  • Transcriptome-wide association study reveals candidate causal genes for lung cancer

    • We have recently completed the largest GWAS on lung cancer including 29,266 cases and 56,450 controls of European descent. The goal of our study has been to integrate the complete GWAS results with a large‐scale expression quantitative trait loci (eQTL) mapping study in human lung tissues (n = 1,038) to identify candidate causal genes for lung cancer. We performed transcriptome‐wide association study (TWAS) for lung cancer overall, by histology (adenocarcinoma, squamous cell carcinoma and small cell lung cancer) and smoking subgroups (never‐ and ever‐smokers). We performed replication analysis using lung data from the Genotype‐Tissue Expression (GTEx) project. DNA damage assays were performed in human lung fibroblasts for selected TWAS genes. As expected, the main TWAS signal for all histological subtypes and ever‐smokers was on chromosome 15q25. The gene most strongly associated with lung cancer at this locus using the TWAS approach was IREB2 (pTWAS = 1.09E−99), where lower predicted expression increased lung cancer risk. A new lung adenocarcinoma susceptibility locus was revealed on 9p13.3 and associated with higher predicted expression of AQP3 (pTWAS = 3.72E−6). Among the 45 previously described lung cancer GWAS loci, we mapped candidate target gene for 17 of them. The association AQP3‐adenocarcinoma on 9p13.3 was replicated using GTEx (pTWAS = 6.55E−5). Consistent with the effect of risk alleles on gene expression levels, IREB2 knockdown and AQP3 overproduction promote endogenous DNA damage. These findings indicate genes whose expression in lung tissue directly influences lung cancer risk.

  • Biological, clinical and population relevance of 95 loci for blood lipids

    • Plasma concentrations of total cholesterol, low-density lipoprotein cholesterol, high-density lipoprotein cholesterol and triglycerides are among the most important risk factors for coronary artery disease (CAD) and are targets for therapeutic intervention. We screened the genome for common variants associated with plasma lipids in >100,000 individuals of European ancestry. Here we report 95 significantly associated loci (P<5 x 10(-8)), with 59 showing genome-wide significant association with lipid traits for the first time. The newly reported associations include single nucleotide polymorphisms (SNPs) near known lipid regulators (for example, CYP7A1, NPC1L1 and SCARB1) as well as in scores of loci not previously implicated in lipoprotein metabolism. The 95 loci contribute not only to normal variation in lipid traits but also to extreme lipid phenotypes and have an impact on lipid traits in three non-European populations (East Asians, South Asians and African Americans). Our results identify several novel loci associated with plasma lipids that are also associated with CAD. Finally, we validated three of the novel genes-GALNT2, PPP1R3B and TTC39B-with experiments in mouse models. Taken together, our findings provide the foundation to develop a broader biological understanding of lipoprotein metabolism and to identify new therapeutic opportunities for the prevention of CAD.

  • Bandgap Tuning of Silver Bismuth Iodide via Controllable Bromide Substitution for Improved Photovoltaic Performance

    • In this work, silver-bismuth-halide thin films, exhibiting low toxicity and good stability, were explored systemically by gradually substituting iodide, I, with bromide, Br, in the AgBi2I7 system. It was found that the optical bandgap can be tuned by varying the I/Br ratio. Moreover, the film quality was improved when introducing a small amount of Br. The solar cell was demonstrated to be more stable at ambient conditions and most efficient when incorporating 10% Br, as a result of decreased recombination originating from the increased grain size. Thus, replacing a small amount of I with Br was beneficial for photovoltaic performance.

  • Climate control of terrestrial carbon exchange across biomes and continents

    • Understanding the relationships between climate and carbon exchange by terrestrial ecosystems is critical to predict future levels of atmospheric carbon dioxide because of the potential accelerating effects of positive climate-carbon cycle feedbacks. However, directly observed relationships between climate and terrestrial CO2 exchange with the atmosphere across biomes and continents are lacking. Here we present data describing the relationships between net ecosystem exchange of carbon (NEE) and climate factors as measured using the eddy covariance method at 125 unique sites in various ecosystems over six continents with a total of 559 site-years. We find that NEE observed at eddy covariance sites is (1) a strong function of mean annual temperature at mid-and high-latitudes, (2) a strong function of dryness at mid-and low-latitudes, and (3) a function of both temperature and dryness around the mid-latitudinal belt (45 degrees N). The sensitivity of NEE to mean annual temperature breaks down at similar to 16 degrees C (a threshold value of mean annual temperature), above which no further increase of CO2 uptake with temperature was observed and dryness influence overrules temperature influence.

  • An Experimental Investigation of a Multi-Cylinder Engine with Gasoline-Like Fuel towards a High Engine Efficiency

    • Partially Premixed Combustion (PPC) is a promising combustion concept with high thermodynamic efficiency and low emission level, and also with minimal modification of standard engine hardware. To use PPC in a production oriented engine, the optimal intake charge conditions for PPC should be included in the analysis. The experiments in this paper investigated and confirmed that the optimal intake conditions of net indicated efficiency for PPC are EGR between 50% and 55% as possible and the lambda close to 1.4. Heat-transfer energy and exhaust gas waste-energy contribute to the majority of the energy loss in the engine. The low EGR region has high heat-transfer and low exhaust gas enthalpy-waste, while the high EGR region has low heat-transfer and high exhaust gas waste-enthalpy. The optimal EGR condition is around 50% where the smallest energy loss is found as a trade-off between heat transfer and exhaust-gas enthalpy-waste. Lambda close to 1.4 results from the trade-off of high gas-exchange efficiency with low lambda and high thermodynamic efficiency with high lambda. The optimal inlet charge condition was also applied to a multi-cylinder engine from low load to high load. The results indicate that the highest net indicated efficiency of the experimental engine was 49.7% at 13.5 bar IMEPn (Net Indicated Mean Effective Pressure), the highest brake efficiency was 44.2% observed at 17.2 bar IMEPn.

    • Partially Premixed combustion (PPC) is a promising combustion concept to achieve high engine efficiency. The combustion-timing of PPC is affected by both inlet-charge condition and the fuel injection, a simple map-based feed-forward control method is not sufficient for controlling the combustion during transient operation. This paper investigates a model-based control method to solve this control problem. A non-linear model was developed to capture the main trend of inlet-gas condition and injection. A Model Predictive Controller (MPC) was designed to control injection timing and the gas-exchange system. The controller was verified in a multi-cylinder heavy-duty PPC engine.

    • Advanced combustion concepts like partially premixed combustion (PPC) are sensitive to inlet gas conditions especially inlet temperature. To estimate the combustion parameters like ignition delay on a cycle-to-cycle basis, both complementary filter and extended Kalman filter were designed to estimate the inlet temperature, and the ignition delay was calculated with properly estimated temperature. A two valve fast thermal management system was used to control the inlet temperature, and a PI controller with the lambda tuning method was used. The estimation and control algorithm were verified on a six-cylinder heavy duty engine.

    • Partially Premixed Combustion (PPC) is a promising advanced combustion mode for future engines. In order to investigate the sensitivity of PPC to exhaust gas recirculation (EGR) rate, intake gas temperature, intake gas pressure, and injection timing, these parameters were swept individually at three different loads in a single cylinder diesel engine with gasoline-like fuel.A factor of sensitivity was defined to indicate the combustion's controllability and sensitivity to inlet gas parameters and injection timings. Through analysis of experimental results, a control window of inlet gas parameters and injection timings is obtained at different loads in PPC mode from 5 bar to 10 bar IMEPg load at 1200 rpm.To further study the PPC controllability with injection timing, main injection timing was adjusted to sustain steady combustion phasing subject to perturbation of inlet gas state. Experimental results show that the main injection timing can resist the interference of intake parameters and maintain constant combustion phasing. Injection timing control is a promising approach to maintain high engine efficiency and low emission levels during transient operation.

    • Silver bismuth iodide (Ag–Bi–I) light absorbers are interesting candidates as lead-free and low-toxic metal-halide materials for solar cell applications. In this work, the partial exchange of bismuth, Bi, with antimony, Sb, is investigated in samples prepared from a solution targeting stoichiometry AgBi2I7. Samples with a gradually increased exchange of Bi by Sb are prepared and light absorption measurements show that the absorption edge is gradually blue-shifted with increasing the amount of Sb. This trend in the shift in combination with the X-ray diffraction and X-ray photoelectron spectroscopy measurements, suggest that new materials with a mixture of Sb and Bi are formed. The density functional theory based electronic structure calculations reproduce the trend observed in the experiments when including spin–orbit coupling, which indicates the importance of relativistic effects in these materials. X-ray photoelectron spectroscopy is used to characterize the materials, and confirms the exchange of Bi to Sb in the samples. When Sb is included in the material, the grain size changes between 50 and 200 nm and the solar cell performance also changes. An optimal power conversion efficiency with excellent reproducibility and stability is obtained for a solar cell with the ratio of Sb/Bi equal to 3.
