content of NFC improved their strength and rigidity in 228 and 317 %, respectively, in comparison with normal papers. The evolution of strength and rigidity from paper to nanopaper was linear in relation to the amount of NFC, which means that the ultimate tensile strength was mainly dependant on nanofibril failure.

    • The dissociative electron-attachment (DEA) phenomena at the N-1-H and N-3-H bonds observed experimentally at low energies (<3 eV) in uracil are studied with the CASSCF/CASPT2 methodology. Two valence bound pi(-) and two dissociative sigma(-) states of the uracil anionic species, together with the ground state of the neutral molecule, are proven to contribute to the shapes appearing in the experimental DEA cross sections. Conical intersections (CI) between the pi(-) and sigma(-) are established as the structures which activate the DEA processes. The N-1-H and N-3-H DEA mechanisms in uracil are described, and experimental observations are interpreted on the basis of two factors: (1) the relative energy of the (U-H)(-) + H fragments obtained after DEA with respect to the ground-state equilibrium structure (S-0) of the neutral molecule (threshold for DEA) and (2) the relative energy of the CIs also with respect to S-0 (band maxima). The pi(-)(1) state is found to be mainly responsible for the N-1-H bond breaking, whereas the pi(-)(2) state is proved to be involved in the cleavage of the N-3-H bond.
