    • Antennal lobe neurones were investigated in the pyralid moth Ostrinia nubilalis using intracellular recording and staining techniques. Response characteristics of antennal lobe neurones from males in the so-called E and Z strains, in F1 hybrids and in parental backcrosses were studied. The antennal lobe of a male O. nubilalis comprises approximately 30 ordinary glomeruli and three enlarged glomeruli making up the macroglomerular complex (MGC). Receptor neurones enter the antennal lobe via the antennal nerve and arborize in single glomeruli. Intracellularly stained, pheromone-responding projection neurones in both parental strains arborized in different glomeruli within the MGC, irrespective of their response characteristics. Neurones were grouped according to their specificity to single pheromone components and to pheromone blends. Component-specific, blend-specific and generalist neurones were found. Specificity only occurred at low stimulus concentrations and disappeared as concentrations increased. Although all neuronal types were present in both pheromone strains and crossings, differences in abundance and sensitivity were found. In the parental strains, neurones responding to the major pheromone component and to the respective strain-specific blend were more abundant than neurones responding to the minor component and the blend produced by the other strain. Neurones investigated in ZxE hybrids responded similarly to those of E-strain males, whereas neurones in EZxZ paternal backcrosses responded similarly to those of Z males. In the hybrids and paternal backcrosses, hybrid-blend-specific neurones were present that were not found in parental-strain males.

  • Developmental changes in the structure and function of the central olfactory system in gregarious and solitary desert locusts

    • Desert locusts are guided by olfactory cues in different behavioural contexts. In order to understand the basis for the variable olfactory guided behaviour displayed by different developmental stages and by solitary and gregarious locusts, we investigated their central olfactory system with neuroanatomical and neurophysiological methods. The primary olfactory centre of the brain, the antennal lobe (AL), increases in size during development due to an increased number and size of glomeruli. These glomeruli are innervated by a constant number of projection neurons that display increased dendritic arborizations during the development of the locust. The anatomical parameters do not differ between gregarious and solitary locusts, In parallel with the observed neuroanatomical changes, neurophysiological changes in response spectra and response specificity of AL neurons were found. During development, the percentage of neurons responding specifically to aggregation pheromone components decreases, whereas an increase in both pheromone-generalists and plant-pheromone generalist neurons is observed. The percentage of neurons responding to green leaf volatiles, however, remains constant. A decrease in the number of nymph blend-specific neurons was also observed. Our data show that anatomical and physiological properties of the AL and its neurons to a large extent reflect the changes in olfactory guided behaviour during development and between phases. The majority of our results are also in accordance with findings that the number of olfactory receptor neurons increases during development, resulting in increasing convergence on AL neurons.

  • Fine mapping of genetic susceptibility loci for melanoma reveals a mixture of single variant and multiple variant regions

    • At least 17 genomic regions are established as harboring melanoma susceptibility variants, in most instances with genome-wide levels of significance and replication in independent samples. Based on genome-wide single nucleotide polymorphism (SNP) data augmented by imputation to the 1,000 Genomes reference panel, we have fine mapped these regions in over 5,000 individuals with melanoma (mainly from the GenoMEL consortium) and over 7,000 ethnically matched controls. A penalized regression approach was used to discover those SNP markers that most parsimoniously explain the observed association in each genomic region. For the majority of the regions, the signal is best explained by a single SNP, which sometimes, as in the tyrosinase region, is a known functional variant. However in five regions the explanation is more complex. At the CDKN2A locus, for example, there is strong evidence that not only multiple SNPs but also multiple genes are involved. Our results illustrate the variability in the biology underlying genome-wide susceptibility loci and make steps toward accounting for some of the missing heritability. What's new? In genome-wide association studies, researchers identify genetic variants that frequently associate with a particular disease, though the variants identified may not contribute to the molecular cause of the disease. This study took a closer look at 17 regions associated with melanoma, fine mapping the regions both in people with melanoma and in healthy controls. Though single SNPs account for the association in some regions, they found that in a few regions, several SNPs - and possibly multiple genes - contributed to the association signal. These findings illustrate the importance of not overlooking the interaction between multiple genetic markers when conducting such studies.

  • Genome-wide association study identifies three new melanoma susceptibility loci

    • We report a genome-wide association study for melanoma that was conducted by the GenoMEL Consortium. Our discovery phase included 2,981 individuals with melanoma and 1,982 study-specific control individuals of European ancestry, as well as an additional 6,426 control subjects from French or British populations, all of whom were genotyped for 317,000 or 610,000 single-nucleotide polymorphisms (SNPs). Our analysis replicated previously known melanoma susceptibility loci. Seven new regions with at least one SNP with P < 10(-5) and further local imputed or genotyped support were selected for replication using two other genome-wide studies (from Australia and Texas, USA). Additional replication came from case-control series from the UK and The Netherlands. Variants at three of the seven loci replicated at P < 10(-3): an SNP in ATM (rs1801516, overall P = 3.4 x 10(-9)), an SNP in MX2 (rs45430, P = 2.9 x 10-9) and an SNP adjacent to CASP8 (rs13016963, P = 8.6 x 10(-10)). A fourth locus near CCND1 remains of potential interest, showing suggestive but inconclusive evidence of replication (rs1485993, overall P = 4.6 x 10(-7) under a fixed-effects model and P = 1.2 x 10(-3) under a random-effects model). These newly associated variants showed no association with nevus or pigmentation phenotypes in a large British case-control series.

    • Odonata (dragonflies and damselflies) present an unparalleled insect model to integrate evolutionary genomics with ecology for the study of insect evolution. Key features of Odonata include their ancient phylogenetic position, extensive phenotypic and ecological diversity, several unique evolutionary innovations, ease of study in the wild and usefulness as bioindicators for freshwater ecosystems worldwide. In this review, we synthesize studies on the evolution, ecology and physiology of odonates, highlighting those areas where the integration of ecology with genomics would yield significant insights into the evolutionary processes that would not be gained easily by working on other animal groups. We argue that the unique features of this group combined with their complex life cycle, flight behaviour, diversity in ecological niches and their sensitivity to anthropogenic change make odonates a promising and fruitful taxon for genomics focused research. Future areas of research that deserve increased attention are also briefly outlined.

    • The paper presents the control server, a real-time scheduling mechanism tailored to control and signal processing applications. A control server creates the abstraction of a control task with a specified period and a fixed input-output latency shorter than the period. Individual tasks can be combined into more complex components without loss of their individual guaranteed fixed-latency properties. I/O occurs at fixed predefined points in time, at which inputs are read or controller outputs become visible. The control server model is especially suited for codesign of real-time control systems. The single parameter linking the scheduling design and the controller design is the task utilization factor. The proposed server is an extension of the constant bandwidth server, which is based on the earliest-deadline-first scheduling algorithm. The server has been implemented in a real-time kernel and has also been validated in control experiments on a ball and beam process.

  • Regional Patterns of Late Medieval and Early Modern European Building Activity Revealed by Felling Dates

    • Although variations in building activity are a useful indicator of societal well-being and demographic development, historical datasets for larger regions and longer periods are still rare. Here, we present 54,045 annually precise dendrochronological felling dates from historical construction timber from across most of Europe between 1250 and 1699 CE to infer variations in building activity. We use geostatistical techniques to compare spatiotemporal dynamics in past European building activity against independent demographic, economic, social and climatic data. We show that the felling dates capture major geographical patterns of demographic trends, especially in regions with dense data coverage. A particularly strong negative association is found between grain prices and the number of felling dates. In addition, a significant positive association is found between the number of felling dates and mining activity. These strong associations, with well-known macro-economic indicators from pre-industrial Europe, corroborate the use of felling dates as an independent source for exploring large-scale fluctuations of societal well-being and demographic development. Three prominent examples are the building boom in the Hanseatic League region of northeastern Germany during the 13th century, the onset of the Late Medieval Crisis in much of Europec. 1300, and the cessation of building activity in large parts of central Europe during armed conflicts such as the Thirty Years’ War (1618–1648 CE). Despite new insights gained from our European-wide felling date inventory, further studies are needed to investigate changes in construction activity of high versus low status buildings, and of urban versus rural buildings, and to compare those results with a variety of historical documentary sources and natural proxy archives.

    • An association between low serum vitamin D levels and poorer melanoma survival has been reported. We have studied inheritance of a polymorphism of the GC gene, rs2282679, coding for the vitamin D-binding protein, which is associated with lower serum levels of vitamin D, in a meta-analysis of 3137 melanoma patients. The aim was to investigate evidence for a causal relationship between vitamin D and outcome (Mendelian randomization). The variant was not associated with reduced overall survival (OS) in the UK cohort, per-allele hazard ratio (HR) for death 1.23 (95% confidence interval (CI) 0.93, 1.64). In the smaller cohorts, HR in OS analysis was 1.07 (95% CI 0.88, 1.3) and for all cohorts combined, HR for OS was 1.09 (95% CI 0.93, 1.29). There was evidence of increased melanoma-specific deaths in the seven cohorts for which these data were available. The lack of unequivocal findings despite the large sample size illustrates the difficulties of implementing Mendelian randomization.

  • Drug diffusion in biomimetic hydrogels : importance for drug transport and delivery in non-vascular tumor tissue

    • Hydrogels of varying complexity are routinely used as scaffolds and 3D structures for in vitro tumor models to increase physiological relevance within pre-clinical cancer research. Relatively simple hydrogels such as agarose are well characterised, meanwhile biomimetic gels containing collagen and fibrin(ogen) have been studied to a much lesser extent. In this study, hydrogels mimicking the biophysical characteristics of liver cancer progression were investigated in terms of their UV-properties and influence on diffusion coefficients of different substances. UV-imaging technology was used to both visualize and quantify the diffusion process in a simple and rapid way. In general, agarose gel diffusion agreed well with predictions using the Stokes-Einstein equation meanwhile the biomimetic gels reduced diffusion coefficients by up to 70%. For doxorubicin, spatio-temporal tissue concentration modelling was used to translate in vitro diffusion to the more clinical context of tumor penetration in a solid liver tumor supplied by arterial blood.
