    • The success of engineering companies is highly dependent on how well product information is managed, engineered and communicated. From marketing through development to after sales activities, data needs to be accessible and used in the best way. Today, geographical distance, the need for close co-operation and data complexity are all natural parts of the working environment. Product modelling techniques are continuously evolving with new requirements and opportunities emerging daily. This paper will outline and discuss some of these trends, and at the same time present some of the areas where research is being carried out within projects in the Product Model Cluster in the national graduate school - The Swedish Engineering Design Research and Education Agenda (ENDREA).

  • High prevalence of pharyngeal bacterial pathogens among healthy adolescents and young adults

    • The pharyngeal mucosa can be colonized with bacteria that have potential to cause pharyngotonsillitis. By the use of culturing techniques and PCR, we aimed to assess the prevalence of bacterial pharyngeal pathogens among healthy adolescents and young adults. We performed a cross-sectional study in a community-based cohort of 217 healthy individuals between 16 and 25 years of age. Samples were analyzed for Group A streptococci (GAS), Group C/G streptococci (SDSE), Fusobacterium necrophorum, and Arcanobacterium haemolyticum. Compared to culturing, the PCR method resulted in more frequent detection, albeit in most cases with low levels of DNA, of GAS (20/217 vs. 5/217; p < 0.01) and F. necrophorum (20/217 vs. 8/217; p < 0.01). Culturing and PCR yielded similar rates of SDSE detection (14/217 vs. 12/217; p = 0.73). Arcanobacterium haemolyticum was rarely detected (3/217), and only by PCR. Overall, in 25.3% (55/217) of these healthy adolescents and young adults at least one of these pathogens was detected, a rate that is higher than previously described. Further studies are needed before clinical adoption of PCR-based detection methods for pharyngeal bacterial pathogens, as our findings suggest a high incidence of asymptomatic carriage among adolescents and young adults without throat infections.

    • A new extension of the Kolmogorov theory, for the two-point pressure–velocity correlation, is studied by LES of homogeneous turbulence with a large inertial subrange in order to capture the high Reynolds number nonlinear dynamics of the flow. Simulations of both decaying and forced anisotropic homogeneous turbulence were performed. The forcing allows the study of higher Reynolds numbers for the same number of modes compared with simulations of decaying turbulence. The forced simulations give statistically stationary turbulence, with a substantial inertial subrange, well suited to test the Kolmogorov theory for turbulence that is locally isotropic but has significant anisotropy of the total energy distribution. This has been investigated in the recent theoretical studies of Lindborg (1996) and Hill (1997) where the role of the pressure terms was given particular attention. On the surface the two somewhat different approaches taken in these two studies may seem to lead to contradictory conclusions, but are here reconciled and (numerically) shown to yield an interesting extension of the traditional Kolmogorov theory. The results from the simulations indeed show that the two-point pressure–velocity correlation closely adheres to the predicted linear relation in the inertial subrange where also the pressure-related term in the general Kolmogorov equation is shown to vanish. Also, second- and third-order structure functions are shown to exhibit the expected dependences on separation.

  • Electrolyte handling in the isolated perfused rat kidney : demonstration of vasopressin V2-receptor-dependent calcium reabsorption

    • Background The most profound effect of vasopressin on the kidney is to increase water reabsorption through V-2-receptor (V2R) stimulation, but there are also data suggesting effects on calcium transport. To address this issue, we have established an isolated perfused kidney model with accurate pressure control, to directly study the effects of V2R stimulation on kidney function, isolated from systemic effects. Methods The role of V2R in renal calcium handling was studied in isolated rat kidneys using a new pressure control system that uses a calibration curve to compensate for the internal pressure drop up to the tip of the perfusion cannula. Results Kidneys subjected to V2R stimulation using desmopressin (DDAVP) displayed stable osmolality and calcium reabsorption throughout the experiment, whereas kidneys not administered DDAVP exhibited a simultaneous fall in urine osmolality and calcium reabsorption. Epithelial sodium channel (ENaC) inhibition using amiloride resulted in a marked increase in potassium reabsorption along with decreased sodium reabsorption. Conclusions A stable isolated perfused kidney model with computer-controlled pressure regulation was developed, which retained key physiological functions. The preparation responds to pharmacological inhibition of ENaC channels and activation of V2R. Using the model, the dynamic effects of V2R stimulation on calcium handling and urine osmolality could be visualised. The study thereby provides evidence for a stimulatory role of V2R in renal calcium reabsorption.
