  • River Flood Detection Using Passive Microwave Remote Sensing in a Data-Scarce Environment: A Case Study for Two River Basins in Malawi

    • Detecting and forecasting riverine floods is of paramount importance for adequate disaster risk management and humanitarian response. However, this is challenging in data-scarce and ungauged river basins in developing countries. Satellite remote sensing data offers a cost-effective, low-maintenance alternative to the limited in-situ data when training, parametrizing and operating flood models. Utilizing the signal difference between a measurement (M) and a dry calibration (C) location in Passive Microwave Remote Sensing (PMRS), the resulting rcm index simulates river discharge in the measurement pixel. Whilst this has been demonstrated for several river basins, it is as of yet unknown at what ratio of the spatial scales of the river width vs. the PMRS pixel resolution it remains effective in East-Africa. This study investigates whether PMRS imagery at 37 GHz can be effectively used for flood preparedness in two small-scale basins in Malawi, the Shire and North Rukuru river basins. Two indices were studied: The m index (rcm expressed as a magnitude relative to the average flow) and a new index that uses an additional wet calibration cell: rcmc. Furthermore, the results of both indices were benchmarked against discharge estimates from the Global Flood Awareness System (GloFAS). The results show that the indices have a similar seasonality as the observed discharge. For the Shire River, rcmc had a stronger correlation with discharge (ρ = 0.548) than m (ρ = 0.476), and the former predicts discharge more accurately (R2 = 0.369) than the latter (R2 = 0.245). In Karonga, the indices performed similarly. The indices do not perform well in detecting individual flood events when comparing the signal to a flood impact database. However, these results are sensitive to the threshold used and the impact database quality. The method presented simulated Shire River discharge and detected floods more accurately than GloFAS. It therefore shows potential for river monitoring in data-scarce areas, especially for rivers of a similar or larger spatial scale than the Shire River. Upstream pixels could not directly be used to forecast floods occurring downstream in these specific basins, as the time lag between discharge peaks did not provide sufficient warning time. © Copyright © 2021 Mokkenstorm, van den Homberg, Winsemius and Persson.

    • A novel heating efficiency analysis of the microwave heated stop-flow (i.e. stagnant liquid) and continuous-flow reactors has been presented. The thermal losses to the surrounding air by natural convection have been taken into account for heating efficiency calculation of the microwave heating process. The effect of the load diameter in the range of 4–29 mm onthe heating efficiency of ethylene glycol was studied in a single mode microwave cavity under continuous flow and stop-flow conditions. The variation of the microwave absorbing properties of the load with temperature was estimated. Under stop-flow conditions, the heating efficiency depends on the load diameter. The highest heating efficiency has been observed at the load diameter close to the half wavelength of the electromagnetic field in the corresponding medium. Under continuous-flow conditions, the heating efficiency increased linearly. However,microwave leakage above the propagation diameter restricted further experimentation at higher load diameters. Contrary to the stop-flow conditions, the load temperature did not raise monotonously from the inlet to outlet under continuous-flow conditions. This was due to the combined effect of lagging convective heat fluxes in comparison to volumetric heating. This severely disturbs the uniformity of the electromagnetic field in the axial direction and creates areas of high and low field intensity along the load length decreasing the heating efficiency as compared to stop-flow conditions.

  • Pyrrolizidine Alkaloids Disturb Bile Acid Homeostasis in the Human Hepatoma Cell Line HepaRG

    • 1,2-unsaturated pyrrolizidine alkaloids (PAs) belong to a group of secondary plant metabolites. Exposure to PA-contaminated feed and food may cause severe hepatotoxicity. A pathway possibly involved in PA toxicity is the disturbance of bile acid homeostasis. Therefore, in this study, the influence of four structurally different PAs on bile acid homeostasis was investigated after single (24 h) and repeated (14 days) exposure using the human hepatoma cell line HepaRG. PAs induce a downregulation of gene expression of various hepatobiliary transporters, enzymes involved in bile acid synthesis, and conjugation, as well as several transcription regulators in HepaRG cells. This repression may lead to a progressive impairment of bile acid homeostasis, having the potential to accumulate toxic bile acids. However, a significant intracellular and extracellular decrease in bile acids was determined, pointing to an overall inhibition of bile acid synthesis and transport. In summary, our data clearly show that PAs structure-dependently impair bile acid homeostasis and secretion by inhibiting the expression of relevant genes involved in bile acid homeostasis. Furthermore, important biliary efflux mechanisms seem to be disturbed due to PA exposure. These mole-cular mechanisms may play an important role in the development of severe liver damage in PA-intoxicated humans.

  • Molecular Characterisation of Titin N2A and Its Binding of CARP Reveals a Titin/Actin Cross-linking Mechanism

    • Striated muscle responds to mechanical overload by rapidly up-regulating the expression of the cardiac ankyrin repeat protein, CARP, which then targets the sarcomere by binding to titin N2A in the I-band region. To date, the role of this interaction in the stress response of muscle remains poorly understood. Here, we characterise the molecular structure of the CARP-receptor site in titin (UN2A) and its binding of CARP. We find that titin UN2A contains a central three-helix bundle fold (ca 45 residues in length) that is joined to N- and C-terminal flanking immunoglobulin domains by long, flexible linkers with partial helical content. CARP binds titin by engaging an a-hairpin in the three-helix fold of UN2A, the C-terminal linker sequence, and the BC loop in Ig81, which jointly form a broad binding interface. Mutagenesis showed that the CARP/N2A association withstands sequence variations in titin N2A and we use this information to evaluate 85 human single nucleotide variants. In addition, actin co-sedimentation, co-transfection in C2C12 cells, proteomics on heart lysates, and the mechanical response of CARP-soaked myofibrils imply that CARP induces the cross-linking of titin and actin myofilaments, thereby increasing myofibril stiffness. We conclude that CARP acts as a regulator of force output in the sarcomere that preserves muscle mechanical performance upon overload stress. Crown Copyright (C) 2021 Published by Elsevier Ltd.
