    • Ekonomibyggnader har alltid behövts för de huvudbyggnader som finns inom våra bevarade kulturmiljöer. Några nyttobyggnader uppskattas och används fortfarande, andra betraktas som problematiska överloppsbyggnader, många rivs. Alltför få har dokumenterats eller fått sin historia klarlagd vilket undanhållit viktig kunskap om samhällets framväxt. Vi vill synliggöra och betona vikten av att bevara och utveckla hela bebyggelsemiljöer, ofta med ett antal hus utöver huvudbyggnaden och tillhörande yttre miljö i staden eller på landet. Denna rapport visar kursdeltagarnas projektarbeten om nyttobyggnader. De har dokumenterat med traditionella och nya arbetsmetoder, inventerat och intervjuat, läst och besökt arkiv, värderat, analyserat och därefter föreslagit hur man ska ta hand om och utveckla nyttobyggnaderna i sina kulturmiljöer.

  • High tumour cannabinoid CB(1) receptor immunoreactivity negatively impacts disease-specific survival in stage II microsatellite stable colorectal cancer

    • Background: There is good evidence in the literature that the cannabinoid system is disturbed in colorectal cancer. In the present study, we have investigated whether CB(1) receptor immunoreactive intensity (CB(1)IR intensity) is associated with disease severity and outcome. Methodology/Principal Findings: CB(1)IR was assessed in formalin-fixed, paraffin-embedded specimens collected with a consecutive intent during primary tumour surgical resection from a series of cases diagnosed with colorectal cancer. Tumour centre (n = 483) and invasive front (n = 486) CB(1)IR was scored from 0 (absent) to 3 (intense staining) and the data was analysed as a median split i.e. CB(1)IR <2 and >= 2. In microsatellite stable, but not microsatellite instable tumours (as adjudged on the basis of immunohistochemical determination of four mismatch repair proteins), there was a significant positive association of the tumour grade with the CB1IR intensity. The difference between the microsatellite stable and instable tumours for this association of CB(1)IR was related to the CpG island methylation status of the cases. Cox proportional hazards regression analyses indicated a significant contribution of CB(1)IR to disease-specific survival in the microsatellite stable tumours when adjusting for tumour stage. For the cases with stage II microsatellite stable tumours, there was a significant effect of both tumour centre and front CB(1)IR upon disease specific survival. The 5 year probabilities of event-free survival were: 8565 and 66+/-8%; tumour interior, 86+/-4% and 63+/-8% for the CB(1)IR<2 and CB(1)IR >= 2 groups, respectively. Conclusions/Significance: The level of CB(1) receptor expression in colorectal cancer is associated with the tumour grade in a manner dependent upon the degree of CpG hypermethylation. A high CB(1)IR is indicative of a poorer prognosis in stage II microsatellite stable tumour patients.

  • Introduktion av värmekameror i undervisningen vid Lillerudsgymnasiet

    • Vi har bedrivit en forskningscirkel med syfte att utvärdera värmekameran som visualiseringsteknik i olika ämnen på naturbruksprogrammet genom att planera, genomföra och reflektera kring lektionsupplägg, där elever har fått möjlighet att använda värmekameror med olika grad av styrning. Resultatet visar att värmekameran har många skilda tillämpningsområden inom lantbruk och djurhållning och kan användas i undervisningen som ett verktyg för elevers praktiska undersökningar av t.ex. djurs fysiologi, eller som ett mätinstrument då tillfälle ges, t.ex. för att studera juverbölder hos suggor eller temperaturökning när en skruv tar fel gäng. En utmaning i undervisningen är att finna en balans mellan öppenhet och styrning, där eleverna upplever att de har möjlighet att undersöka fenomen de själva är intresserade av, men utifrån ett etablerat, systematiskt arbetssätt. Sammanfattningsvis har vi upplevt användning av värmekameror som ett effektivt och intresseväckande sätt att konkretisera och individanpassa undervisningen på naturbruksprogrammet, vilket vi kan rekommendera andra lärare att prova.

  • Evolving Perspectives of Stewardship in the Seafood Industry

    • Humanity has never benefited more from the ocean as a source of food, livelihoods, and well-being, yet on a global scale this has been accompanied by trajectories of degradation and persistent inequity. Awareness of this has spurred policymakers to develop an expanding network of ocean governance instruments, catalyzed civil society pressure on the public and private sector, and motivated engagement by the general public as consumers and constituents. Among local communities, diverse examples of stewardship have rested on the foundation of care, knowledge and agency. But does an analog for stewardship exist in the context of globally active multinational corporations? Here, we consider the seafood industry and its efforts to navigate this new reality through private governance. We examine paradigmatic events in the history of the sustainable seafood movement, from seafood boycotts in the 1970s through to the emergence of certification measures, benchmarks, and diverse voluntary environmental programs. We note four dimensions of stewardship in which efforts by actors within the seafood industry have aligned with theoretical concepts of stewardship, which we describe as (1) moving beyond compliance, (2) taking a systems perspective, (3) living with uncertainty, and (4) understanding humans as embedded elements of the biosphere. In conclusion, we identify emerging stewardship challenges for the seafood industry and suggest the urgent need to embrace a broader notion of ocean stewardship that extends beyond seafood.

  • Colorectal Cancer Cells Activate Adjacent Fibroblasts Resulting in FGF1/FGFR3 Signaling and Increased Invasion.

    • Cancer-associated fibroblasts expressing fibroblast activation protein (FAP) have been implicated in the invasive behavior of colorectal cancer. In this study, we use FAP expression as a marker of fibroblast activation and analyze the effect of activated fibroblasts on colorectal cancer migration and invasion in experimental cell studies. We also investigated the expression pattern of FAP in cancer-associated fibroblasts during transformation from benign to malignant colorectal tumors. In immunohistochemical analyses, FAP was expressed in fibroblasts in all colorectal cancer samples examined, whereas all normal colon, hyperplastic polyps, or adenoma samples were negative. In in vitro studies, conditioned medium from colon cancer cells, but not adenoma cells, activated fibroblasts by inducing FAP expression. These activated fibroblasts increased the migration and invasion of colon cancer cells in Boyden chamber experiments and in a three-dimensional cell culture model. We identify fibroblast growth factor 1/fibroblast growth factor receptor 3 (FGF1/FGFR-3) signaling as mediators leading to the increased migration and invasion. Activated fibroblasts increase their expression of FGF1, and by adding a fibroblast growth factor receptor inhibitor, as well as an FGF1-neutralizing antibody, we reduced the migration of colon cancer cells. Our findings provide evidence of a possible molecular mechanism involved in the cross talk between cancer cells and fibroblasts leading to cancer cell invasion.

  • Topsoil removal enhances plant target species occurrence in sandy calcareous grassland

    • Species-rich sandy calcareous grasslands are threatened by land use change, eutrophication and acidification. Disturbance is vital for the persistence of characteristic sandy calcareous grassland species because it provides habitats with low competition, and can lead to increased pH and decreased nutrient availability, which favour many threatened species. Today many sandy calcareous grasslands lack a proper disturbance regime. The outcome of topsoil removal, applied as restoration measure, was investigated in a multi-site study. Successful restoration was associated with colonisation of plants representing conservation target species, as well as increased pH and reduced amounts of nutrients and organic matter. The impact of topsoil removal on plant species composition and soil properties was examined, with adjacent degraded plots serving as controls. Non-degraded target vegetation was included as reference areas. Restored plots had a significantly higher mean proportion of target species compared to control plots. However, control plots had higher species richness. Topsoil removal increased the pH value and decreased contents of ammonium and organic matter in the newly established topsoil layer, while no effect was seen on nitrate, phosphate and lime. Restored plots had a high cover of bare sand, but in contrast to target plots, a rather low proportion of forbs in relation to graminoids. Control plots had a higher vegetation cover, mainly represented by graminoids, forbs and the dwarf shrub species Calluna vulgaris (L.) Hull. Our study suggests that topsoil removal is an effective restoration measure due to its positive influence on soil characteristics and target species. However, the degraded sandy grasslands that were chosen as controls in this study should also be recognised as important habitats with conservation values due to the high species richness, suggesting that a mosaic pattern of the different successional stages is desirable.

  • Molecular characterization of the cancer susceptibility protein wrap53β in Cajal body formation and DNA repair

    • WRAP53β is a multifaceted protein involved in several biological processes including Cajal body maintenance, cancer cell survival and DNA damage repair. By directing factors to Cajal bodies and DNA double-strand breaks (DSBs), WRAP53β facilitates site-specific interactions necessary for proper biological responses. The Cajal body is a subnuclear organelle implicated in cellular processes such as splicing machinery maturation and telomere maintenance. In Paper I, we reveal that WRAP53β is an essential structural component of Cajal bodies. Furthermore, WRAP53β is required for the intracellular targeting of factors to this site. WRAP53β associates with the survival of motor neuron (SMN) complex in the cytoplasm, mediates its nuclear import and subsequent Cajal body accumulation. In addition, we find that the interaction between WRAP53β and SMN is disrupted in the severe neurodegenerative disorder spinal muscular atrophy, suggesting clinical relevance of WRAP53β-mediated SMN transport. In Paper II, we study the relationship between WRAP53β expression and cancer cell survival. We demonstrate that WRAP53β is overexpressed in a panel of different cancer cell lines in comparison to primary cells. WRAP53β depletion results in massive induction of cancer cell death, whereas normal human fibroblasts are largely insensitive to WRAP53β knockdown. The cell death associated with WRAP53β silencing occurs via the intrinsic mitochondrial pathway as demonstrated by Bax/Bak activation, loss of mitochondrial membrane potential and release of cytochrome c. Finally, we show that high WRAP53β expression levels correlate with poor prognosis and radioresistance of head and neck cancer patients. In Paper III, we establish WRAP53β as a novel player in the DNA damage response. We show that WRAP53β rapidly and transiently localizes to DNA DSBs in an ATM- and PARP-dependent manner. WRAP53β binds the E3 ligase RNF8 and facilitates its interaction with MDC1, which is essential for the downstream recruitment of repair proteins 53BP1, BRCA1 and RAD51 to damaged sites. Knockdown of WRAP53β results in deficient DNA DSB repair, whereas WRAP53β overexpression enhances repair efficiency and provides resistance to DNA damaging agents. Furthermore, reduced expression of WRAP53β is related to decreased ovarian cancer patient survival. In summary, our data identify WRAP53β as a novel structural and regulatory component of Cajal bodies as well as an important factor in carcinogenesis and DNA repair.
