    • Properties of starch triacetate films from different botanical origins were evaluated. Tensile and barrier properties, glass transition temperatures and moisture uptake were measured, and the molecular structure was characterised. High-amylose starches were good film formers, normal potato starch formed continuous but brittle films and potato amylopectin formed very brittle films even after plasticisation. Barley amylopectin did not form continuous films. Different plasticisers were studied and diacetin was shown to perform better than the standard plasticiser triacetin. All films were water resistant, and the best films gave water vapour barriers in the range of films of polylactic acid, cellulose acetates and gluten. The inherent oxygen barrier of native starches was lost when the starches were highly acetylated. Starch triacetates are commonly dissolved in organic solvents such as chloroform. When testing different approaches to dissolve the starch triacetates it was found that acetic acid as well as acetylacetone were good solvents without imposing negative effects on material properties.

Som kungligt slott har Ulriksdal genomgått ombyggnader under 1600 - och 1700-talet, men präglas idag mest av Karl XVs omfattande omdaning på 1850-talet och Gustav VI Adolfs lika genomgripande förändringar på 1920-talet.Slottet har bjudit på rika studiemöjligheter av stilideal, planlösning, interiörgestaltning, snickerikultur och stormtekniska system från hela 350-årsperioden. Genom noggranna uppmätningar och inventeringar på plats kopplade till arkivstudier har slottets komplicerade förändringshistoria kunnat analyseras,

  • A novel reciprocating tribometer for friction and wear measurements with high contact pressure and large area contact configurations

    • There are many moving machine assemblies with conformal tribological contacts at very high contact pressures, e.g., sliding bearings, propeller shaft bearings and machine guideways. Furthermore, applications such as trunnion and guide vane bearing in Kaplan turbines have very low sliding speeds and oscillatory types of motion. Although there is a vast selection of tribology test rigs available, there is still a lack of test equipment to perform friction and wear tests under high contact pressure, reciprocatory sliding and large area contact. The aim of this work is thus to develop a novel reciprocating tribometer and test method that enables friction and wear tests under low-speed reciprocatory sliding with contact pressures up to 90 MPa in a flat-on-flat contact configuration. First, a thorough description of the test rig design is given. Secondly, the influence of contact pressure and stroke length on the tribological properties of a stainless steel and polymer composite material combination is studied. The significance of considering creep, friction during the stroke and contact temperature is specifically highlighted. The novel tribometer can be used to screen different bearing and shaft material combinations and to evaluate the friction and wear performance of self-lubricating bearings for the specific operating conditions found in Kaplan turbines.

    • This study was carried out to evaluate the effect of cellulose-based hydrocolloids on the rheology of flour doughs based on corn starch and potato starch. Adding cellulosebased hydrocolloids or a chemically modified waxy starch to this gluten-free dough imparted a shear-thinning behavior similar to that of wheat flour dough.

    • This paper describes an electrical system layout for a wave power plant connecting linear generators to the grid. The electrical power out from the wave energy converters must be converted before they can be connected to the grid. The conversion is carried out in marine substations that will be placed on the seabed.The paper presents experimental power data from a wave energy converter that has been in operation at the Lysekil research site since March 2006. Moreover, results and analyses from experiments and simulations from tests with the generator connected to a rectifier and filter are presented. A simulation is made to show the difference between having the generator connected to a linear load and a nonlinear load, which would be the case when the generator is connected to the grid.

  • Pan-cancer analysis of whole genomes

    • Cancer is driven by genetic change, and the advent of massively parallel sequencing has enabled systematic documentation of this variation at the whole-genome scale1–3. Here we report the integrative analysis of 2,658 whole-cancer genomes and their matching normal tissues across 38 tumour types from the Pan-Cancer Analysis of Whole Genomes (PCAWG) Consortium of the International Cancer Genome Consortium (ICGC) and The Cancer Genome Atlas (TCGA). We describe the generation of the PCAWG resource, facilitated by international data sharing using compute clouds. On average, cancer genomes contained 4–5 driver mutations when combining coding and non-coding genomic elements; however, in around 5% of cases no drivers were identified, suggesting that cancer driver discovery is not yet complete. Chromothripsis, in which many clustered structural variants arise in a single catastrophic event, is frequently an early event in tumour evolution; in acral melanoma, for example, these events precede most somatic point mutations and affect several cancer-associated genes simultaneously. Cancers with abnormal telomere maintenance often originate from tissues with low replicative activity and show several mechanisms of preventing telomere attrition to critical levels. Common and rare germline variants affect patterns of somatic mutation, including point mutations, structural variants and somatic retrotransposition. A collection of papers from the PCAWG Consortium describes non-coding mutations that drive cancer beyond those in the TERT promoter4; identifies new signatures of mutational processes that cause base substitutions, small insertions and deletions and structural variation5,6; analyses timings and patterns of tumour evolution7; describes the diverse transcriptional consequences of somatic mutation on splicing, expression levels, fusion genes and promoter activity8,9; and evaluates a range of more-specialized features of cancer genomes8,10–18.

    • BACKGROUND: Turnover of RNA is a regulated process that in part controls gene expression. This process is partly controlled by the scavenger decapping enzyme (DcpS). This study aimed to investigate the expression of DcpS in colorectal cancer (CRC) tissue, to evaluate its prognostic significance in patients with CRC and to investigate potentially targeted genes by DcpS.METHODS: Immunohistochemical analysis was used to determine localization of DcpS in normal and CRC tissue, western blot analysis for quantification of protein expression and qPCR for mRNA expression in normal and CRC tissue and expression in cell lines after silencing using siRNA. Gene array analysis was used to study regulation of genes after silencing of DcpS. Proliferation was studied using BRDU.RESULTS: DcpS expression was localized to the epithelial cells of both control and cancer tissue. Tumor and paired control tissue samples from 100 patients who underwent surgical resection for primary colorectal adenocarcinomas were utilized. mRNA and protein of DcpS was significantly up-regulated in the patients with CRC and the mRNA level was higher in rectal cancer tissue compared to colon cancer tissue (p < 0.05). Lowest tertile levels of DcpS mRNA in cancer tissue was associated with a decreased cancer-specific survival rate with a hazard ratio (HR) of 4.7 (95% CI=1.02-12.3), independent of disease stage. The low level of DcpS mRNA was a predictor of poorer survival in patients with rectal and disseminated cancer and in patients receiving adjuvant treatment (p < 0.05). After silencing DcpS in Caco-2 cancer cells, altered expression of several genes associated with RNA, cell cycle regulation, alternative splicing and microRNA was observed and resulted in 23% increase in proliferation.CONCLUSIONS: These results indicate that DcpS has potential as a prognostic factor for CRC but further studies in a broader cohort are warranted to evaluate the significance of the findings in the clinic.
