  • Emerging Adult Parents’ Coordination of Work and Family Commitments

    • It has been proposed that the evaluation and coordination of commitments in different areas of life, such as work and family, becomes an important developmental task in late emerging adulthood (e.g., Pals, 1999). In the present study we explore how this coordination takes place by investigating how 33 (23 women) Swedish parents in late emerging adulthood reason around priorities between work and family. The participants took part in a semi-structured interview concerning their attitudes towards work/family priorities. Thematic analysis was used to analyze the material (Braun & Clarke, 2006). When describing their work/family priorities most parents talked about family as being more important than work, but many also talked about a strive for work/family balance. A small group said that work and family were equally important, but none of the participants said that they prioritized work over family. However, the results showed that wanting to prioritize family had different consequences for how different participants organized their work life. Most participants talked about how they were simply working their “regular hours”. Some, but not all, of these participants thought that working regular hours meant that their children had to spend too many hours in daycare, and some of them expressed a wish to work part time. A smaller group of participants (only women) work part time. These participants often talked about wanting to downshift and spend time with their children. In contrast, some participants talked about working long hours and putting in a lot of overtime. These participants often explained how this choice was a way for them to prioritize their family. For example, they talked about how working long hours would benefit their family financially or give them other advantages. Many participants said that their view of work/family priorities had changed when they became parents. Some of these participants said that before becoming parents they had thought that it would not be so difficult to combine work and family as it later turned out to be. Some also said that before becoming parents they had not thought much about work/family issues at all, or that their career had been more important to them before they became parents. A smaller group of participants also talked about how they expected, or had already experienced, that their view on work/family priorities would change during parenthood. For example, when their child got older and more independent they would be able to engage more in their work life. In conclusion, this study indicates that to prioritize family before work is normative among emerging adult Swedish parent, but that doing so can mean very different things to different people. Moreover, the results show that when emerging adults become parents work/family priorities is often a more pressing issue than it was before. This suggests that the transition to parenthood requires young people to negotiate their commitments in different life areas in relation to the new social realities of adult life.

  • TRAF6 ubiquitinates TGFβ type I receptor to promote its cleavage and nuclear translocation in cancer

    • Transforming growth factor β (TGFβ) is a pluripotent cytokine promoting epithelial cell plasticity during morphogenesis and tumour progression. TGFβ binding to type II and type I serine/threonine kinase receptors (TβRII and TβRI) causes activation of different intracellular signaling pathways. TβRI is associated with the ubiquitin ligase tumor necrosis factor receptor (TNFR)-associated factor 6 (TRAF6). Here we show that TGFβ, via TRAF6, causes Lys63-linked polyubiquitination of TβRI, promoting cleavage of TβRI by TNF-alpha converting enzyme (TACE), in a PKCζ-dependent manner. The liberated intracellular domain (ICD) of TβRI associates with the transcriptional regulator p300 to activate genes involved in tumour cell invasiveness, such as Snail and MMP2. Moreover, TGFβ-induced invasion of cancer cells is TACE- and PKCζ- dependent and the TβRI ICD is localized in the nuclei of different kinds of tumour cells in tissue sections. Thus, our data reveal a specific role for TβRI in TGFβ mediated tumour invasion.

    • Moving bed biofilm reactors (MBBRs) are increasingly used for nitrogen removal with nitritation-anaerobic ammonium oxidation (anammox) processes in wastewater treatment. Carriers provide protected surfaces where ammonia oxidizing bacteria (AOB) and anammox bacteria form complex biofilms. However, the knowledge about the organization of microbial communities in MBBR biofilms is sparse. We used new cryosectioning and imaging methods for fluorescence in situ hybridization (FISH) to study the structure of biofilms retrieved from carriers in a nitritation-anammox MBBR. The dimensions of the carrier compartments and the biofilm cryosections after FISH showed good correlation, indicating little disturbance of biofilm samples by the treatment. FISH showed that Nitrosomonas europaea/eutropha-related cells dominated the AOB and Candidatus Brocadia fulgida-related cells dominated the anammox guild. New carriers were initially colonized by AOB, followed by anammox bacteria proliferating in the deeper biofilm layers, probably in anaerobic microhabitats created by AOB activity. Mature biofilms showed a pronounced three-dimensional stratification where AOB dominated closer to the biofilm-water interface, whereas anammox were dominant deeper into the carrier space and towards the walls. Our results suggest that current mathematical models may be oversimplifying these three-dimensional systems and unless the multidimensionality of these systems is considered, models may result in suboptimal design of MBBR carriers.

    • This roadmap presents the transformational research ideas proposed by "BATTERY 2030+," the European large-scale research initiative for future battery chemistries. A "chemistry-neutral" roadmap to advance battery research, particularly at low technology readiness levels, is outlined, with a time horizon of more than ten years. The roadmap is centered around six themes: 1) accelerated materials discovery platform, 2) battery interface genome, with the integration of smart functionalities such as 3) sensing and 4) self-healing processes. Beyond chemistry related aspects also include crosscutting research regarding 5) manufacturability and 6) recyclability. This roadmap should be seen as an enabling complement to the global battery roadmaps which focus on expected ultrahigh battery performance, especially for the future of transport. Batteries are used in many applications and are considered to be one technology necessary to reach the climate goals. Currently the market is dominated by lithium-ion batteries, which perform well, but despite new generations coming in the near future, they will soon approach their performance limits. Without major breakthroughs, battery performance and production requirements will not be sufficient to enable the building of a climate-neutral society. Through this "chemistry neutral" approach a generic toolbox transforming the way batteries are developed, designed and manufactured, will be created.

  • Isolation and sequencing of the replication region of plasmid pBFp1 isolated from a marine biofilm

    • A 24 kb plasmid, pBFp1, encoding mercury resistance was previously isolated from a marine biofilm. Isolation and sequencing of a 4280 bp DNA fragment containing the plasmid replicon (rep-pBFp1) revealed a putative open reading frame encoding a RepA protein and an oriV-like region containing an A + T rich sub-region, iterons, and DnaA boxes. Sequence comparisons showed significant similarities to the incW plasmid pSa both for the RepA amino acid sequence and in the iteron DNA sequence. Plasmid pBFp1 was also shown to be incompatible with pSa in standard incompatibility testing. A probe from the repA gene of pBFp1 was further made and tested on a collection of plasmids exogenously isolated from marine habitats in a previous study

    • Reinforced concrete has been employed worldwide as a leading building material for public and private structures as well as in modern sculptural art. Although the unrivalled mechanical strength and modelling versatility of this material, several interrelated processes are responsible for its progressive degradation (e.g., carbonation, penetration of aging-promoting agents), decreasing its long-last durability and representing a risk for the public security or the cultural heritage. With the aim to tackle this issue, the present work reports a novel configuration of a screen-printed sensor, obtained by the combination of flexible and robust polyester support and wax-printed filter paper device for the direct application on the concrete surface. Our sensor consists of a polyester-printed three-electrochemical cell that allows dual measurements on reinforced concrete, namely (i) the evaluation of corrosion probability of the metallic reinforcements (which outperforms the half-cell potential standard method) and (ii) the employment of a pH-sensitive iridium oxide film for the measurement of the pH of concrete. The paper was used as a porous material capable of ensuring the electrochemical connection between the Ag/AgCl printed electrode and the concrete solid matrix, acting also as a protective envelope for the electrode. After the laboratory tests, which revealed the noteworthy performances of the sensors in distinguishing among different levels of corrosion as well as measuring the pH of concrete, the developed sensor was applied for on-site measurement at the Giacomo Manzu Museum (Ardea, Italy), demonstrating its suitability for the real application to cultural heritage conservation. Overall, this easy-to-handle and non-invasive diagnostic device provides an innovative analytical approach for the on-site and prompt multiparametric monitoring of the physico-chemical phenomena that endanger the long-lasting preservation of reinforced concrete structures.

    • Corrosion occurring in reinforced concrete has turned into a primary concern of the current century, concrete being the most ubiquitous and predominant material used in the construction industry. Among the many interrelated processes that trigger corrosion of metallic reinforcements, the penetration of chloride ions into the concrete matrix is the most insidious threat. Herein, we developed the first electrochemical device entirely made of paper that allows for the direct, prompt, and noninvasive evaluation of free chloride ion contamination in concrete-based constructions. Our device is based on a three-layer wax-modified filter paper, consisting of two Ag/AgCl screen-printed electrodes that are interfaced by a junction pad in a sandwich-like configuration. Filter paper allows for generating a vertical-flow potentiometric device capable of measuring the electrochemical potential between two solutions containing different concentrations of chloride ions, which are separately drop-cast on the top and bottom layers. After demonstrating the analytical performance of the device, the same principle was applied to the evaluation of the chloride contents in different concrete samples, exploiting paper as a suitable interfacing material for potentiometric measurements on the cement solid surface. Laboratory-prepared concrete samples with known chloride contents were first assessed, and then, the paper-based vertical-flow device was applied to real concrete structures at the Giacomo Manzu Museum (Ardea, Italy) for the evaluation of chloride contamination caused by the proximity to the seaside. The capability of our device to provide timely warning of the risk conditions of concrete-based artifacts was demonstrated.
