    • Of the many types of DNA damage, DNA double-strand breaks (DSBs) are probably the most deleterious. Mounting evidence points to an intricate relationship between DSBs and transcription. A cell system in which the impact on transcription can be investigated at precisely mapped genomic DSBs is essential to study this relationship. Here in a human cell line, we map genome-wide and at high resolution the DSBs induced by a restriction enzyme, and we characterize their impact on gene expression by four independent approaches by monitoring steady-state RNA levels, rates of RNA synthesis, transcription initiation and RNA polymerase II elongation. We consistently observe transcriptional repression in proximity to DSBs. Downregulation of transcription depends on ATM kinase activity and on the distance from the DSB. Our study couples for the first time, to the best of our knowledge, high-resolution mapping of DSBs with multilayered transcriptomics to dissect the events shaping gene expression after DSB induction at multiple endogenous sites.

  • Defining Global Benchmarks in Bariatric Surgery A Retrospective Multicenter Analysis of Minimally Invasive Roux-en-Y Gastric Bypass and Sleeve Gastrectomy

    • Objective: To define "best possible'' outcomes for bariatric surgery (BS)(Roux-en-Y gastric bypass [RYGB] and sleeve gastrectomy [SG]). Background: Reference values for optimal surgical outcomes in welldefined low-risk bariatric patients have not been established so far. Consequently, outcome comparison across centers and over time is impeded by heterogeneity in case-mix. Methods: Out of 39,424 elective BS performed in 19 high-volume academic centers from 3 continents between June 2012 and May 2017, we identified 4120 RYGB and 1457 SG low-risk cases defined by absence of previous abdominal surgery, concomitant procedures, diabetes mellitus, sleep apnea, cardiopathy, renal insufficiency, inflammatory bowel disease, immunosuppression, anticoagulation, BMI>50 kg/m(2) and age>65 years. We chose clinically relevant endpoints covering the intra- and postoperative course. Complications were graded by severity using the comprehensive complication index. Benchmark values were defined as the 75th percentile of the participating centers' median values for respective quality indicators. Results: Patients were mainly females (78%), aged 38+/-11 years, with a baseline BMI 40.8 +/- 5.8 kg/m(2). Over 90 days, 7.2% of RYGB and 6.2% of SG patients presented at least 1 complication and no patients died (mortality in nonbenchmark cases: 0.06%). The most frequent reasons for readmission after 90-days following both procedures were symptomatic cholelithiasis and abdominal pain of unknown origin. Benchmark values for both RYGB and SG at 90-days postoperatively were 5.5% Clavien-Dindo grade >= IIIa complication rate, 5.5% readmission rate, and comprehensive complication index <= 33.73 in the subgroup of patients presenting at least 1 grade >= II complication. Conclusion: Benchmark cutoffs targeting perioperative outcomes in BS offer a new tool in surgical quality-metrics and may be implemented in qualityimprovement cycle.     • Conditions for existence and stability of stationary periodic solutions of uncertain periodic systems are addressed in this paper. Existence and harmonic performance are studied using integral quadratic constraints (IQCs) defined on the space of square integrable periodic functions. Stability is investigated using IQCs defined on the usual space of square integrable functions. The analysis results in criteria formulated as affinely parameterized operator inequalities. Methods of convex optimization are used to find feasible parameters. Two applications will be discussed. The first is harmonic analysis of an electronic circuit and the second involves estimation of the amplitude of a periodic disturbance in a nonlinear control system.

    • Dither signals are commonly used in electronics for implementing different type of modulations in power converters, which represent a very interesting class of hybrid systems. It was recently shown that a non-smooth dithered system can be approximated by an averaged system if the dither frequency is sufficiently high and that the amplitude distribution function of the dither is continuous and has bounded derivative. In power electronics, a large class of systems can be modeled as hybrid dynamical systems due to the presence of switching devices (for instance diodes or transistors) that "instantaneously" change the dynamical behavior of the system. This class of systems can be adequately represented by hybrid systems or nonlinear differential equations with discontinuous nonlinearities. One of the standard approaches for designing control algorithms for power converters is to use modulation schemes (e.g. pulse width modulation, PWM).

    • Averaging of fast switching systems is an effective technique used in many engineering applications. Practical stability and control design for a nonsmooth switched system can be inferred by analyzing the smooth averaged system. In this paper we overview the few formal approaches proposed in the literature to deal with the averaging of nonsmooth systems. The dithering, the phasor dynamics and the hybrid framework techniques are recast and compared by considering pulse-modulated switched linear systems as the common modeling platform.

    • Switched systems provide a tempting modelling framework for power electronics converters. At a certain level of abstraction, switched models are able to capture some interesting properties of the converters behaviour while ignoring irrelevant details. Switched models of power converter have rich dynamical behaviour, so the formal analysis of these systems could be difficult. An effective widely used approach to circumvent some of these difficulties is the averaging technique. In this chapter, the formal approaches proposed in the literature to deal with the averaging of switched systems are presented. The general averaging over a moving time window, the dithering and the dynamic phasor models are recast and compared by considering models for various type of power converters as a common framework.
