    • The liver-specific organic cation transport protein (OCT1; SLC22A1) transports several cationic drugs including the antidiabetic drug metformin and the anticancer agents oxaliplatin and imatinib. In this study, we explored the chemical space of registered oral drugs with the aim of studying the inhibition pattern of OCT1 and of developing predictive computational models of OCT1 inhibition. In total, 191 structurally diverse compounds were examined in HEK293-OCT1 cells. The assay identified 47 novel inhibitors and confirmed 15 previously known inhibitors. The enrichment of OCT1 inhibitors was seen in several drug classes including antidepressants. High lipophilicity and a positive net charge were found to be the key physicochemical properties for OCT1 inhibition, whereas a high molecular dipole moment and many hydrogen bonds were negatively correlated to OCT1 inhibition. The data were used to generate OPLS-DA models for OCT1 inhibitors; the final model correctly predicted 82% of the inhibitors and 88% of the noninhibitors of the test set.

Vi föreslår därför ett ”utsläppsmoratorium”: inga beslut får tas som ökar utsläppen av växthusgaser. Principen måste kopplas till mål om exempelvis förnybar energi och grön infrastruktur, skriver 23 forskare och debattörer.

We assayed the components of the uPA system in homogenates of 64 primary epithelial ovarian tumors and 5 metastases and evaluated the association of these parameters to prognosis in the 51 malignant cases. The levels of uPA, PAI-2 and the uPA: PAI-I complex increased with progressive loss of histological differentiation (P-trend <0.001, <0.05 and <0.001). The level of PAI-I was higher in poorly than in well/moderately differentiated tumors (p = 0.03). The content of uPAR was lower in benign tumors as compared to borderline malignancies (p = 0.002), invasive primary tumors (p < 0.001), and metastases (p = 0.002). Surprisingly, the level of uPAR was lower in poorly differentiated as compared to both borderline (p = 0.01) and well differentiated malignant tumors (p = 0.005). Also, the level of uPAR was lower in advanced as compared to early stages of the disease (P-trend = 0.002). The median follow-up time for patients was 5.8 years. High tumor tissue levels of uPAR were associated with longer postoperative survival (HR = 0.4, 95% CI = 0.2-0.8, p = 0.01). In contrast, shorter survival was evident in patients with high tumor levels of uPA from 2 years on after operation (HR = 4.6, 95% CI = 1.2-17, p = 0.02). High tPA levels tended to be associated with shorter overall survival after 2 years (HR = 2.9, 95% 95% Cl = 0.9-9.8, p = 0.08). Although high tumor tissue content of uPAR was associated with a less aggressive phenotype characterized by well differentiated histology and longer survival, low content of uPAR in the poorly differentiated tumors and metastases presumably results from increased elimination of uPAR.   • Influence of Ladle-slag Additions on BOF-Process Parameters

    • The influence of ladle-slag additions on the BOF-process performance were investigated in plant trials. The aim of the study was to recycle ladle slag from secondary steelmaking to the LD-converter to save lime and improve the slag formation. More specifically, two plant trial campaigns covering in total 83 heats, whereof 47 with ladle-slag additions and 36 without ladle-slag additions, were performed.  Slag and steel sampling of the process were performed at tapping as well as during blowing at 15, 35, and 65% of the total blowing time. During the first campaign, ladle slag was added through the chute and lime reductions were made manually to correct for the ladle-slag addition.  In the second campaign, a development of the approach was made to suite a normal production practice. More specifically, the ladle slag was added through the weight-hopper system and implemented in the process-control system. In this way, the lime additions were reduced automatically by approximately 260 kg per heat. Moreover, the heat balance was compensated with a reduction in the iron-ore consumption. Additionally, the lance program was modified and the lance was lowered in the initial stages of the blow.  On the positive side, it was found that no demerits in the metallurgical performance of the process occur when ladle slag is recycled to the BOF-process. Furthermore, only slight affections on the slag composition were found, mainly with respect to the Al2O3 and FeO-content. In addition, the ladle slag was shown to melt during the initial stages of the blow. This contributed to an increased slag weight both during the blow and at tapping. However, a negative effect on the blowing time was experienced during the trials. Although, this effect was more pronounced during the first campaign and could be reduced with a controlled heat balance during the second campaign.

  • No serious late cardiac effects after adjuvant radiotherapy following mastectomy in premenopausal women with early breast cancer

    • PURPOSE: To assess cardiac mortality, coronary artery disease, myocardial dysfunction, and valvular heart disease in women younger than 65 years of age, at least 10 years after adjuvant radiotherapy following mastectomy in early breast cancer. METHODS AND MATERIALS: Ninety women (45-64 years old) with Stage II breast cancer without relapse, included in the South Sweden Breast Cancer Trial (premenopausal arm), with or without adjuvant postoperative radiotherapy +/- cyclophosphamide were examined with myocardial scintigraphy and echocardiography/Doppler, 10-17 years after radiotherapy. Thirty-four patients had been irradiated for left-sided tumors, 33 for right-sided tumors, and 23 patients had not been treated with radiotherapy. The radiotherapy (conventional roentgen, electron beams, and high-energy photon beams combined, in each patient) included the chest wall and the regional lymph nodes, with a specified target dose of 38-48 Gy, administered in daily fractions of 1.9-2.4 Gy, 5 days/week. RESULTS: No cardiac deaths were found among the original 275 patients randomized to adjuvant therapy. In the 90 patients examined, abnormal findings were recorded for ECG (14 patients), exercise test (5 patients), myocardial scintigraphy (6 patients), thickening of valve cusps (14 patients), and mild valvular regurgitation (20 patients). All patients had normal systolic function. Diastolic dysfunction was observed in 6 patients (abnormal relaxation in 4 patients and restrictive filling abnormality in 2 patients). Although no significant differences were found between the 3 study groups, there was a tendency to more abnormal findings after radiotherapy. CONCLUSION: Women younger than 50 years of age at the time of adjuvant radiotherapy following mastectomy in early breast cancer, had no serious cardiac sequelae 13 years (median) later, despite partly old-fashioned radiation techniques.

    • The equilibrium conditions for silicon transfer from ash to the liquid metal phase via SiO gas have been discussed by several authors. However, no published calculations have been found using the most modern thermodynamic models available. Since there are major differences in the results of calculations using different thermodynamic models and the models are continuously being improved, new equilibrium calculations on SiO and SiS gas formation have been performed using the recently developed models. Different ingoing compositions of coke ash, coal powder ash and blast air were used in the calculations. The compositions chosen represent blast furnace no. 3 at SSAB in Lulea, Sweden. Temperature was found to be the major factor influencing the equilibrium silicon level in the gas phase. At low temperatures (below 1 600 degrees C the total gas pressure was also seen to influence the silicon content in the gas phase. The main reason for this is that below 1 600 degrees C, the amount of liquid slag at equilibrium increases with the total gas pressure. Liquid slag contains large amounts of silica that then can not be found in the gas phase. Higher carbon activity is usually expected to result in higher SiO gas levels in the blast furnace. The equilibrium calculations show that increased carbon activity increases the amount of silicon in the gas phase at temperatures up to about 1 600 degrees C, but that at higher temperatures SiC is formed that decreases the equilibrium silicon level in the gas phase.

  • Characteristics of metal droplets in slag tapped from the blast furnace

    • Slag samples, hot-metal samples and hot-metal temperatures were obtained during tapping of two blast furnaces. Sampling was carried out at different time points during tapping of three separate heats. The size distribution and composition of metal droplets found in the slag were determined using scanning electron microscopy. Only metal droplets above 0.75 mu m could be counted and analysed. All droplets were below 8 mu m in diameter and the great majority of these droplets were found to be between 0.75 and 2 Pm. The size distribution did not differ significantly for different slag samples. Iron was the main droplet component. Electron probe microanalysis showed that the droplets contained small amounts of carbon. The percentage of the area in a studied cross-section that was covered with metal droplets varied between 0.01 and 0.07%. Calculations based on Stoke's law showed that the distance droplets travel in the slag is in the micron range. Slag samples taken in the beginning of slag tapping contained more droplets than those taken in the middle of slag tapping, an indication that most droplets can be found in the area near the furnace wall. Some droplets were determined to have magnesium enrichment at the external surface.
