    • Previously, we reported germline DNA variants associated with risk of urinary bladder cancer (UBC) in Dutch and Icelandic subjects. Here we expanded the Icelandic sample set and tested the top 20 markers from the combined analysis in several European case-control sample sets, with a total of 4,739 cases and 45,549 controls. The T allele of rs798766 on 4p16.3 was found to associate with UBC (odds ratio = 1.24, P = 9.9 x 10(-12)). rs798766 is located in an intron of TACC3, 70 kb from FGFR3, which often harbors activating somatic mutations in low-grade, noninvasive UBC. Notably, rs798766[T] shows stronger association with low-grade and low-stage UBC than with more aggressive forms of the disease and is associated with higher risk of recurrence in low-grade stage Ta tumors. The frequency of rs798766[T] is higher in Ta tumors that carry an activating mutation in FGFR3 than in Ta tumors with wild-type FGFR3. Our results show a link between germline variants, somatic mutations of FGFR3 and risk of UBC.

    • Three genome-wide association studies in Europe and the USA have reported eight urinary bladder cancer (UBC) susceptibility loci. Using extended case and control series and 1000 Genomes imputations of 5 340 737 single-nucleotide polymorphisms (SNPs), we searched for additional loci in the European GWAS. The discovery sample set consisted of 1631 cases and 3822 controls from the Netherlands and 603 cases and 37 781 controls from Iceland. For follow-up, we used 3790 cases and 7507 controls from 13 sample sets of European and Iranian ancestry. Based on the discovery analysis, we followed up signals in the urea transporter (UT) gene SLC14A. The strongest signal at this locus was represented by a SNP in intron 3, rs17674580, that reached genome-wide significance in the overall analysis of the discovery and follow-up groups: odds ratio = 1.17, P = 7.6 × 10(-11). SLC14A1 codes for UTs that define the Kidd blood group and are crucial for the maintenance of a constant urea concentration gradient in the renal medulla and, through this, the kidney's ability to concentrate urine. It is speculated that rs17674580, or other sequence variants in LD with it, indirectly modifies UBC risk by affecting urine production. If confirmed, this would support the 'urogenous contact hypothesis' that urine production and voiding frequency modify the risk of UBC.

    • The surgical treatment of spinal metastases is evolving. The major problem is the selection of patients who may benefit from surgical treatment. One of the criteria is an expected survival of at least 3 months. A prediction model has been previously developed. The present study has been performed in order to validate externally the model and to demonstrate that this model can be generalized to other institutions and other countries than the Netherlands. Data of 356 patients from five centers in Germany, Spain, Sweden, and the Netherlands who were treated for metastatic epidural spinal cord compression were collected. Hazard ratios in the test population corresponded with those of the developmental population. However, the observed and the expected survival were different. Analysis revealed that the baseline hazard function was significantly different. This tempted us to combine the data and develop a new prediction model. Estimating iteratively, a baseline hazard was composed. An adapted prediction model is presented. External validation of a prediction model revealed a difference in expected survival, although the relative contribution of the specific hazard ratios was the same as in the developmental population. This study emphasized the need to check the baseline hazard function in external validation. A new model has been developed using an estimated baseline hazard.

  • Dutch tenants at risk of eviction : Identifying predictors of eviction orders

    • In order to prevent evictions, it is important to gain more insight into factors predicting whether or not tenants receive an eviction order. In this study, ten potential risk factors for evictions were tested. Tenants who were at risk of eviction due to rent arrears in five Dutch cities were interviewed using a structured questionnaire, and six months later their housing associations were asked to provide information about the tenants' current situation. Multiple logistic regression analyses with data on 344 tenants revealed that the amount of rent arrears was a strong predictor for receiving an eviction order. Furthermore, single tenants and tenants who had already been summoned to appear in court were more likely to receive an eviction order. These results can contribute to identifying households at risk of eviction at an early stage, and to develop targeted interventions to prevent evictions.

    • Purpose: Stripping contaminants from sediments with granular activated carbon (GAC) is a promising remediation technique in which the effectiveness depends on the rate of contaminant extraction from the sediment by the GAC. The purpose of the present study was to investigate the effect of mixing intensity on the short-term extraction rate of polycyclic aromatic hydrocarbons (PAHs) from contaminated sediment.Materials and methods: PAH desorption from sediment at a wide range of rotational speeds (min−1; rotations per minute (rpm)) was monitored by uptake in Tenax polymeric resins using a completely mixed batch reactor. Desorption data were interpreted using a radial diffusion model. Desorption parameters obtained with the radial diffusion model were correlated with particle size measurements and interpreted mechanistically.Results and discussion: Fast desorption rate constants, De/r2, with De the effective diffusion coefficient and r the particle radius, ranged from 3.7 × 10−3 to 1.1 × 10−1 day−1 (PHE) and 6 × 10−6 to 1.9 × 10−4 day−1(CHR), respectively, and increased with the intensity of mixing. The De/r2 values would correspond to De ranges of 1.8 × 10−14–1.2 × 10−16 m2 × day−1 and 1.8 × 10−12–3.7 × 10−15 m2 × day−1, assuming fast desorption from the measured smallest particle size (9 μm) classes at 200 and 600 rpm, respectively.Conclusions: Desorption of PAHs was significantly accelerated by a reduction of particle aggregate size caused by shear forces that were induced by mixing. The effective intra-particle diffusion coefficients, De, were larger at higher mixing rates.
