    • A method was developed for the characterisation of carotenoid pigments in algal species using Raman spectroscopy in combination with multivariate hyperspectral analysis. Target orthogonal partial least squares (T-OPLS) operates by designating one known reference spectrum as the target. The target spectrum is put as the single y column in an OPLS regression model where the X matrix consists of the unfolded image spectra as variables in its columns. The spectral shape of the OPLS first orthogonal target score enabled us to verify the peak positions of the standard, and detect new peaks, not present in the reference standard. It was shown that the mixture of carotenoids present in the algae did not fully match the reference spectrum, however, the method provided enough information to make an analysis possible also in this case. The image results were constructed from the OPLS loading vectors that were showing a correlation map for the reference spectrum from the predictive loadings and maps of the occurrence of deviations from the orthogonal loadings.

    • Roll compaction is gaining importance in pharmaceutical industry for the dry granulation of heat or moisture sensitive powder blends with poor flowing properties prior to tabletting. We studied the influence of microcrystalline cellulose (MCC) properties on the roll compaction process and the consecutive steps in tablet manufacturing. Four dissimilar MCC grades, selected by subjecting their physical characteristics to principal components analysis, and three speed ratios, i.e. the ratio of the feed screw speed and the roll speed of the roll compactor, were included in a full factorial design. Orthogonal projection to latent structures was then used to model the properties of the resulting roll compacted products (ribbons, granules and tablets) as a function of the physical MCC properties and the speed ratio. This modified version of partial least squares regression separates variation in the design correlated to the considered response from the variation orthogonal to that response. The contributions of the MCC properties and the speed ratio to the predictive and orthogonal components of the models were used to evaluate the effect of the design variation. The models indicated that several MCC properties, e.g. bulk density and compressibility, affected all granule and tablet properties, but only one studied ribbon property: porosity. After roll compaction, Ceolus KG 1000 resulted in tablets with obvious higher tensile strength and lower disintegration time compared to the other MCC grades. This study confirmed that the particle size increase caused by roll compaction is highly responsible for the tensile strength decrease of the tablets.

    • Two new orthogonal projections to latent structures (OPLS) based methods were proposed to analyze hyperspectral images, enabling the visualization ofmultiple chemical compounds in onematrix without the need of extensive preprocessing. Both proposed methods delivered images representing the chemical distribution in the ribbon similar to the more traditional multivariate curve resolution–alternating least squares (MCR-ALS) method, but their image background was less dynamic resulting in a stronger chemical contrast. This indicated that the methods successfully removed structured variation orthogonal to the chemical information (pure spectra of individual compounds), which was confirmed by the fact that physical scattering effects caused by grooves and edges were captured in the images visualizing the orthogonal components of the model. Hereby, the OPLS-based method employing the pure spectra as weights in the OPLS algorithm was more successful in distinguishing compounds with a similar spectral signal than the transposed OPLS algorithm(pure spectra of individual compounds were used as response in OPLS model). It should be noted that for the main compounds, the MCR-ALS method enabled easier visual interpretation compared to the OPLS-based methods by setting all values below zero to zero, resulting in a higher contrast between pixels containing the studied compound and pixels not containing that compound.

    • Surface enhanced Raman spectroscopy (SERS) was used to study the uptake of rhodamine 6G in human lymphocytes. In total four Raman images of lymphocytes were used. The aim was to find a multivariate methodology capable of separating spectra with chemical information from those that mainly contained the surface enhanced background, in order to create chemical images. The standard PCA procedure was compared with PCA of standard normal variate (SNV) corrected spectra, spectra baseline corrected in the wavelet domain, and variable trimming before PCA, to isolate unique spectra. It was not straightforward to perform a standard PCA for overview, since the small background variation in many variables dominated over the Raman band variation that only occur in few variables. It was shown that wavelet filtering could remove background variations and that variable trimming followed by PCA modelling left the unique Raman spectra as outliers, which facilitated interpretation of the Raman score images.

    • This paper is aimed to show the possibility to determine individual organic compounds introduced into single living cells with surface-enhanced Raman spectroscopy (SERS). Surface enhancement was achieved with gold colloids that were allowed to diffuse into lymphocytes. An introduced analyte, rhodamine 6G, could be imaged together with for example nucleotides and amino acids of the cell. Multivariate evaluation of surface-enhanced Raman images proved to be a powerful tool for the separation of spectral information of various intracellular components. The principal component analysis (PCA) enabled identification of spectra containing different chemical information and separation of the spectral contribution of rhodamine 6G from the complex cellular matrix.

    • In this paper, measurements with a THz imaging system at 346 GHz together with electromagnetic simulations have demonstrated the capability of THz sensing for the non-destructive density determination of intermediate products in the pharmaceutical industry. The results confirm the ability of THz imaging to detect density changes in pharmaceutical samples by measuring the phase variation of the wave transmitted through the sample.
