    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • This paper presents YouPower, an open source platform designed to make people more aware of their energy consumption and encourage sustainable consumption with local communities. The platform is designed iteratively in collaboration with users in the Swedish and Italian test sites of the project to improve the design and increase active user participation. The community-oriented design is composed of parts that link energy data to energy actions, provide comparisons at different levels, generate dynamic time-of-use signals, offer energy conservation suggestions, and support social sharing. The goal is to bridge people's attitude-behavior gap in energy consumption and to facilitate the behavior change process towards sustainable energy consumption that is implementable in people's daily life. Preliminary results show that community-oriented energy intervention has the potential to improve user engagement significantly.

  • C-extended : Extending the erasmus experience beyond mobility

    • The EU innovation effort to address grand societal challenges is facing an increasing divide. The C-Extended Erasmus+ Strategic Partnership project has the vision to help join this divide by extending the Erasmus experience beyond geographical mobility. It aims to enable stronger contamination between diverse knowledge domains on the one hand, and to foster tighter cooperation between higher education institutions and public/private organizations on the other hand. To this end, mobility opportunities have been explored and piloted through a new mobility scheme, called IIBMS, that results from evolution of existing programmes combined with blended learning, in order to engage students in interdisciplinary and intersectoral educational paths. This paper presents this scheme and discusses the rationale of the design, the current state of development and the prospective outcome of the project.

    • A recyclable and regenerable magnetic polysaccharide absorbent for methylene blue (MB) removal was prepared by coating magnetic polyethyleneimine nanoparticles (PEI@MNPs) with sulfonated chitosan (SCS) and further cross -linked with glutaraldehyde. The driving force for coating is the electrostactic interaction between positively charged PEI and negatively charged SCS. Infrared spectra, zeta potential, thermal gravimetric analysis and X-ray diffraction demonstrated the successful synthesis of magnetic polysaccharide absorbent. The self-assembly of polysaccharide with magnetic nanopartices did not alter the saturation magnetization value of the absorbent confirmed by vibrating sample magnetometer. The nanoparticles showed fast removal (about 30 min reached equilibrium) of MB. In particular, the removal ability of MB after desorption did not reduce, demonstrating an excellent regeneration ability. Our study provides new insights into utilizing polysaccharides for environmental remediation and creating advanced magnetic materials for various promising applications.

    • BACKGROUND: Intestinal injury is a key feature in sepsis. Inhibitors of heat shock protein 90 (Hsp90) have been shown to exert protective effects in models of inflammation. Herein, we hypothesized that Hsp90 might regulate intestinal inflammation and leakage in abdominal sepsis. MATERIALS AND METHODS: Male C57BL/6 mice were pretreated with radicicol (60 mg/kg), which is a specific inhibitor of Hsp90, prior to cecal ligation and puncture (CLP). Intravital fluorescence microscopy was used to quantify leukocyte-endothelium interactions in the colonic microcirculation 6 h after CLP. Colonic tissue was harvested to determine levels of myeloperoxidase, tumor necrosis factor-α and CXC chemokines. Intestinal injury was examined by histology. Intestinal barrier function was quantified by leakage of fluorescein isothiocyanate-dextran from the vascular system out into the abdominal cavity after intravenous injection. RESULTS: We found that radicicol significantly decreased CLP-induced leukocyte rolling and adhesion in colonic venules. Inhibition of Hsp90 reduced colonic levels of myeloperoxidase by 24% in septic animals. Moreover, radicicol significantly decreased CLP-provoked formation of CXC chemokines but had no significant effect on tumor necrosis factor-α levels in the colon. Notably, Hsp90 inhibition significantly attenuated intestinal tissue injury evoked by CLP. Lastly, it was found that radicicol reduced sepsis-induced intestinal leakage by 43%. CONCLUSION: Our novel findings suggest that targeting Hsp90 protects against intestinal inflammation and leakage and might be a useful strategy to ameliorate intestinal failure in polymicrobial sepsis.
