  • Multi-ancestry genetic study of type 2 diabetes highlights the power of diverse populations for discovery and translation

    • We assembled an ancestrally diverse collection of genome-wide association studies (GWAS) of type 2 diabetes (T2D) in 180,834 affected individuals and 1,159,055 controls (48.9% non-European descent) through the Diabetes Meta-Analysis of Trans-Ethnic association studies (DIAMANTE) Consortium. Multi-ancestry GWAS meta-analysis identified 237 loci attaining stringent genome-wide significance (P < 5 x 10(-9)), which were delineated to 338 distinct association signals. Fine-mapping of these signals was enhanced by the increased sample size and expanded population diversity of the multi-ancestry meta-analysis, which localized 54.4% of T2D associations to a single variant with >50% posterior probability. This improved fine-mapping enabled systematic assessment of candidate causal genes and molecular mechanisms through which T2D associations are mediated, laying the foundations for functional investigations. Multi-ancestry genetic risk scores enhanced transferability of T2D prediction across diverse populations. Our study provides a step toward more effective clinical translation of T2D GWAS to improve global health for all, irrespective of genetic background. Genome-wide association and fine-mapping analyses in ancestrally diverse populations implicate candidate causal genes and mechanisms underlying type 2 diabetes. Trans-ancestry genetic risk scores enhance transferability across populations.

  • Impact of common genetic determinants of Hemoglobin A1c on type 2 diabetes risk and diagnosis in ancestrally diverse populations : A transethnic genome-wide meta-analysis

    • Background: Glycated hemoglobin (HbA1c) is used to diagnose type 2 diabetes (T2D) and assess glycemic control in patients with diabetes. Previous genome-wide association studies (GWAS) have identified 18 HbA1c-associated genetic variants. These variants proved to be classifiable by their likely biological action as erythrocytic (also associated with erythrocyte traits) or glycemic (associated with other glucose-related traits). In this study, we tested the hypotheses that, in a very large scale GWAS, we would identify more genetic variants associated with HbA1c and that HbA1c variants implicated in erythrocytic biology would affect the diagnostic accuracy of HbA1c. We therefore expanded the number of HbA1c-associated loci and tested the effect of genetic risk-scores comprised of erythrocytic or glycemic variants on incident diabetes prediction and on prevalent diabetes screening performance. Throughout this multiancestry study, we kept a focus on interancestry differences in HbA1c genetics performance that might influence race-ancestry differences in health outcomes.Methods & findings: Using genome-wide association meta-analyses in up to 159,940 individuals from 82 cohorts of European, African, East Asian, and South Asian ancestry, we identified 60 common genetic variants associated with HbA1c. We classified variants as implicated in glycemic, erythrocytic, or unclassified biology and tested whether additive genetic scores of erythrocytic variants (GS-E) or glycemic variants (GS-G) were associated with higher T2D incidence in multiethnic longitudinal cohorts (N = 33,241). Nineteen glycemic and 22 erythrocytic variants were associated with HbA1c at genome-wide significance. GS-G was associated with higher T2D risk (incidence OR = 1.05, 95% CI 1.04-1.06, per HbA1c-raising allele, p = 3 x 10-29); whereas GS-E was not (OR = 1.00, 95% CI 0.99-1.01, p = 0.60). In Europeans and Asians, erythrocytic variants in aggregate had only modest effects on the diagnostic accuracy of HbA1c. Yet, in African Americans, the X-linked G6PD G202A variant (T-allele frequency 11%) was associated with an absolute decrease in HbA1c of 0.81%-units (95% CI 0.66-0.96) per allele in hemizygous men, and 0.68%-units (95% CI 0.38-0.97) in homozygous women. The G6PD variant may cause approximately 2% (N = 0.65 million, 95% CI0.55-0.74) of African American adults with T2Dto remain undiagnosed when screened with HbA1c. Limitations include the smaller sample sizes for non-European ancestries and the inability to classify approximately one-third of the variants. Further studies in large multiethnic cohorts with HbA1c, glycemic, and erythrocytic traits are required to better determine the biological action of the unclassified variants.Conclusions: As G6PD deficiency can be clinically silent until illness strikes, we recommend investigation of the possible benefits of screening for the G6PD genotype along with using HbA1c to diagnose T2D in populations of African ancestry or groups where G6PD deficiency is common. Screening with direct glucose measurements, or genetically-informed HbA1c diagnostic thresholds in people with G6PD deficiency, may be required to avoid missed or delayed diagnoses.

    • There is a growing interest in analysing freshness of data in networked systems. Age of Information (AoI) has emerged as a relevant metric to quantify this freshness at a receiver, and minimizing this metric for different system models has received significant research attention. However, a fundamental question remains: what is the minimum achievable AoI in any single-server-single-source queuing system for a given service-time distribution? We address this question for the average peak AoI (PAoI) statistic by considering generate-at-will source model, service preemptions, and request delays. Our main result is on the characterization of the minimum achievable average PAoI, and we show that it is achieved by a fixed-threshold policy among the set of all causal policies. We use the characterization to provide necessary and sufficient condition for preemptions to be beneficial for a given service-time distribution. Our numerical results, obtained using well-known distributions, demonstrate that the heavier the tail of a distribution the higher the performance gains of using preemptions.

    • There is a growing interest in analysing the freshness of data in networked systems. Age of Information (AoI) has emerged as a popular metric to quantify this freshness at a given destination. There has been a significant research effort in optimizing this metric in communication and networking systems under different settings. In contrast to previous works, we are interested in a fundamental question, what is the minimum achievable AoI in any single-server-single-source queuing system for a given service-time distribution? To address this question, we study a problem of optimizing AoI under service preemptions. Our main result is on the characterization of the minimum achievable average peak AoI (PAoI). We obtain this result by showing that a fixed-threshold policy is optimal in the set of all randomized-threshold causal policies. We use the characterization to provide necessary and sufficient conditions for the service-time distributions under which preemptions are beneficial.

    • The abundance of spectrum in the millimeter-wave (mm-wave) bands makes it an attractive alternative for future wireless communication systems. Such systems are expected to provide data transmission rates in the order of multi-gigabits per second in order to satisfy the ever-increasing demand for high rate data communication. Unfortunately, mm-wave radio is subject to severe path loss, which limits its usability for long-range outdoor communication. In this paper, we propose a multi-hop mm-wave wireless network for outdoor communication, where multiple full-duplex buffered relays are used to extend the communication range, while providing end-to-end performance guarantees to the traffic traversing the network. We provide a cumulative service process characterization for the mm-wave propagation channel with self-interference in terms of the moment generating function of its channel capacity. Then, we then use this characterization to compute probabilistic upper bounds on the overall network performance, i.e., total backlog and end-to-end delay. Furthermore, we study the effect of self-interference on the network performance and propose an optimal power allocation scheme to mitigate its impact in order to enhance network performance. Finally, we investigate the relation between relay density and network performance under a sum power constraint. We show that increasing relay density may have adverse effects on network performance, unless the selfinterference can be kept sufficiently small.

    • In wireless local area networks, often a station can potentially associate with more than one access point (AP). Therefore, a relevant question is which AP to select 'best' from a list of candidate ones. In IEEE 802.11, the user simply associates to the AP with the strongest received signal strength. However, this may result in a significant load imbalance between several APs. Moreover, the multi-rate flexibility provided by several IEEE 802.11 variants can cause low bit rate stations to negatively affect high bit rate ones and consequently degrade the overall network throughput. This paper investigates the various aspects of 'best' AP selection for IEEE 802.11 systems. In detail, we first derive a new decision metric which can be used for AP selection. Using this metric, we propose two new selection mechanisms which are decentralised in the sense that the decision is performed by each station, given appropriate status information of each AP. In fact, only few bytes of status information have to be added to the Beacon and Probe Response frames which does not impose significant overhead. We show that our mechanism improves mean quality of service of all stations and better utilises network resources compared to the conventional one implemented today in IEEE 802.11 devices. Also, the schemes are appealing in terms of stability and provide their performance improvement even for denser or lighter network configurations.

    • This paper presents and evaluates an Inter-Access Point Coordination protocol for dynamic channel selection in IEEE 802.11 WLANs. It addresses an open issue for the implementation of many distributed and centralized dynamic channel selection policies proposed to mitigate interference problems in Wireless LANs (WLANs). The presented protocol provides services to a wide range of policies that require different levels of coordination among APs by enabling them to actively communicate and exchange information. An Intra-Cell protocol that enables interaction between the AP and its accommodated stations to handle channel switching within the same cell is also presented.

    • Resource-constrained Edge Devices (EDs), e.g., IoT sensors and microcontroller units, are expected to make intelligent decisions using Deep Learning (DL) inference at the edge of the network. Toward this end, developing tinyML models is an area of active research - DL models with reduced computation and memory storage requirements - that can be embedded on these devices. However, tinyML models have lower inference accuracy. On a different front, DNN partitioning and inference offloading techniques were studied for distributed DL inference between EDs and Edge Servers (ESs). In this paper, we explore Hierarchical Inference (HI), a novel approach proposed in [19] for performing distributed DL inference at the edge. Under HI, for each data sample, an ED first uses a local algorithm (e.g., a tinyML model) for inference. Depending on the application, if the inference provided by the local algorithm is incorrect or further assistance is required from large DL models on edge or cloud, only then the ED offloads the data sample. At the outset, HI seems infeasible as the ED, in general, cannot know if the local inference is sufficient or not. Nevertheless, we present the feasibility of implementing HI for image classification applications. We demonstrate its benefits using quantitative analysis and show that HI provides a better trade-off between offloading cost, throughput, and inference accuracy compared to alternate approaches.

    • Since its first release in the late 1990s, Wi-Fi has been updated to keep up with evolving user needs. Recently, Wi-Fi and other radio access technologies have been pushed to their edge when serving Augmented Reality (AR) applications. AR applications require high throughput, low latency, and high reliability to ensure a high-quality user experience. The 802.11be amendment - which will be marketed as Wi-Fi 7 - introduces several features that aim to enhance its capabilities to support challenging applications like AR. One of the main features introduced in this amendment is Multi-Link Operation (MLO) which allows nodes to transmit and receive over multiple links concurrently. When using MLO, traffic is distributed among links using an implementation-specific traffic-to-link allocation policy. This paper aims to evaluate the performance of MLO, using different policies, in serving AR applications compared to Single-Link (SL). Experimental simulations using an event-based Wi-Fi simulator have been conducted. Our results show the general superiority of MLO when serving AR applications. MLO achieves lower latency and serves a higher number of AR users compared to SL with the same frequency resources. In addition, increasing the number of links can improve the performance of MLO. Regarding traffic-to-link allocation policies, we found that policies can be more susceptible to channel blocking, resulting in possible performance degradation.
