  • APOE ε4 is associated with longer telomeres, and longer telomeres among ε4 carriers predicts worse episodic memory

    • Both leukocyte telomere length and the apolipoprotein ε4 allele have been associated with mortality, cardiovascular disease, cognition, and dementia. The authors investigated whether leukocyte telomere length was associated with APOE genotype or cognitive abilities in the context of APOE genotype. The setting for this cross-sectional study was 427 nondemented individuals aged 41–81 yr. The authors found that ε4 carriers overall exhibited significantly longer telomeres compared with non-carriers (difference of 268 bp, p = 0.001). This difference was greatest at the lower limit of the age span and nonsignificant at the upper limit, which translated into a significantly higher telomere attrition rate (p = 0.049) among ε4 carriers (37 bp/years) compared with non-carriers (21 bp/year). Further, longer telomeres among ε4 carriers significantly predicted worse performance on episodic memory tasks. No significant associations were found on tasks tapping semantic and visuospatial ability, or among ε3/ε3 carriers. In conclusion, APOE ε4 carriers had longer telomeres compared with non-carriers, but higher rate of attrition. Among them, longer telomeres predicted worse performance on episodic memory tasks. These observations suggest that the ε4 allele is associated with abnormal cell turnover of functional and possibly clinical significance.

  • Longer Leukocyte Telomere Length Is Associated with Smaller Hippocampal Volume among Non-Demented APOE epsilon 3/epsilon 3 Subjects

    • Telomere length shortens with cellular division, and leukocyte telomere length is used as a marker for systemic telomere length. The hippocampus hosts adult neurogenesis and is an important structure for episodic memory, and carriers of the apolipoprotein E epsilon 4 allele exhibit higher hippocampal atrophy rates and differing telomere dynamics compared with non-carriers. The authors investigated whether leukocyte telomere length was associated with hippocampal volume in 57 cognitively intact subjects (29 epsilon 3/epsilon 3 carriers; 28 epsilon 4 carriers) aged 49-79 yr. Leukocyte telomere length correlated inversely with left (r(s) = -0.465; p = 0.011), right (r(s) = -0.414; p = 0.025), and total hippocampus volume (r(s) = -0.519; p = 0.004) among APOE epsilon 3/epsilon 3 carriers, but not among epsilon 4 carriers. However, the epsilon 4 carriers fit with the general correlation pattern exhibited by the epsilon 3/epsilon 3 carriers, as epsilon 4 carriers on average had longer telomeres and smaller hippocampi compared with epsilon 3/epsilon 3 carriers. The relationship observed can be interpreted as long telomeres representing a history of relatively low cellular proliferation, reflected in smaller hippocampal volumes. The results support the potential of leukocyte telomere length being used as a biomarker for tapping functional and structural processes of the aging brain.

    • The Astrid-2 mission has dual primary objectives. First, it is an orbiting instrument platform for studying auroral electrodynamics. Second, it is a technology demonstration of the feasibility of using micro-satellites for innovative space plasma physics research. The EMMA instrument, which we discuss in the present paper, is designed to provide simultaneous sampling of two electric and three magnetic field components up to about 1 kHz. The spin plane components of the electric field are measured by two pairs of opposing probes extended by wire booms with a separation distance of 6.7 m. The probes have titanium nitride (TiN) surfaces. which has proved to be a material with excellent properties for providing good electrical contact between probe and plasma. The wire booms are of a new design in which the booms in the stowed position are wound around the exterior of the spacecraft body. The boom system was flown for the first time on this mission and worked flawlessly. The magnetic field is measured by a tri-axial fluxgate sensor located at the tip of a rigid. hinged boom extended along the spacecraft spin axis and facing away from the Sun. The new advanced-design fluxgate magnetometer uses digital signal processors for detection and feedback, thereby reducing the analogue circuitry to a minimum. The instrument characteristics as well as a brief review of the science accomplished and planned are presented.

  • Evaluating the performance of TEWA systems

    • It is in military engagements the task of the air defense to protect valuable assets such as air bases from being destroyed by hostile aircrafts and missiles. In order to fulfill this mission, the defenders are equipped with sensors and firing units. To infer whether a target is hostile and threatening or not is far from a trivial task. This is dealt with in a threat evaluation process, in which the targets are ranked based upon their estimated level of threat posed to the defended assets. Once the degree of threat has been estimated, the problem of weapon allocation comes into the picture. Given that a number of threatening targets have been identified, the defenders need to decide on whether any firing units shall be allocated to the targets, and if so, which firing unit to engage which target. To complicate matters, the outcomes of such engagements are usually stochastic. Moreover, there are often tight time constraints on how fast the threat evaluation and weapon allocation processes need to be executed. There are already today a large number of threat evaluation and weapon allocation (TEWA) systems in use, i.e. decision support systems aiding military decision makers with the threat evaluation and weapon allocation processes. However, despite the critical role of such systems, it is not clear how to evaluate the performance of the systems and their algorithms. Hence, the work in thesis is focused on the development and evaluation of TEWA systems, and the algorithms for threat evaluation and weapon allocation being part of such systems. A number of algorithms for threat evaluation and static weapon allocation are suggested and implemented, and testbeds for facilitating the evaluation of these are developed. Experimental results show that the use of particle swarm optimization is suitable for real-time target-based weapon allocation in situations involving up to approximately ten targets and ten firing units, while it for larger problem sizes gives better results to make use of an enhanced greedy maximum marginal return algorithm, or a genetic algorithm seeded with the solution returned by the greedy algorithm.

  • Denaturant-assisted formation of a stabilizing disulfide bridge from engineered cysteines in nonideal conformations

    • The engineered disulfide bridge A23C/L203C in human carbonic anhydrase II, inserted from homology modeling of Neisseria gonorrhoeae carbonic anhydrase, significantly stabilizes the native state of the protein. The inserted cysteine residues are placed in the interior of the structure, and because of the conformationally restrained localization, the protein is expressed in the reduced state and the cysteines are not readily oxidized. However, upon exposure to low concentrations of denaturant (0.6 M guanidine hydrochloride), corresponding to the lower part of the denaturation curve for the first unfolding transition, the oxidation rate of correctly formed disulfide bridges was markedly increased. By entropy estimations it appears that the increased flexibility, induced by the denaturant, enables the cysteines to find each other and hence to form the disulfide bridge. The outlined strategy of facilitating formation of disulfide bonds by addition of adjusted concentrations of a denaturant should be applicable to other proteins in which engineered cysteine residues are located in nonideal conformations. Moreover, a S99C/V242C variant was constructed, in which the cysteine residues are located on the surface. In this mutant the disulfide bridge was spontaneously formed and the native state was considerably stabilized (midpoint concentration of unfolding was increased from 1.0 to 1.4 M guanidine hydrochloride).
