  • Paleomagnetic and geochronological studies on Paleoproterozoic diabase dykes of Karelia, East Finland-Key for testing the Superia supercraton

    • Paleomagnetic results are presented for two Paleoproterozoic mafic dykes in the Taivalkoski area in northern Karelia Province of the Fennoscandian shield where, based on K-Ar data, the crust has seen minimal effects of the otherwise pervasive 1.8-1.9 Ga Svecofennian orogeny. Within this study a new U-Pb baddeleyite age of 2339 +/- 18 Ma has been determined for one of the E-W trending dykes (dyke AD13). The paleomagnetic results show that a strong Svecofennian overprinting is pervasive in the area. Upon thermal or AF demagnetization four remanence directions were obtained. Most typical are the secondary Svecofennian remanence direction A (intermediate down to the NNW) and remanence direction B (intermediate down to the NNE). Component D (D = 115.4 degrees, 1=50.5 degrees, alpha(95) =2.6 degrees) yielding a virtual geomagnetic pole (VGP) D (Plat= -19.5 degrees N, Plon= 263.3 degrees, A95 = 3.1 degrees) is obtained from baked rocks for dyke WD, and based on a positive baked contact test is interpreted to represent the primary magnetization dating from about 2.4 Ga. Dyke AD13 carries only secondary A and B components, its unbaked host migmatites carry reversed A (A(R)) component, and the baked host rock carries a component D' (D = 134.5 degrees, 1= -7.3 degrees, alpha(95) = 8.8 degrees), which yields a VGP pole D' (Plat= -20.4 degrees N, Plon = 257.3 degrees, A(95) = 7.6 degrees), possibly representing magnetization at 2.3 Ga. The new paleomagnetic data from the Karelia Province compared to similar-aged paleomagnetic data from the Superior Province does not support the recently proposed Superia configuration, based upon dyke swarm trajectories.   • ABO Genotyping finds more A2 to B kidney transplant opportunities than lectin-based subtyping

    • ABO compatibility is important for kidney transplantation, with longer waitlist times for blood group B kidney transplant candidates. However, kidneys from non-A1 (eg, A2) subtype donors, which express less A antigen, can be safely transplanted into group B recipients. ABO subtyping is routinely performed using anti-A1 lectin, but DNA-based genotyping is also possible. Here, we compare lectin and genotyping testing. Lectin and genotype subtyping was performed on 554 group A deceased donor samples at 2 transplant laboratories. The findings were supported by 2 additional data sets of 210 group A living kidney donors and 124 samples with unclear lectin testing sent to a reference laboratory. In deceased donors, genotyping found 65% more A2 donors than lectin testing, most with weak lectin reactivity, a finding supported in living donors and samples sent for reference testing. DNA sequencing and flow cytometry showed that the discordances were because of several factors, including transfusion, small variability in A antigen levels, and rare ABO∗A2.06 and ABO∗A2.16 sequences. Although lectin testing is the current standard for transplantation subtyping, genotyping is accurate and could increase A2 kidney transplant opportunities for group B candidates, a difference that should reduce group B wait times and improve transplant equity.

  • Interaction with Caveolin-1 Modulates G Protein Coupling of Mouse beta(3)-Adrenoceptor

    • Caveolins act as scaffold proteins in multiprotein complexes and have been implicated in signaling by G protein-coupled receptors. Studies using knock-out mice suggest that beta(3)-adrenoceptor (beta(3)-AR) signaling is dependent on caveolin-1; however, it is not known whether caveolin-1 is associated with the beta(3)-AR or solely with downstream signaling proteins. We have addressed this question by examining the impact of membrane rafts and caveolin-1 on the differential signaling of mouse beta(3a)- and beta(3b)-AR isoforms that diverge at the distal C terminus. Only the beta(3b)-AR promotes pertussis toxin (PTX)-sensitive cAMP accumulation. When cells expressing the beta(3a)-AR were treated with filipin III to disrupt membrane rafts or transfected with caveolin-1 siRNA, the cyclic AMP response to the beta(3)-AR agonist CL316243 became PTX-sensitive, suggesting G alpha(i/o) coupling. The beta(3a)-AR C terminus, S (P-384) under bar PLNR (P-389) under bar DG (Y-392) under bar EGARP (P-398) under bar PT, resembles a caveolin interaction motif. Mutant beta(3a)-ARs (F389A/Y392A/F398A or P384S/F389A) promoted PTX-sensitive cAMP responses, and in situ proximity assays demonstrated an association between caveolin-1 and the wild type beta(3a)-AR but not the mutant receptors. In membrane preparations, the beta(3b)-AR activated G alpha(o) and mediated PTX-sensitive cAMP responses, whereas the beta(3a)-AR did not activate G alpha(i/o) proteins. The endogenous beta(3a)-AR displayed G alpha(i/o) coupling in brown adipocytes from caveolin-1 knock-out mice or in wild type adipocytes treated with filipin III. Our studies indicate that interaction of the beta(3a)-AR with caveolin inhibits coupling to G alpha(i/o) proteins and suggest that signaling is modulated by a raft-enriched complex containing the beta(3a)-AR, caveolin-1, G alpha(s), and adenylyl cyclase.
