    • Myocardial triglycerides stored in lipid droplets are important in regulating the intracellular delivery of fatty acids for energy generation in mitochondria, for membrane biosynthesis, and as agonists for intracellular signaling. Previously, we showed that deficiency in the lipid droplet protein perilipin 5 (Plin5) markedly reduces triglyceride storage in cardiomyocytes and increases the flux of fatty acids into phospholipids. Here, we investigated whether Plin5 deficiency in cardiomyocytes alters mitochondrial function. We found that Plin5 deficiency reduced mitochondrial oxidative capacity. Furthermore, in mitochondria from Plin5((-/)(-)) hearts, the fatty acyl composition of phospholipids in mitochondrial membranes was altered and mitochondrial membrane depolarization was markedly compromised. These findings suggest that mitochondria isolated from hearts deficient in Plin5, have specific functional defects.

  • Hypoxic regulation of secreted proteoglycans in macrophages.

    • Macrophages are prominent in hypoxic areas of atherosclerotic lesions, and their secreted proteoglycans (PG), such as versican, can modulate the retention of lipoproteins and the activity of enzymes, cytokines, and growth factors involved in atherogenesis. In this study, we report the effects of hypoxia on PG secreted by human monocyte-derived macrophages (HMDM) and the potential regulation by the transcription factor hypoxia-inducible factor (HIF-1alpha and HIF-2alpha). We found that versican co-localized with HIF-1alpha in macrophage-rich areas in human advanced atherosclerotic lesions. Versican and perlecan mRNA expression increased after exposure to 0.5% O(2) (hypoxia) compared with 21% O(2) (control cells). Using precursors to GAG biosynthesis combined with immunoabsorption with a versican antibody an increased versican synthesis was detected at hypoxia. Furthermore, siRNA knockdown of HIF-1alpha and HIF-2alpha in THP-1 cells showed that the hypoxic induction of versican and perlecan mRNA expression involved HIF signaling. Versican expression was co-regulated by HIF-1alpha and HIF-2alpha but expression of perlecan was influenced only by HIF-1alpha and not by HIF-2alpha knockdown. The results show that oxygen concentration is an important modulator of PG expression in macrophages. This may be a novel component of the complex role of macrophages in atherosclerosis.

  • Perilipin 5 is protective in the ischemic heart

    • Background: Myocardial ischemia is associated with alterations in cardiac metabolism, resulting in decreased fatty acid oxidation and increased lipid accumulation. Here we investigate how myocardial lipid content and dynamics affect the function of the ischemic heart, and focus on the role of the lipid droplet protein perilipin 5 (Plin5) in the pathophysiology of myocardial ischemia. Methods and results: We generated Plin5(-/-) mice and found that Plin5 deficiency dramatically reduced the triglyceride content in the heart. Under normal conditions, Plin5(-/-) mice maintained a close to normal heart function by decreasing fatty acid uptake and increasing glucose uptake, thus preserving the energy balance. However, during stress or myocardial ischemia, Plin5 deficiency resulted in myocardial reduced substrate availability, severely reduced heart function and increased mortality. Importantly, analysis of a human cohort with suspected coronary artery disease showed that a common noncoding polymorphism, rs884164, decreases the cardiac expression of PLIN5 and is associated with reduced heart function following myocardial ischemia, indicating a role for Plin5 in cardiac dysfunction. Conclusion: Our findings indicate that Plin5 deficiency alters cardiac lipid metabolism and associates with reduced survival following myocardial ischemia, suggesting that Plin5 plays a beneficial role in the heart following ischemia.     • CD14 is a predictor of inflammation and associated with atherosclerosis. We analyzed 118 carotid plaques from patients with symptomatic carotid artery stenosis for expression of the macrophage markers CD14, CD68 and the angiotensin II type 1 receptor (AT(1)-R). CD14 staining was significantly increased in thrombotic carotid plaques. AT1-R staining was found in macrophage-rich areas, and AT1-R mRNA was detected in plaque macrophages isolated with anti-CD14 immunobeads. In patients treated with an angiotensin receptor blocker, expression of CD14 and CD68 in carotid plaque and serum levels of inflammatory markers were lower than in untreated patients. In vitro, expression of CD14 in human monocyte-derived macrophages was increased by exposure to lipopolysaccharide and decreased by exposure to an angiotensin receptor blocker. Thus, inhibition of the innate immune responsive lipopolysaccharide receptor CD14 in macrophages, rather than AT(1)-R inhibition, may help explain the anti-inflammatory effects of angiotensin receptor blockade.     • The innate immune system and, in particular, activation of the multi-protein complex known as the inflammasome complex are involved in ischemic injury in myocardial cells. The nucleotide-binding leucine-rich repeat-containing pyrin receptor 3 (NLRP3) inflammasome has been linked to inflammation and NLRP3 is especially important for increased inflammation in atherosclerosis, which may lead to myocardial infarction. Here we investigated how inflammasome molecules are affected in human ischemic heart tissue. Surprisingly the important member of the inflammasome complex, NLRP3, displayed markedly decreased levels in human ischemic heart tissue compared with non ischemic control heart tissue. However, subsequent gene analysis revealed mutations in NLRP3 in human ischemic heart tissues but not in non-ischemic control tissue. Gene polymorphisms in the NLRP3 inflammasome have been shown to be associated with increased IL-1β and IL-18 production and severe inflammation. The autoinflammatory disorder familial Mediterranean fever (FMF) is associated with decreased expression of the Mediterranean fever gene (MEFV) and increased inflammation. We also observed reduced expression of MEFV in ischemic versus non-ischemic heart tissue. Further analyses showed a mutation in MEFV in human ischemic heart tissue but not in non-ischemic control tissue. Our data show that defects in the inflammasome and associated proteins may be involved in promoting ischemic heart disease.

  • New food waste data for reference year 2022 from manufacturing sector and from retail and distribution sector in Sweden : Deliverable “D1.1 Project report” of the project SWEFOODWASTE

    • The purpose of this project was to improve the capacity and quality of food waste reporting in Sweden, specifically for two sectors: Processing and Manufacturing, and Retail and other distribution of food.During the first reporting year (reference year 2020) of the collection of food waste data, several challenges were found within these two sectors. In the Processing and Manufacturing sector, there was a need for improvement due to uncertainties according to the EU food waste definition in terms of what residues ought to be classified as food waste, food loss, by-product or other waste. There were also uncertainties how water content should be reported according to the EU food waste reporting obligation. For the Retail and other distribution of food sector, data from only few actors were available for wholesalers, convenience stores, and e-stores which affected data quality.Processing and ManufacturingTo improve reporting and data quality from the food processing and manufacturing sector, a survey was produced asking about food waste and other residues that may constitute food losses. This was to get self-reported data from a large random sample of respondents and to better understand food loss and waste flows in this sector.The survey of Processing and Manufacturing revealed a large volume of previously unknown food waste. 305 000 tonnes of food waste were reported for reference year 2022 compared to 53 000 tonnes for 2020. This is attributed to a better representation of the reality rather than an actual increase in food waste generation.Amounts of food losses seem to be even larger. However, these amounts are also very uncertain due to few respondents from most sectors. Apart from food waste, the dairy and ice cream sector accounted for 180 500 tonnes of food loss, which mainly is whey that goes to feed. Food loss from other sectors cannot be published due to high uncertainties and risk or revealing respondents.The new results on food waste generation, corresponding to 29 kg food waste per capita 2022, align better with international figures. In a European context, the Swedish food waste generation in the industry was low, with 5 kg per capita in 2020 compared to the EU27 average of 27 kg per capita.The new method of self-reporting by respondents improves the accuracy of results, as they know their own operations best. However, there was a challenge in aligning the new EU common definition of food waste with the common understanding of food waste in the Swedish food sector.The different types of food waste to include in the food waste definition were broadened to better align with the intent of EU legislation. The EU definition was for this study interpreted to include most parts of animals and plants that have entered processing as food and become waste. This is a change from former interpretation and the main reason for accounting larger amounts of food waste than previous years. The survey was also designed to enable the distribution of the resulting quantities between different subsectors of manufacturing and processing, as they are grouped by “NACE-codes” in the Business register. Retail and other distribution of foodMost national and regional wholesalers, convenience store chains and e-stores were contacted but unfortunately only a few were able or willing to provide data. This study revealed 13 800 tonnes of food waste from these businesses 2022. This is less compared to 18 200 tonnes in 2021. Note that both these numbers exclude retail and that no scaling occurred for wholesale or e-stores. Although several smaller companies started sharing data, a few large wholesalers did not submit data for 2022 as in 2021 which is the reason for the decrease of reported amounts.Data collection in the wholesale sector yielded limited insights due to only a few companies providing data, and mostly aggregated data. This seems to be due to low technical maturity of their internal data collection and sharing. However, these companies collectively constitute a substantial portion of the wholesale sector in Sweden. It is challenging to precisely quantify their contribution, since many food industry and retail entities also engage in wholesale activities without being formally registered as wholesalers.Convenience stores provided data that could be upscaled to the national level together with data from retail, showing that these do not add up much food waste compared to retail stores. For specialized e-stores selling food, their contribution to food waste is estimated to be minimal, as their operational models and product types result in low waste generation compared to other retail sectors. A large portion of e-stores selling food are part of the retail sector and already included in reported food waste amounts from retail.

  • Augmented levels of CD44 in macrophages from atherosclerotic subjects: a possible IL-6-CD44 feedback loop?

    • The cell-adhesion molecule CD44 likely participates in atherosclerosis development. We have shown previously that pro-inflammatory cytokines affect CD44 expression. Therefore, this work examined the role of elevated CD44 levels in human macrophages. Macrophages from human atherosclerotic subjects (n=15) showed elevated levels of CD44 transcript and protein (1.5-fold) compared to matched controls (n=15) (P=0.050 and 0.044, respectively). To test whether genetic factors influence CD44 expression, two single nucleotide polymorphisms in the CD44 gene were analyzed but these were not associated with coronary artery disease. We also examined the potential connection between plasma cytokine levels and CD44 expression. In atherosclerotic subjects, elevated CD44 expression correlates (P=0.012) with enhanced macrophage IL-6 secretion (3.13+/-2.5 pg/mL versus 0.32+/-0.16 pg/mL in controls, P=0.021). Additionally, CD44-deficient mice exhibit less circulating IL-6 than wild-type controls (9.8+/-0.7 pg/mL versus 14.3+/-0.7 pg/mL; P=0.032). Furthermore, IL-6 augments CD44 expression in primary human macrophages after 24 h (P=0.038) and 48 h (P=0.015). Taken together, our data show an IL-6-CD44 feedback loop in macrophages. Such a positive feedback loop may aggravate atherosclerosis development.
