  • Ten golden rules for optimal antibiotic use in hospital settings: the WARNING call to action

    • Antibiotics are recognized widely for their benefits when used appropriately. However, they are often used inappropriately despite the importance of responsible use within good clinical practice. Effective antibiotic treatment is an essential component of universal healthcare, and it is a global responsibility to ensure appropriate use. Currently, pharmaceutical companies have little incentive to develop new antibiotics due to scientific, regulatory, and financial barriers, further emphasizing the importance of appropriate antibiotic use. To address this issue, the Global Alliance for Infections in Surgery established an international multidisciplinary task force of 295 experts from 115 countries with different backgrounds. The task force developed a position statement called WARNING (Worldwide Antimicrobial Resistance National/International Network Group) aimed at raising awareness of antimicrobial resistance and improving antibiotic prescribing practices worldwide. The statement outlined is 10 axioms, or "golden rules," for the appropriate use of antibiotics that all healthcare workers should consistently adhere in clinical practice.

    • The ecotropic viral integration site 1 (Evi1) oncogenic transcription factor is one of a number of alternative transcripts encoded by the Mds1 and Evi1 complex locus (Mecom). Overexpression of Evi1 has been observed in a number of myeloid disorders and is associated with poor patient survival. It is also amplified and/or overexpressed in many epithelial cancers including nasopharyngeal carcinoma, ovarian carcinoma, ependymomas, and lung and colorectal cancers. Two murine knockout models have also demonstrated Evi1's critical role in the maintenance of hematopoietic stem cell renewal with its absence resulting in the death of mutant embryos due to hematopoietic failure. Here we characterize a novel mouse model (designated Evi1(fl3)) in which Evi1 exon 3, which carries the ATG start, is flanked by loxP sites. Unexpectedly, we found that germline deletion of exon3 produces a hypomorphic allele due to the use of an alternative ATG start site located in exon 4, resulting in a minor Evi1 N-terminal truncation and a block in expression of the Mds1-Evi1 fusion transcript. Evi1(δex3/δex3) mutant embryos showed only a mild non-lethal hematopoietic phenotype and bone marrow failure was only observed in adult Vav-iCre/+, Evi1(fl3/fl3) mice in which exon 3 was specifically deleted in the hematopoietic system. Evi1(δex3/δex3) knockout pups are born in normal numbers but die during the perinatal period from congenital heart defects. Database searches identified 143 genes with similar mutant heart phenotypes as those observed in Evi1(δex3/δex3) mutant pups. Interestingly, 42 of these congenital heart defect genes contain known Evi1-binding sites, and expression of 18 of these genes are also effected by Evi1 siRNA knockdown. These results show a potential functional involvement of Evi1 target genes in heart development and indicate that Evi1 is part of a transcriptional program that regulates cardiac development in addition to the development of blood.

  • Control of the Oceanic Heat Content of the Getz-Dotson Trough, Antarctica, by the Amundsen Sea Low

    • The changing supply of warm Circumpolar Deep Water (CDW) to the West Antarctic continental shelf is responsible for the basal melting and thinning of the West Antarctic ice shelves that has occurred in recent decades. Here we assess the variability in CDW supply, and its drivers, from a multiyear mooring deployed in, and a regional ocean model spanning, the Getz-Dotson Trough, Amundsen Sea. Between 2010 to 2015, the CDW within the trough underwent a pronounced cooling and freshening, associated with changes in thermohaline properties on isopycnals. Variability in the rate of CDW inflow is controlled by local wind forcing of a shelf break undercurrent, which determines the hydrographic properties of inflowing CDW via tilting of density surfaces above the continental slope. Local wind is coupled to the Amundsen Sea Low (ASL) low-pressure system, which is modulated by large-scale climatic modes via atmospheric teleconnections. For the period analyzed, a deeper ASL was associated with westward wind anomaly at the shelf break. Changes in the sea surface slope decelerated the shelf break undercurrent, resulting in less heat accessing the continental shelf and, consequently, a cooling of the Getz-Dotson Trough. Therefore, the present work suggests that the fate of the West Antarctic ice shelves is closely tied to the future evolution of the ASL.

  • Where shadows lie : reconstruction of anisotropies in the neutrino sky

    • The Cosmic Neutrino Background (CNB) encodes a wealth of information, but has not yet been observed directly. To determine the prospects of detection and to study its information content, we reconstruct the phase-space distribution of local relic neutrinos from the three-dimensional distribution of matter within 200 h-1 Mpc of the Milky Way. Our analysis relies on constrained realization simulations and forward modelling of the 2M++ galaxy catalogue. We find that the angular distribution of neutrinos is anti-correlated with the projected matter density, due to the capture and deflection of neutrinos by massive structures along the line of sight. Of relevance to tritium capture experiments, we find that the gravitational clustering effect of the large-scale structure on the local number density of neutrinos is more important than that of the Milky Way for neutrino masses less than 0.1 eV. Nevertheless, we predict that the density of relic neutrinos is close to the cosmic average, with a suppression or enhancement over the mean of (-0.3%, +7%, +27%) for masses of (0.01, 0.05, 0.1) eV. This implies no more than a marginal increase in the event rate for tritium capture experiments like PTOLEMY. We also predict that the CNB and CMB rest frames coincide for 0.01 eV neutrinos, but that neutrino velocities are significantly perturbed for masses larger than 0.05 eV. Regardless of mass, we find that the angle between the neutrino dipole and the ecliptic plane is small, implying a near-maximal annual modulation in the bulk velocity. Along with this paper, we publicly release our simulation data, comprising more than 100 simulations for six different neutrino masses.

    • [1] Ice shelves are critical features in the debate about West Antarctic ice sheet change and sea level rise, both because they limit ice discharge and because they are sensitive to change in the surrounding ocean. The Pine Island Glacier ice shelf has been thinning rapidly since at least the early 1990s, which has caused its trunk to accelerate and retreat. Although the ice shelf front has remained stable for the past six decades, past periods of ice shelf collapse have been inferred from relict seabed “corrugations” (corrugated ridges), preserved 340 km from the glacier in Pine Island Trough. Here we present high-resolution bathymetry gathered by an autonomous underwater vehicle operating beneath an Antarctic ice shelf, which provides evidence of long-term change in Pine Island Glacier. Corrugations and ploughmarks on a sub-ice shelf ridge that was a former grounding line closely resemble those observed offshore, interpreted previously as the result of iceberg grounding. The same interpretation here would indicate a significantly reduced ice shelf extent within the last 11 kyr, implying Holocene glacier retreat beyond present limits, or a past tidewater glacier regime different from today. The alternative, that corrugations were not formed in open water, would question ice shelf collapse events interpreted from the geological record, revealing detail of another bed-shaping process occurring at glacier margins. We assess hypotheses for corrugation formation and suggest periodic grounding of ice shelf keels during glacier unpinning as a viable origin. This interpretation requires neither loss of the ice shelf nor glacier retreat and is consistent with a “stable” grounding-line configuration throughout the Holocene.

    • Ice Shelves in the Amundsen Sea Embayment have thinned, accelerating the seaward flow of ice sheets upstream over recent decades. This imbalance is caused by an increase in the ocean-driven melting of the ice shelves. Observations and models show that the ocean heat content reaching the ice shelves is sensitive to the depth of thermocline, which separates the cool, fresh surface waters from warm, salty waters. Yet the processes controlling the variability of thermocline depth remain poorly constrained. Here, we quantify the oceanic conditions and ocean-driven melting of Cosgrove, Pine Island Glacier (PIG), Thwaites, Crosson and Dotson ice shelves in the Amundsen Sea Embayment from 1991 to 2014 using a general circulation model. Ice shelf melting is coupled to variability in the wind field and the sea-ice motions over the continental shelf break and associated onshore advection of warm waters in deep troughs. The layer of warm, salty waters at the calving front of PIG and Thwaites is thicker in austral spring (June to October) than in austral summer (December to March), whereas the seasonal cycle at the calving front of Dotson is reversed. Furthermore, the ocean-driven melting in PIG is enhanced by an asymmetric response to changes in ocean heat transport anomalies at the continental shelf break: melting responds more rapidly to increases in ocean heat transport than to decreases. This asymmetry is caused by the inland deepening of bathymetry and the glacial meltwater circulation around the ice shelf.

    • The large rotating Coriolis platform was used to investigate oceanographic flows of particular importance in the Antarctic region. Topographic features (such as canyons and ridges) on continental slopes were found to direct dense overflows downslope and increase the entrainment of ambient water into the overflow, as compared to flows on smooth slopes. In cases where dense flows on slopes generate eddies in the overlying fluid, simulated ice shelves were found to present a substantial obstacle to the flow, diverting the eddies downslope and impeding the flow of dense water into the ice shelf cavity. However, if the ambient water was stratified (as is the case in the winter around Antarctica) the ice shelf may not obstruct the flow, leading to seasonal differences in the flow of dense water into the ice shelf cavities. The exchange of flow between ice shelf cavities and the open water, driven by the generation of dense High Salinity Shelf Water (HSSW) outside the cavity and relatively light meltwater within the cavity, was also modelled.

    • We present SIBELIUS-DARK, a constrained realization simulation of the local volume to a distance of 200 Mpc from the Milky Way. SIBELIUS-DARK is the first study of the ‘Simulations Beyond The Local Universe’ (SIBELIUS) project, which has the goal of embedding a model Local Group-like system within the correct cosmic environment. The simulation is dark-matter-only, with the galaxy population calculated using the semi-analytic model of galaxy formation, GALFORM. We demonstrate that the large-scale structure that emerges from the SIBELIUS constrained initial conditions matches well the observational data. The inferred galaxy population of SIBELIUS-DARK also match well the observational data, both statistically for the whole volume and on an object-by-object basis for the most massive clusters. For example, the K-band number counts across the whole sky, and when divided between the northern and southern Galactic hemispheres, are well reproduced by SIBELIUS-DARK. We find that the local volume is somewhat unusual in the wider context of ΛCDM: it contains an abnormally high number of supermassive clusters, as well as an overall large-scale underdensity at the level of ≈5 per cent relative to the cosmic mean. However, whilst rare, the extent of these peculiarities does not significantly challenge the ΛCDM model. SIBELIUS-DARK is the most comprehensive constrained realization simulation of the local volume to date, and with this paper we publicly release the halo and galaxy catalogues at z = 0, which we hope will be useful to the wider astronomy community.
