  • A three-dimensional movie of structural changes in bacteriorhodopsin
  • 2016
  • Ingår i: Science. - : American Association for the Advancement of Science (AAAS). - 0036-8075 .- 1095-9203. ; 354:6319, s. 1552-1557
  • Tidskriftsartikel (refereegranskat)abstract
    • Bacteriorhodopsin (bR) is a light-driven proton pump and a model membrane transport protein. We used time-resolved serial femtosecond crystallography at an x-ray free electron laser to visualize conformational changes in bR from nanoseconds to milliseconds following photoactivation. An initially twisted retinal chromophore displaces a conserved tryptophan residue of transmembrane helix F on the cytoplasmic side of the protein while dislodging a key water molecule on the extracellular side. The resulting cascade of structural changes throughout the protein shows how motions are choreographed as bR transports protons uphill against a transmembrane concentration gradient.

  • Nanosecond pump-probe device for time-resolved serial femtosecond crystallography developed at SACLA
  • 2017
  • Ingår i: Journal of Synchrotron Radiation. - : International Union of Crystallography (IUCr). - 1600-5775. ; 24, s. 1086-1091
  • Tidskriftsartikel (refereegranskat)abstract
    • X-ray free-electron lasers (XFELs) have opened new opportunities for timeresolved X-ray crystallography. Here a nanosecond optical-pump XFEL-probe device developed for time-resolved serial femtosecond crystallography (TRSFX) studies of photo-induced reactions in proteins at the SPring-8 Angstrom Compact free-electron LAser (SACLA) is reported. The optical-fiber-based system is a good choice for a quick setup in a limited beam time and allows pump illumination from two directions to achieve high excitation efficiency of protein microcrystals. Two types of injectors are used: one for extruding highly viscous samples such as lipidic cubic phase (LCP) and the other for pulsed liquid droplets. Under standard sample flow conditions from the viscous-sample injector, delay times from nanoseconds to tens of milliseconds are accessible, typical time scales required to study large protein conformational changes. A first demonstration of a TR-SFX experiment on bacteriorhodopsin in bicelle using a setup with a droplet-type injector is also presented.

  • Deep Inner-Shell Multiphoton Ionization by Intense X-Ray Free-Electron Laser Pulses
  • 2013
  • Ingår i: Physical Review Letters. - 1079-7114 .- 0031-9007. ; 110:17
  • Tidskriftsartikel (refereegranskat)abstract
    • We have investigated multiphoton multiple ionization dynamics of xenon atoms using a new x-ray free-electron laser facility, SPring-8 Angstrom Compact free electron LAser (SACLA) in Japan, and identified that Xen+ with n up to 26 is produced at a photon energy of 5.5 keV. The observed high charge states (n >= 24) are produced via five-photon absorption, evidencing the occurrence of multiphoton absorption involving deep inner shells. A newly developed theoretical model, which shows good agreement with the experiment, elucidates the complex pathways of sequential electronic decay cascades accessible in heavy atoms. The present study of heavy-atom ionization dynamics in high-intensity hard-x-ray pulses makes a step forward towards molecular structure determination with x-ray free-electron lasers. • Sequential multiphoton multiple ionization of atomic argon and xenon irradiated by X-ray free-electron laser pulses from SACLA
  • 2013
  • Ingår i: Journal of Physics B: Atomic, Molecular and Optical Physics. - : IOP Publishing. - 0953-4075 .- 1361-6455. ; 46:16
  • Tidskriftsartikel (refereegranskat)abstract
    • We have investigated multiphoton multiple ionization of argon and xenon atoms at 5 keV using a new x-ray free electron laser (XFEL) facility, the SPring-8 Angstrom Compact free electron LAser (SACLA) in Japan. The experimental results are compared with the new theoretical results presented here. The absolute fluence of the XFEL pulse has been determined with the help of the calculations utilizing two-photon processes in the argon atom. The high charge states up to +22 observed for Xe in comparison with the calculations point to the occurrence of sequential L-shell multiphoton absorption and of resonance-enabled x-ray multiple ionization.

  • Valence excitations observed in resonant soft X-ray emission spectra of K2Ni(CN)4.H2O at the Ni 2p edge
  • 2001
  • Ingår i: Journal of Electron Spectroscopy and Related Phenomena. - 0368-2048 .- 1873-2526. ; 114-116, s. 909-913
  • Tidskriftsartikel (refereegranskat)abstract
    • Resonant soft X-ray emission spectra have been measured at the Ni 2p edge of a planar nickel complex K2Ni(CN)4·H2O and interpreted by performing ab initio configuration interaction calculations. This cyano complex shows characteristic metal-to-ligand charge transfer (MLCT) bands in the Ni 2p photoabsorption spectra. Resonant soft X-ray emission via the MLCT intermediate states can be interpreted as valence MLCT excitations within a one-electron picture. On the other hand, resonant soft X-ray emission via the intra-atomic Ni 2p–3d intermediate state corresponding to the white line excitation shows significant electron correlation effects.
