    • Diabetes mellitus is a chronic disease, which has an increasing trend all over the world. Type 2 diabetes constitutes 90% of all diabetes. It is associated with weight gain and insulin resistance. Research during recent years has suggested that shift work could be a risk factor of type 2 diabetes. Since shift work is becoming more common, it could contribute to the increasing trend of diabetes. In this systematic review, we have studied the potential association between shift work and type 2 diabetes. We have also reviewed studies on control of diabetes in relation to shift work.

  • A genomic snapshot of demographic and cultural dynamism in Upper Mesopotamia during the Neolithic Transition

    • Upper Mesopotamia played a key role in the Neolithic Transition in Southwest Asia through marked innovations in symbolism, technology, and diet. We present 13 ancient genomes (c. 8500 to 7500 cal BCE) from Pre-Pottery Neolithic Çayönü in the Tigris basin together with bioarchaeological and material culture data. Our findings reveal that Çayönü was a genetically diverse population, carrying mixed ancestry from western and eastern Fertile Crescent, and that the community received immigrants. Our results further suggest that the community was organized along biological family lines. We document bodily interventions such as head shaping and cauterization among the individuals examined, reflecting Çayönü’s cultural ingenuity. Last, we identify Upper Mesopotamia as the likely source of eastern gene flow into Neolithic Anatolia, in line with material culture evidence. We hypothesize that Upper Mesopotamia’s cultural dynamism during the Neolithic Transition was the product not only of its fertile lands but also of its interregional demographic connections.

    • This presentation briefly covers the ongoing development of a therapy center with multiple simultaneous radiation modalities at Karolinska university hospital. The hearth of the facility will most likely be a superconducting cyclotron capable of delivering around 400 MeV/u carbon ions simultaneously to two separate excentric gantries who service four treatment rooms each. A number of different stable ions will be available ranging from hydrogen to oxygen but also PET emitting C11 ions and possibly B8 the lightest existing PET emitter. Several treatment rooms will also be equipped for narrow scanned high energy photon and electron beam treatments and a light ion research facility for physics and biology studies will be set up on two separate beamlines. The centre will include an advanced PET-CT and MRSI based diagnostic centre on the same floor and close to both the ion treatment facility and the high energy photon and electron facility. By docking the stereotactic treatment coach both to the treatment units and before and after the treatment to the diagnostic PET-CT units, iso dose delivery can be rapidly examined by imaging the radiation induced C11 and O15 activity produced during the treatment.

    • The integrated dual-polarisation receivers utilize a dual probe concept, efficiently integrating the antenna and MMIC package environment which allows for polarisation discrimination without the use of bulky and lossy external orthomode transducers. This concept increases the sensitivity of the instrument and reduces its size, enabling the development of future earth observation arrays. Omnisys Instruments AB (Sweden) and Chalmers University of Technology (Sweden) are working to demonstrate state-of-the-art dual polarisation capability with two integrated receiver modules at 325 GHz and 650 GHz.

  • Development of antimicrobial peptides : Methodological aspects, design, synthesis and evaluation of a new class of antimicrobial peptides

    • A new protective group for the indole nitrogen of the amino acid tryptophan, the Boc-sarcosinoyl-sarcosinolyl (Boc-Sar-Sar) is described. This protective group shows good stability to the reaction conditions used in solid phase peptide synthesis and can be incorporated into the peptide as Fmoc-Trp(Boc-Sar-Sar)-OH. When the peptide is cleaved from the resin with trifluororoactetic acid, the Boc group is simultaneously cleaved and the N-terminal nitrogen in the Sar-Sar group is exposed. At low pH this amino group is protonated and thereby the peptide will have higher solubility in aqueous solution. High solubility of the peptide is an important factor for the purification of peptides with reversed phase HPLC. During HPLC purification, the protonated Sar-Sar group will remain on the peptide. When the purified peptide is exposed to physiological pH, the Sar-Sar group will be cleaved and the peptide will be obtained in its fully deprotected form.Gramicidin A (GA) is an extremely hydrophobic peptide with four tryptophan residues. To improve the solubility of GA, Fmoc-Trp(Boc-Sar-Sar)-OH was used in the solid phase synthesis of GA resulting in (H-Sar-Sar)4-GA. When this peptide was exposed to physiological pH native GA was formed.A new class of antimicrobial peptides, represented by the enantiomers; cyclo[D-Tle-D-Lys-D-Tle-L-Ala-D-Tle-L-Ala-D-Tle-L-Ala] and cyclo[L-Tle-L-Lys-L-Tle-D-Ala-L-Tle-D-Ala-L-Tle-D-Ala] is described. Neither of the two peptides shows any significant antimicrobial activity against Bacillus megaterium. However, when both peptides are present during the experiment a potent antimicrobial activity with an IC50 value of 2 µM could be observed. The peptides also show low hemolytic activity with an HC50 value of 316 µM. The synergistic mode of action of these peptides is dependent on the presence of both enantiomers. Circumstantial evidences indicate that the antimicrobial effect is the result of formation of peptide nanotubes within the bacterial membrane.It is also shown that the cyclic peptides described above can be modified at the lysine residue with three arginine residues in which the N-terminus is acylated with a short fatty acid. Togheter these peptides show a synergistic mode of action against Staphylococcus aureus resulting in a MIC value of 9 µM.

    • Background: After over a decade of developments in field collection, laboratory methods and advances in high-throughput sequencing, contamination remains a key issue in ancient DNA research. Currently, human and microbial contaminant DNA still impose challenges on cost-effective sequencing and accurate interpretation of ancient DNA data.Results: Here we investigate whether human contaminating DNA can be found in ancient faunal sequencing datasets. We identify variable levels of human contamination, which persists even after the sequence reads have been mapped to the faunal reference genomes. This contamination has the potential to affect a range of downstream analyses.Conclusions: We propose a fast and simple method, based on competitive mapping, which allows identifying and removing human contamination from ancient faunal DNA datasets with limited losses of true ancient data. This method could represent an important tool for the ancient DNA field.

    • Syftet med denna rapport är att illustrera hur ett praktiskt inriktat ramverk, tekno- logiska innovationssystem (TIS), kan användas av analytiker och beslutsfattare vid departement och myndigheter för att analysera strategiskt viktiga teknikområden ?????????????????????????????????????????????????????????????????????????????I rapporten analyseras fem TIS centrerade kring havsbaserad vindkraft, marin energi, ????????????????????????????????????????????????????????????????????????????????????? systemsvagheter som bromsar områdets vidare utveckling, vilka som kan åtgärdas av systemets aktörer och vilka som motiverar särskilda politiska åtaganden. Rapporten utgör därmed ett underlag för att formulera åtgärder för att åstadkomma ökad innova- tion, teknikspridning och industrialisering inom ovan nämnda teknikområden.Studien har även möjliggjort en jämförande analys av likheter och skillnader ???????????????????????????????????????????????????????????????????????????????? ?????????????????????????????????????????????????????????????????????????????????????? mellan områdena – de är starka respektive svaga av olika orsaker. Detta visar att ???????????????????????????????????????????????????????????????????????Samtidigt har områdena gemensamma drag. De behövs för att investeringar i kunskapsutveckling ska kunna nyttiggöras och för att en bred industriell utveckling inom nya områden skall göras möjlig i Sverige.Vidare presenteras lärdomar kring vad en aktiv teknikpolitik innebär. Två huvud- ??????????????????????????????????????????????????????????????????????????????- hällsbygget och därför bör vara ett politikområde bland många samt att den skarpa ??????????????????????????????????????????????????????????????????????????????- ?????????????????????????????????????????????????????????????????????????????????? ?????????????????????????????????????????????????????????????????????????????????? olika faser av innovationssystemets utveckling.För att lyckas med en aktiv teknikpolitik behövs en hög grad av koordinering ??????????????????????????????????????????????????????????????????????? teknikområden så att ”rätt” typ av åtgärder kan sättas in vid ”rätt” tidpunkt av ”rätt” aktör. TIS-ramverket lyfts här fram som en metod för att skapa ett sådant underlag.   • Advances in Separation Science : . Molecular Imprinting: Development of Spherical Beads and Optimization of the Formulation by Chemometrics.

    • An intrinsic mathematical model for simulation of fixed bed chromatography was demonstrated and compared to more simplified models. The former model was shown to describe variations in the physical, kinetic, and operating parameters better than the latter ones. This resulted in a more reliable prediction of the chromatography process as well as a better understanding of the underlying mechanisms responsible for the separation. A procedure based on frontal liquid chromatography and a detailed mathematical model was developed to determine effective diffusion coefficients of proteins in chromatographic gels. The procedure was applied to lysozyme, bovine serum albumin, and immunoglobulin γ in Sepharose™ CL-4B. The effective diffusion coefficients were comparable to those determined by other methods.Molecularly imprinted polymers (MIPs) are traditionally prepared as irregular particles by grinding monoliths. In this thesis, a suspension polymerization providing spherical MIP beads is presented. Droplets of pre-polymerization solution were formed in mineral oil with no need of stabilizers by vigorous stirring. The droplets were transformed into solid spherical beads by free-radical polymerization. The method is fast and the performance of the beads comparable to that of irregular particles. Optimizing a MIP formulation requires a large number of experiments since the possible combinations of the components are huge. To facilitate the optimization, chemometrics was applied. The amounts of monomer, cross-linker, and porogen were chosen as the factors in the model. Multivariate data analysis indicated the influence of the factors on the binding and an optimized MIP composition was identified. The combined use of the suspension polymerization method to produce spherical beads with the application of chemometrics was shown in this thesis to drastically reduce the number of experiments and the time needed to design and optimize a new MIP.
