A cis-acting structural variation at the ZNF558 locus controls a gene regulatory network in human brain development

- The human forebrain has expanded in size and complexity compared to chimpanzees despite limited changes in protein-coding genes, suggesting that gene expression regulation is an important driver of brain evolution. Here, we identify a KRAB-ZFP transcription factor, ZNF558, that is expressed in human but not chimpanzee forebrain neural progenitor cells. ZNF558 evolved as a suppressor of LINE-1 transposons but has been co-opted to regulate a single target, the mitophagy gene SPATA18. ZNF558 plays a role in mitochondrial homeostasis, and loss-of-function experiments in cerebral organoids suggests that ZNF558 influences developmental timing during early human brain development. Expression of ZNF558 is controlled by the size of a variable number tandem repeat that is longer in chimpanzees compared to humans, and variable in the human population. Thus, this work provides mechanistic insight into how a cis-acting structural variation establishes a regulatory network that affects human brain evolution.

- MicroRNAs (miRNAs) have been implicated in regulating multiple processes during brain development in various species. However, the function of miRNAs in human brain development remains largely unexplored. Here, we provide a comprehensive analysis of miRNA expression of regionalized neural progenitor cells derived from human embryonic stem cells and human foetal brain. We found miR-92b-3p and miR-130b-5p to be specifically associated with neural progenitors and several miRNAs that display both age-specific and region-specific expression patterns. Among these miRNAs, we identified miR-10 to be specifically expressed in the human hindbrain and spinal cord, while being absent from rostral regions. We found that miR-10 regulates a large number of genes enriched for functions including transcription, actin cytoskeleton and ephrin receptor signalling. When overexpressed, miR-10 influences caudalization of human neural progenitor cells. Together, these data confirm a role for miRNAs in establishing different human neural progenitor populations. This dataset also provides a comprehensive resource for future studies investigating the functional role of different miRNAs in human brain development.

- The splice variant INS-IGF2 entails the preproinsulin signal peptide, the insulin B-chain, eight amino acids of the C-peptide and 138 unique amino acids from an ORF in the IGF2 gene. The aim of this study was to determine whether levels of specific INS-IGF2 autoantibodies (INS-IGF2A) were related to age at diagnosis, islet autoantibodies, HLA-DQ or both, in patients and controls with newly diagnosed type 1 diabetes. Patients (n = 676), 0-18 years of age, diagnosed with type 1 diabetes in 1996-2005 and controls (n = 363) were analysed for specific INS-IGF2A after displacement with both cold insulin and INS-IGF2 to correct for non-specific binding and identify double reactive sera. GADA, IA-2A, IAA, ICA, ZnT8RA, ZnT8WA, ZnT8QA and HLA-DQ genotypes were also determined. The median level of specific INS-IGF2A was higher in patients than in controls (P < 0.001). Irrespective of age at diagnosis, 19% (126/676) of the patients had INS-IGF2A when the cut-off was the 95th percentile of the controls (P < 0.001). The risk of INS-IGF2A was increased among HLA-DQ2/8 (OR = 1.509; 95th CI 1.011, 2.252; P = 0.045) but not in 2/2, 2/X, 8/8, 8/X or X/X (X is neither 2 nor 8) patients. The association with HLA-DQ2/8 suggests that this autoantigen may be presented on HLA-DQ trans-heterodimers, rather than cis-heterodimers. Autoantibodies reactive with both insulin and INS-IGF2A at diagnosis support the notion that INS-IGF2 autoimmunity contributes to type 1 diabetes.

- Within several cooperative projects, KTH (Royal Institute of Technology), Ovako Steel AB, and MEFOS have investigated the desulphurisation of bearing steel during vacuum degassing. The work includes thermodynamic calculations of the slag-metal equilibrium, CFD modelling of slag-metal reactions, and plant trials. Results from the various studies are presented and discussed in this paper. Models for predicting slag properties (sulphide capacity, viscosity, and oxide activities) in liquid slags as functions of slag composition and temperature have been used for the calculation of data which have been employed in static and dynamic modelling of sulphur refining. The results from static modelling show that the method allows fast and easy evaluation of the theoretical desulphurisation conditions during degassing at Ovako Steel AB, as well as theoretical determination of the parameters that have the greatest influence on the equilibrium sulphur distribution. The conclusion from dynamic modelling is that the vacuum degassing operation can be described dynamically with the present knowledge of sulphide capacity, sulphur distribution, viscosity, and oxide activities of ladle slags if this knowledge is combined with fluid flow modelling to derive the overall kinetics. The presented model approaches have been found useful in understanding the sulphur refining process at Ovako Steel AB. The dynamic modelling concept is also believed to have potential for dynamic descriptions of other slag-metal reactions in steelmaking.

- Förmågan att effektivt utnyttja och kritiskt förhålla sig till olika informationsresurser – ”information literacy” eller informationskompetens – är en central färdighet i det moderna samhället. Det utgör också en allt viktigare del av lärandet i högre utbildning. Syftet med detta paper är tvåfaldigt: 1) att problematisera relationen mellan informationskompetens som en generell färdighet och mer specifika ämnesorienterade kunskaper och färdigheter; 2) ge exempel på och utveckla redskap för lärare att på ett ämnesmässigt integrerat sätt arbeta med, utveckla och examinera studenternas informationskompetens. Ett huvudspår i framställningen är att synliggöra hur träningen i informationskompetens kan stimulera och hjälpa studenterna till fördjupade ämneskunskaper.

- Stigma and discrimination have been identified as important obstacles to the integration of people with mental illness in society. In efforts to reduce stigma and discrimination, health professionals play an important role as they have frequent contact with and responsibility for treatment and rehabilitation of consumers. The aim of the present study was to investigate attitudes towards mental illness and people with mental illness among nursing staff working in psychiatric or somatic care. The sample consisted of 120 registered or assistant nurses who were interviewed about intimacy with mental illness and attitudes about seven different mental illnesses. The results showed that nursing staff in somatic care, to a higher degree than nursing staff in mental health, reported more negative attitudes with regard to people with schizophrenia as being more dangerous and unpredictable. In contrast, professional experience, intimacy with mental illness and type of care organization were found to be more associated with attitudes to specific mental illnesses concerning the prospect of improvement with treatment and the prospect of recovery. In conclusion, attitudes among nursing staff are in several respects comparable with public opinions about mental illness and mentally ill persons. In order to elucidate if negative attitudes about dangerousness and unpredictability of persons with specific mental illnesses are associated with realistic experiences or with prejudices further studies with a qualitative design are suggested.
