Effects of various squalene epoxides on coenzyme Q and cholesterol synthesis

- 2,3-Oxidosqualene is an intermediate in cholesterol biosynthesis and 2,3:22,23-dioxidosqualene act as the substrate for an alternative pathway that produces 24(S),25-epoxycholesterol which effects cholesterol homeostasis. In light of our previous findings concerning the biological effects of certain epoxidated all-trans-polyisoprenes, the effects of squalene carrying epoxy moieties on the second and third isoprene residues were investigated here. In cultures of HepG2 cells both monoepoxides of squalene and one of their hydrolytic products inhibited cholesterol synthesis and stimulated the synthesis of coenzyme Q (CoQ). Upon prolonged treatment the cholesterol content of these cells and its labeling with [H-3]mevalonate were reduced, while the amount and labeling of CoQ increased. Injection of the squalene monoepoxides into mice once daily for 6 days elevated the level of CoQ in their blood, but did not change the cholesterol level. The same effects were observed upon treatment of apoE-deficient mice and diabetic GK-rats. This treatment increased the hepatic level of CoQ10 in mice, but the amount of CoQ9, which is the major form, was unaffected. The presence of the active compounds in the blood was supported by the finding that cholesterol synthesis in the white blood cells was inhibited. Since the ratio of CoQ9/CoQ10 varies depending on the experimental conditions, the cells were titrated with substrate and inhibitors, leading to the conclusion that the intracellular isopentenyl-PP pool is a regulator of this ratio. Our present findings indicate that oxidosqualenes may be useful for stimulating both the synthesis and level of CoQ both in vitro and in vivo.

Older immigrants in need of care in times of digitalization : The case of filial piety in transition among Chinese families in Sweden

- Globalization has facilitated international communication and economic activities that motivate more international migration. Different cultures and values from the West and the East are interwoven in the same social context. Meanwhile, aging has become a notable phenomenon in most developed countries due to declining fertility and increased life expectancy. A digital transformation of society is taking place alongside the aging and international migration that will start to question the image of older adults and the patterns of providing elderly care. Chinese elderly care is characterized by familism providing family-based informal care to an older family member. Despite the influence of migration and modern lifestyles, filial piety is expected to continue to guide the care responsibilities and care providing among Chinese families. This thesis aims to understand the cultural provision of support and care for Chinese older adults living in Sweden and under the influence of the digital transformation of society. It starts by exploring the disparities, barriers, and facilitators for older immigrants in need of home care to utilize digital technology. After constructing knowledge about the influences of the foreign environment and continuous digitalization in home care, the thesis seeks to understand how filial piety continues to be enacted; what part of the tradition is preserved, and what is changed; how the internal and external factors influence the decision of elderly care; and the role of digitalization in this process. The thesis utilizes qualitative in-depth interviews to continue investigations among eight midlife and older Chinese immigrants living in Sweden. Thematic analysis was employed for data analysis. Berry’s acculturation framework and Yeh’s Dual Filial Piety Model provide the theoretical ground for the research. The results indicated that filial piety is in transition in terms of filial responsibilities, filial behaviors, and intergenerational relationships. Despite the differences in the acculturation level, the authoritarian element of filial piety lacked ground in the Swedish social context. Respondents showed their willingness to strengthen the reciprocity with their adult children by providing additional support, valuing affections rather than hands-on care, respecting adult children’s decisions. The uptake of digital technologies has enabled midlife and older Chinese immigrants to maintain more frequent contact with family members and friends in other countries. It also improves the autonomy and capabilities for respondents to plan more independent later lives. The conclusion is that traditional values such as filial piety was not as prominent in Sweden as they are in China.

Beyond the nation-state Polish national identity and cultural intimacy online

- This article analyses the debate on ;new patriotism' in a Polish online discussion forum. We study the ways in which national identity is constructed in this setting. Digital communication contributes further to expanding discourse on national identities beyond nation-state borders. We analyse close to 6000 posts from a large Polish Internet discussion forum through the methods of quantitative concept mapping and qualitative close readings. Our results show that patriotism is negotiated beyond strictly national frameworks. It is not merely a question of national interest as it also connects people through a process of establishing and maintaining of cultural intimacy.

- Critical Internet and media scholarship has primarily focused on contributing to theoretical debates within the field of media and communications but few empirical studies have applied this theoretical approach. This article uses data on older active ICT users’ understandings of digitisation. It draws inspiration from Boltanski’s pragmatist sociology of critique and the notion that people’s own take on their situation are fruitful sources of information in the quest for emancipation. It employs the notions of ideology, power and inclusion – which are central to critical scholarship – to make sense of older active ICT users’ understandings of digitisation. In doing so, it explores the fruitfulness of the critical lens for studies of ICT users while bringing attention to older active ICT users’ critical capacities.
