  • Volumetric dried blood spots for determination of phosphatidylethanol: Validation of a liquid chromatography tandem masspectrometry method and clinical application
  • 2024
  • Ingår i: DRUG TESTING AND ANALYSIS. - 1942-7603 .- 1942-7611.
  • Tidskriftsartikel (refereegranskat)abstract
    • Phosphatidylethanol (PEth) measurement in whole blood samples is established as a specific alcohol biomarker with clinical and forensic applications. Establishment of dried blood spots (DBSs) as a specimen for PEth determination offers several advantages and was the focus of this work. A liquid chromatography tandem mass spectrometry method using a 96-well format for sample preparation was developed and validated. PEth was extracted from DBSs by using isopropanol containing PEth-d5 as internal standard. The blood sampling used a commercial volumetric DBS device having a phosholipase D inhibitor incorporated to stop continuous PEth formation. The method quantified PEth in the range of 0.05-10 mu mol/L, with a bias and imprecision of less than 15%. In a clinical study (n = 25) using fingerprick blood, the volumetric device offered more precise quantifications (CV 4.6%) compared with the Whatman 903 Protein Saver card device (CV 16.6%). In another clinical study (n = 48), the use of dried venous and capillary blood, and liquid venous blood was compared under real-life conditions with samples sent by postal service. The capillary and venous DBS samples gave identical results while the liquid blood gave slightly higher values. Calculation of elimination half-life (PEth 16:0/18:1) in 31 cases based on two consecutive samples with 2-9 days in between gave results (mean 6.2 days) that agree with literature but several cases with values over 10 days. In conclusion, this study demonstrates that volumetric DBS is a valid specimen for determination of PEth blood concentrations, offering several advantages. Phosphatidylethanol, a specific alcohol biomarker, measured in dried blood spots was the focus of this work. A liquid chromatography tandem mass spectrometry method was developed, validated, and applied. The blood sampling used a volumetric DBS device. The volumetric device gave accurate and precise quantifications and the dried capillary blood gave the same results as that of venous blood. image

  • Isolation and sequencing of the replication region of plasmid pBFp1 isolated from a marine biofilm
  • 2004
  • Ingår i: Plasmid. - : Elsevier BV. - 0147-619X. ; 51:3, s. 179-184
  • Tidskriftsartikel (refereegranskat)abstract
    • A 24 kb plasmid, pBFp1, encoding mercury resistance was previously isolated from a marine biofilm. Isolation and sequencing of a 4280 bp DNA fragment containing the plasmid replicon (rep-pBFp1) revealed a putative open reading frame encoding a RepA protein and an oriV-like region containing an A + T rich sub-region, iterons, and DnaA boxes. Sequence comparisons showed significant similarities to the incW plasmid pSa both for the RepA amino acid sequence and in the iteron DNA sequence. Plasmid pBFp1 was also shown to be incompatible with pSa in standard incompatibility testing. A probe from the repA gene of pBFp1 was further made and tested on a collection of plasmids exogenously isolated from marine habitats in a previous study

  • NIMBYs for the rich and YIMBYs for the poor : analyzing the property price effects of infill development
  • 2020
  • Ingår i: Journal of European Real Estate Research. - : EMERALD GROUP PUBLISHING LTD. - 1753-9269 .- 1753-9277. ; 13:1, s. 55-81
  • Tidskriftsartikel (refereegranskat)abstract
    • Purpose This paper aims to analyze how nearby property prices are affected by new construction projects in Stockholm. If there is an impact on property prices, the authors endeavor to investigate whether the effects vary among different areas within the municipality, for different groups of inhabitants and for different types of housing (i.e. public versus private housing). Design/methodology/approach The authors use a difference-in-difference specification in a hedonic model, and the sample consists of more than 90,000 observations over the period 2005-2013. Findings The results are robust and indicate that house prices in nearby areas increase following the completion of infill development. The results also indicate that infill development has a positive spillover effect on nearby dwelling prices only in areas with lower incomes, more public housing units and more inhabitants born abroad. Originality/value It provides an analysis on how nearby property prices are affected by new construction projects by creating a restricted control area, so as to make the treatment group and the control group more homogeneous. Thus, it mitigates any potential problems with spatial dependency, which can cause biased standard errors.

  • NIMBYs for the rich and YIMBYs for the poor: Analyzing the property price effects of infill development
  • 2016
  • Rapport (övrigt vetenskapligt/konstnärligt)abstract
    • A combination of strong urbanization and shortage of land in many European city areas prompts an impetus of infill development, with current residents often raising concerns that infill development leads to lower nearby property prices. The aim of this paper is to analyze how nearby property prices are affected by new construction projects in Stockholm, Sweden. We use a difference-in-difference specification in a hedonic model, and our sample consists of more than 40,000 observations over the period 2005–2013. Our results are robust and indicate that house prices in nearby areas increase following the completion of infill development. Our results also indicate that infill development only has a positive spillover effect on nearby house prices only in areas with lower incomes, more public housing units and more inhabitants born abroad.

  • Translation and linguistic validation of self-administered Swedish version of WHODAS 2.0
  • 2015
  • Konferensbidrag (refereegranskat)abstract
    • Background: Use of Patient-Reported Outcomes (PROs) is increasingly requested in clinical practice and research. The World Health Organization Disability Assessment Schedule 2.0 (WHODAS 2.0) is a generic instrument for self-report capturing functioning in the life-domains of understanding and communicating, getting around, self-care, getting along with people, life activities and participation in society. It is based on the same conceptual foundation as the International Classification of Functioning Disability and Health (ICF). Several language versions of WHODAS 2.0 are available and a Swedish version is in progress.Aim: To translate the self-administered 36-item version of WHODAS 2.0 into Swedish and examine the understanding and interpretation of the instruments´ content and response processes. Methods: Translation process was made following WHO guidelines and the WHODAS 2.0 translation package (version 1.0) including: forward-translation, expert panel discussion, back-translation, pre-test of preliminary version, final version, and pilot-testing of the final version.Pre-test was conducted through structured interviews in purposeful samples of Swedish-speaking adults including two patients with bi-polar disorder and a sample from the general population. Pilot-testing of the final version will be performed by structured interviews in a purposeful sample of outpatients. Results: Forward-translation, expert panel discussion, and back-translation resulted in a preliminary Swedish version of WHODAS 2.0. Pre-test revealed some issues related to items in the Participation in society domain, and in the overall lay-out. This resulted in minor changes in selection of words in the final version and suggestions for future revision of the instrument.Pilot-testing of the final version in outpatients are under progress and the result will be ready to present before Conference date in November 2015.Conclusion: The preliminary Swedish self-administered version of WHODAS 2.0 is easy to understand and respond to. Further studies are needed to provide evidence for validity of the final Swedish WHODAS 2.0.
