    • We report on Be-10 and optically stimulated luminescence ages from moraines and glaciolacustrine sediments on eastern Jameson Land, East Greenland. Sampled landforms and sediment are associated with advances of outlet glaciers from the local Liverpool Land ice cap situated in the coastal Scoresby Sund region. Previous studies have tentatively correlated these advances with the Milne Land Stade moraines, which are prominent moraine sets deposited by mountain glaciers in the inner Scoresby Sund region. Recent constraints on the formation of the outer and inner of these moraines have suggested two advances of local glaciers, one prior to or during the Younger Dryas and another during the Preboreal. In this paper, we test the correlation of the Liverpool Land glacial advance with the Milne Land Stade. Our results show that outlet glaciers from the Liverpool Land ice cap reached ice-marginal positions marked by moraines in east-facing valleys on Jameson Land sometime during late glacial-early Holocene time (ca. 13-11 Kya). This confirms the correlation of these moraines with the Milne Land Stade moraines described elsewhere in the Scoresby Sund region.

  • The impact of ADHD and autism spectrum disorders on temperament, character, and personality development.

    • OBJECTIVE: The authors describe personality development and disorders in relation to symptoms of attention deficit hyperactivity disorder (ADHD) and autism spectrum disorders. METHOD: Consecutive adults referred for neuropsychiatric investigation (N=240) were assessed for current and lifetime ADHD and autism spectrum disorders and completed the Temperament and Character Inventory. In a subgroup of subjects (N=174), presence of axis II personality disorders was also assessed with the Structured Clinical Interview for DSM-IV Personality Disorders (SCID-II). RESULTS: Patients with ADHD reported high novelty seeking and high harm avoidance. Patients with autism spectrum disorders reported low novelty seeking, low reward dependence, and high harm avoidance. Character scores (self-directedness and cooperativeness) were extremely low among subjects with neuropsychiatric disorders, indicating a high overall prevalence of personality disorders, which was confirmed with the SCID-II. Cluster B personality disorders were more common in subjects with ADHD, while cluster A and C disorders were more common in those with autism spectrum disorders. The overlap between DSM-IV personality disorder categories was high, and they seem less clinically useful in this context. CONCLUSIONS: ADHD and autism spectrum disorders are associated with specific temperament configurations and an increased risk of personality disorders and deficits in character maturation.

    • Along the northeast Greenland continental margin, bedrock on interfjord plateaus is highly weathered, whereas rock surfaces in fjord troughs are characterized by glacial scour. Based on the intense bedrock weathering and lack of glacial deposits from the last glaciation, interfjord plateaus have long been thought to be ice-free throughout the last glacial maximum (LGM). In recent years there is growing evidence from shelf and fjord settings that the northeast Greenland continental margin was more extensively glaciated during the LGM than previously thought. However, little is still known from interfjord settings. We present cosmogenic (10)Be data from meltwater channels and weathered sandstone outcrops on Jameson Land, an interfjord highland north of Scoresby Sund. The mean exposure age of samples from channel beds (n = 3) constrains on the onset of deglaciation on interior Jameson Land to 18.5 +/- 1.3-21.4 +/- 1.9 ka (for erosion conditions of 0-10 mm/ka, respectively). This finding adds to growing evidence that the northeast Greenland continental margin was more heavily glaciated during the LGM than previously thought.

  • A DHODH inhibitor increases p53 synthesis and enhances tumor cell killing by p53 degradation blockage

    • The development of non-genotoxic therapies that activate wild-type p53 in tumors is of great interest since the discovery of p53 as a tumor suppressor. Here we report the identification of over 100 small-molecules activating p53 in cells. We elucidate the mechanism of action of a chiral tetrahydroindazole (HZ00), and through target deconvolution, we deduce that its active enantiomer (R)-HZ00, inhibits dihydroorotate dehydrogenase (DHODH). The chiral specificity of HZ05, a more potent analog, is revealed by the crystal structure of the (R)-HZ05/DHODH complex. Twelve other DHODH inhibitor chemotypes are detailed among the p53 activators, which identifies DHODH as a frequent target for structurally diverse compounds. We observe that HZ compounds accumulate cancer cells in S-phase, increase p53 synthesis, and synergize with an inhibitor of p53 degradation to reduce tumor growth in vivo. We, therefore, propose a strategy to promote cancer cell killing by p53 instead of its reversible cell cycle arresting effect.

    • Background Previously, nitric oxide has been shown to possess antimicrobial effects. In this study, we aim to test the effect of glyceryl trinitrate (GTN) on Staphylococcus aureus growth during simulated extracorporeal circulation (SECC) and also to examine the effect of S. aureus, alone and in combination with GTN, on activation markers of the innate immune system during SECC. Methods In an in vitro system of SECC, we measured GTN-induced changes in markers of leukocyte activation in whole blood caused by S. aureus infestation, as well as the effect of GTN on S. aureus growth. Results GTN had no effect on S. aureus growth after 240 minutes SECC. Staphylococcus aureus reduced the expression of granulocyte Fc gamma-receptor CD32 but stimulated the expression of monocyte CD32. Staphylococcus aureus stimulated expression of some leukocyte adhesion key proteins, activation marker CD66b, lipopolysaccharide-receptor CD14, and C3b-receptor CD35. Staphylococcus aureus and GTN addition induced significant increases in monocyte CD63 (lysosomal granule protein) levels. Conclusion GTN does not affect S. aureus growth during SECC and has no effect on SECC-induced leukocyte activation.

    • This Data in Brief paper contains data (including images) from Quaternary sedimentary successions investigated along the Bol'shaya Balakhnya River and the Luktakh-Upper Taimyra-Logata river system on southern Taimyr Peninsula, NW Siberia (Russia). Marine foraminifera and mollusc fauna composition, extracted from sediment samples, is presented. The chronology (time of deposition) of the sediment successions is reconstructed from three dating methods; (i) radiocarbon dating of organic detritus (from lacustrine/fluvial sediment) and molluscs (marine sediment) as finite ages (usually <42 000 years) or as non-finite ages (>42 000-48 000 years) on samples/sediments beyond the radiocarbon dating limit; (ii) Electron Spin Resonance (ESR) dating on marine molluscs (up to ages >400 000 years); (iii) Optically Stimulated Luminescence (OSL) dating, usually effective up to 100-150 0000 years. Terrestrial Cosmogenic Nuclide (TCN) exposure dating has been applied to boulders resting on top of moraine ridges (Ice Marginal Zones).
