  • Species and abundance of ectoparasitic flies (Diptera) in pied flycatcher nests in Fennoscandia

    • Background: Birds host several ectoparasitic fly species with negative effects on nestling health and reproductive output, and with the capability of transmitting avian blood parasites. Information on the abundance and distribution of the ectoparasitic fly genera Ornithomya (Hippoboscidae) and Protocalliphora (Calliphoridae) in northern Europe is still generally poor, and we thus explored their geographic range and occurrence of these flies in the nests of a common avian model species, the pied flycatcher Ficedula hypoleuca. Methods: Nests of F. hypoleuca were collected from 21 locations across Fennoscandia in summer 2013, across a latitudinal gradient (between 56 degrees N - 70 degrees N) and examined for the presence of fly puparia. Adult specimens of Ornithomya spp. were also collected for species identification. Fly species were identified morphologically and identifications confirmed with DNA barcoding. Results: We found three species: two louse-flies - Ornithomya chloropus and O. avicularia - and one blow-fly, Protocalliphora azurea. The prevalence of O. avicularia was higher in southern latitudes and this species was not encountered beyond 62 degrees N whereas O. chloropus and P. azurea occurred across the whole range of latitudes. The prevalence of O. chloropus further increased with increasing distance from the coast - a pattern not documented before. The three fly species showed no interspecific associations in their prevalence. Conclusions: Our study revealed relatively high prevalence for all the species (O. chloropus 59 %, O. avicularia 20 %, P. azurea 32 %), and an interesting spatial pattern in the prevalence of the two louse fly species. Our sample did not indicate any major range shifts towards the north for the southern species as compared to the information from the past. Morphological identification of O. chloropus did not match with the corresponding sequences published in the GenBank and taxonomy of this group calls for further studies.

    • Enzymes are effective biological catalysts that accelerate almost all metabolic reactions in living organisms. Synthetic modulators of enzymes are useful tools for the study of enzymatic reactions and can provide starting points for the design of new drugs. Here, we report on the discovery of a class of biologically active compounds that covalently modifies lysine residues in human liver pyruvate kinase (PKL), leading to allosteric activation of the enzyme (EC50=0.29 μM). Surprisingly, the allosteric activation control point resides on the lysine residue K282 present in the catalytic site of PKL. These findings were confirmed by structural data, MS/MS experiments, and molecular modelling studies. Altogether, our study provides a molecular basis for the activation mechanism and establishes a framework for further development of human liver pyruvate kinase covalent activators.

    • A large portion of CubeSat projects have either been demonstrations or educational missions, where the science or operational concept has not been in focus. For efficient use of CubeSat platforms and realization of efficient services, either for scientific or commercial purposes, a full end-to-end design is needed, where the operational concept as well as a focused scientific or commercial rationale is taken into consideration. The SEAM project (funded within European Union’s Seventh Framework Programme) addresses parts of this challenge and develops operational concepts as well as on-board systems for scientific missions. The SEAM platform is using S-Band for downlink and uplink and is fully compliant to the CCSDS standards for satellite link services thus allowing compatibility with a global commercial ground station network. The project is led by the Royal Institute of Technology KTH and SSC is an industrial partner in the consortium. The 3U SEAM CubeSat is designed with an operational concept that includes on-board selection of data to download with earth in the loop, and flexible ground network scheduling. The spacecraft S-Band transceiver is full duplex with a downlink data-rate capability of 3 Mbps and uplink capability up to 100 Kbps. The communication link is CCSDS compatible in both directions, and operates with COTS multi-mission ground station equipment. A newly developed on-board module, that integrates mass memory and CCSDS functionality with a direct transceiver interface, is being demonstrated in the project. The data link layer of the CCSDS standards is implemented in hardware while the network layer and the data storage coordination in the mass memory are handled by software. This functionality partitioning ensures high data throughput and performance while providing flexibility in data collection and handling. It is noted that although the satellite is small, the complexity of such spacecraft is fully comparable to scientific microsatellites and its communication systems and operational concept use technology, equipment and procedures often found in much bigger satellites. The SEAM CubeSat is planned to be launched in early 2017 and the presentation will include the latest news from the mission operations.

    • A feasibility study is presented and the achieved key design milestones towards the first application of Chemical Species Tomography by Near-IR Absorption Tomography on a heavy duty, large-bore marine engine to visualise relative mixture strength are described. The engine is equipped with pre-chamber ignition and operates using Liquefied Natural Gas with >88.9% methane content. Operation of the engine under maximum-load conditions is a key design requirement, with peak cylinder pressure and mean temperature exceeding 127,510. Torr (170. bar) and 850. K respectively. The near-IR spectroscopic behaviour of methane is examined for suitable absorption and reference regions for the above application. In particular, the spectroscopic absorption around the 2ν3 transition region near 1666. nm is approximated by simulation using data from the HITRAN database under worst-case conditions. The simulation results are compared with methane spectra experimentally acquired at high temperature and ambient pressure. Interference from other chemical species as well as measurement linearity is also investigated. A 31-laser beam tomographic imaging array is proposed, which has been optimised to achieve higher spatial resolution performance in the vicinity of the pre-chamber's orifices. To enable optical access, a novel, minimally-intrusive method is presented, utilising standard fibre-optics and collimators.
