  • Parameter and modeling uncertainty simulated by GLUE and a formal Bayesian method for a conceptual hydrological model

    • Quantification of uncertainty of hydrological models has attracted much attention in hydrologic research in recent years. Many methods for quantification of uncertainty have been reported in the literature, of which GLUE and formal Bayesian method are the two most popular methods. There have been many discussions in the literature concerning differences between these two methods in theory (mathematics) and results, and this paper focuses on the computational efficiency and differences in their results, but not on philosophies and mathematical rigor that both methods rely on. By assessing parameter and modeling uncertainty of a simple conceptual water balance model (WASMOD) with the use of GLUE and formal Bayesian method, the paper evaluates differences in the results of the two methods and discusses the reasons for these differences. The main findings of the study are that: (1) the parameter posterior distributions generated by the Bayesian method are slightly less scattered than those by the GLUE method; (2) using a higher threshold value (>0.8) GLUE results in very similar estimates of parameter and model uncertainty as does the Bayesian method; and (3) GLUE is sensitive to the threshold value used to select behavioral parameter sets and lower threshold values resulting in a wider uncertainty interval of the posterior distribution of parameters, and a wider confidence interval of model uncertainty.   • Regionalization study of a conceptual hydrological model in Dongjiang basin, south China

    • The seasonal variability and spatial distribution of precipitation are the main cause of flood and drought events. The study of spatial distribution and temporal trend of precipitation in river basins has been paid more and more attention. However, in China, the precipitation data are measured by weather stations (WS) of China Meteorological Administration and hydrological rain gauges (RG) of national and local hydrology bureau. The WS data usually have long record with fewer stations, while the RG data usually have short record with more stations. The consistency and correlation of these two data sets have not been well understood. In this paper, the precipitation data from 30 weather stations for 1958-2007 and 248 rain gauges for 1995-2004 in the Haihe River basin are examined and compared using linear regression, 5-year moving average, Mann-Kendall trend analysis, Kolmogorov-Smirnov test, Z test and F test methods. The results show that the annual precipitation from both WS and RG records are normally distributed with minor difference in the mean value and variance. It is statistically feasible to extend the precipitation of RG by WS data sets. Using the extended precipitation data, the detailed spatial distribution of the annual and seasonal precipitation amounts as well as their temporal trends are calculated and mapped. The various distribution maps produced in the study show that for the whole basin the precipitation of 1958-2007 has been decreasing except for spring season. The decline trend is significant in summer, and this trend is stronger after the 1980s. The annual and seasonal precipitation amounts and changing trends are different in different regions and seasons. The precipitation is decreasing from south to north, from coastal zone to inland area.

    • Amyloid fibrils—nanoscale fibrillar aggregates with high levels of order—are pathogenic in some today incurable human diseases; however, there are also many physiologically functioning amyloids in nature. The process of amyloid formation is typically nucleation-elongation-dependent, as exemplified by the pathogenic amyloid-β peptide (Aβ) that is associated with Alzheimer's disease. Spider silk, one of the toughest biomaterials, shares characteristics with amyloid. In this study, it is shown that forming amyloid-like nanofibrils is an inherent property preserved by various spider silk proteins (spidroins). Both spidroins and Aβ capped by spidroin N- and C-terminal domains, can assemble into macroscopic spider silk-like fibers that consist of straight nanofibrils parallel to the fiber axis as observed in native spider silk. While Aβ forms amyloid nanofibrils through a nucleation-dependent pathway and exhibits strong cytotoxicity and seeding effects, spidroins spontaneously and rapidly form amyloid-like nanofibrils via a non-nucleation-dependent polymerization pathway that involves lateral packing of fibrils. Spidroin nanofibrils share amyloid-like properties but lack strong cytotoxicity and the ability to self-seed or cross-seed human amyloidogenic peptides. These results suggest that spidroins' unique primary structures have evolved to allow functional properties of amyloid, and at the same time direct their fibrillization pathways to avoid formation of cytotoxic intermediates.

  • Exome sequencing reveals genetic heterogeneity and clinically actionable findings in children with cerebral palsy

    • Cerebral palsy (CP) is the most common motor disability in children. To ascertain the role of major genetic variants in the etiology of CP, we conducted exome sequencing on a large-scale cohort with clinical manifestations of CP. The study cohort comprised 505 girls and 1,073 boys. Utilizing the current gold standard in genetic diagnostics, 387 of these 1,578 children (24.5%) received genetic diagnoses. We identified 412 pathogenic and likely pathogenic (P/LP) variants across 219 genes associated with neurodevelopmental disorders, and 59 P/LP copy number variants. The genetic diagnostic rate of children with CP labeled at birth with perinatal asphyxia was higher than the rate in children without asphyxia (P = 0.0033). Also, 33 children with CP manifestations (8.5%, 33 of 387) had findings that were clinically actionable. These results highlight the need for early genetic testing in children with CP, especially those with risk factors like perinatal asphyxia, to enable evidence-based medical decision-making. Using exome sequencing data from one of the largest cohorts of children with cerebral palsy, the genetic diagnostic rates of single-nucleotide and copy number variants were assessed and a sizeable fraction found to be clinically actionable.

  • Prognostic value and biological function of LRRN4 in colorectal cancer

    • Background Several nervous and nerve-related biomarkers have been detected in colorectal cancer (CRC) and can contribute to the progression of CRC. However, the role of leucine-rich repeat neuronal 4 (LRRN4), a recently identified neurogenic marker, in CRC remains unclear. Methods We examined the expression and clinical outcomes of LRRN4 in CRC from TCGA-COREAD mRNA-sequencing datasets and immunohistochemistry in a Chinese cohort. Furthermore, colony formation, flow cytometry, wound healing assays and mouse xenograft models were used to investigate the biological significance of LRRN4 in CRC cell lines with LRRN4 knockdown or overexpression in vitro and in vivo. In addition, weighted coexpression network analysis, DAVID and western blot analysis were used to explore the potential molecular mechanism. Results We provide the first evidence that LRRN4 expression, at both the mRNA and protein levels, was remarkably high in CRC compared to controls and positively correlated with the clinical outcome of CRC patients. Specifically, LRRN4 was an independent prognostic factor for progression-free survival and overall survival in CRC patients. Further functional experiments showed that LRRN4 promoted cell proliferation, cell DNA synthesis and cell migration and inhibited apoptosis. Knockdown of LRRN4 can correspondingly decrease these effects in vitro and can significantly suppress the growth of xenografts. Several biological functions and signaling pathways were regulated by LRRN4, including proteoglycans in cancer, glutamatergic synapse, Ras, MAPK and PI3K. LRRN4 knockdown resulted in downregulation of Akt, p-Akt, ERK1/2 and p-ERK1/2, the downstream of the Ras/MAPK signaling pathway, overexpression of LRRN4 leaded to the upregulation of these proteins. Conclusions Our results suggest that LRRN4 could be a biological and molecular determinant to stratify CRC patients into distinct risk categories, and mechanistically, this is likely attributable to LRRN4 regulating several malignant phenotypes of neoplastic cells via RAS/MAPK signal pathways.
