  • Guidelines for the use and interpretation of assays for monitoring autophagy
  • 2012
  • Ingår i: Autophagy. - : Informa UK Limited. - 1554-8635 .- 1554-8627. ; 8:4, s. 445-544
  • Forskningsöversikt (refereegranskat)abstract
    • In 2008 we published the first set of guidelines for standardizing research in autophagy. Since then, research on this topic has continued to accelerate, and many new scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Accordingly, it is important to update these guidelines for monitoring autophagy in different organisms. Various reviews have described the range of assays that have been used for this purpose. Nevertheless, there continues to be confusion regarding acceptable methods to measure autophagy, especially in multicellular eukaryotes. A key point that needs to be emphasized is that there is a difference between measurements that monitor the numbers or volume of autophagic elements (e.g., autophagosomes or autolysosomes) at any stage of the autophagic process vs. those that measure flux through the autophagy pathway (i.e., the complete process); thus, a block in macroautophagy that results in autophagosome accumulation needs to be differentiated from stimuli that result in increased autophagic activity, defined as increased autophagy induction coupled with increased delivery to, and degradation within, lysosomes (in most higher eukaryotes and some protists such as Dictyostelium) or the vacuole (in plants and fungi). In other words, it is especially important that investigators new to the field understand that the appearance of more autophagosomes does not necessarily equate with more autophagy. In fact, in many cases, autophagosomes accumulate because of a block in trafficking to lysosomes without a concomitant change in autophagosome biogenesis, whereas an increase in autolysosomes may reflect a reduction in degradative activity. Here, we present a set of guidelines for the selection and interpretation of methods for use by investigators who aim to examine macroautophagy and related processes, as well as for reviewers who need to provide realistic and reasonable critiques of papers that are focused on these processes. These guidelines are not meant to be a formulaic set of rules, because the appropriate assays depend in part on the question being asked and the system being used. In addition, we emphasize that no individual assay is guaranteed to be the most appropriate one in every situation, and we strongly recommend the use of multiple assays to monitor autophagy. In these guidelines, we consider these various methods of assessing autophagy and what information can, or cannot, be obtained from them. Finally, by discussing the merits and limits of particular autophagy assays, we hope to encourage technical innovation in the field.

  • Development, validation, and comparison of a nomogram based on radiologic findings for predicting malignancy in intraductal papillary mucinous neoplasms of the pancreas : An international multicenter study
  • 2021
  • Ingår i: Journal of hepato-biliary-pancreatic sciences. - : Wiley-Blackwell. - 1868-6974 .- 1868-6982.
  • Tidskriftsartikel (refereegranskat)abstract
    • BACKGROUND: Although we previously proposed a nomogram to predict malignancy in intraductal papillary mucinous neoplasms (IPMN) and validated it in an external cohort, its application is challenging without data on tumor markers. Moreover, existing nomograms have not been compared. This study aimed to develop a nomogram based on radiologic findings and to compare its performance with previously proposed American and Korean/Japanese nomograms.METHODS: We recruited 3708 patients who underwent surgical resection at 31 tertiary institutions in eight countries, and patients with main pancreatic duct >10 mm were excluded. To construct the nomogram, 2606 patients were randomly allocated 1:1 into training and internal validation sets, and area under the receiver operating characteristics curve (AUC) was calculated using 10-fold cross validation by exhaustive search. This nomogram was then validated and compared to the American and Korean/Japanese nomograms using 1102 patients.RESULTS: Among the 2606 patients, 90 had main-duct type, 900 had branch-duct type, and 1616 had mixed-type IPMN. Pathologic results revealed 1628 low-grade dysplasia, 476 high-grade dysplasia, and 502 invasive carcinoma. Location, cyst size, duct dilatation, and mural nodule were selected to construct the nomogram. AUC of this nomogram was higher than the American nomogram (0.691 vs 0.664, P = .014) and comparable with the Korean/Japanese nomogram (0.659 vs 0.653, P = .255).CONCLUSIONS: A novel nomogram based on radiologic findings of IPMN is competitive for predicting risk of malignancy. This nomogram would be clinically helpful in circumstances where tumor markers are not available. The nomogram is freely available at http://statgen.snu.ac.kr/software/nomogramIPMN.

  • Competence development at workplace : a work integrated informal learning (WIIL) approach
  • 2016
  • Ingår i: Far East Journal of Electronics and Communications. - Allahabad, India : Pushpa Publishing House. - 0973-7006. ; 2, s. 35-42
  • Tidskriftsartikel (refereegranskat)abstract
    • Learning within the organization is not only linked to the formal training and education of the employees, but also to informal processes of sharing knowledge. The knowledge is often embedded in organizational activities, practices and norms, as well as in social ties among individuals in the organization. We analyzed employees' behavior and acceptance of online social networking as an information system (IS) of learning. The analysis is performed by a descriptive case survey within a telecom corporation in Pakistan. The purpose of our study is to bring an informal learning system at workplace. In this study we obtain quantitative data from 105 out of 180 targeted employees. The results show that a majority of the studied employees favor social networking to connect colleagues at different levels and more than half of the respondents share their knowledge and experiences using social sites. Nearly half of the respondents strongly agree that knowledge sharing is an important role of social sites within the organization and many are in favor of social media as an informal learning system. The results also show how the professionals use social networking sites to share their experiences and to build a virtual break in hierarchical levels of professions. This paper illustrates the benefits of social networking sites as an informal work integrated learning tool that also serves as an IS model for the organization.

  • Social Networking sites for Interdisciplinary Learning at Workplace : Work integrated informal Learning (WiiL)
  • 2016
  • Ingår i: International Conference on Advanced Computing, Communication and Information Sciences (ICACCI-2016). - : Pushpa Publishing House. ; , s. 9-9
  • Konferensbidrag (refereegranskat)abstract
    • Work Integrated Learning (WIL) studies usually focuses on educational situations when professional organizations are actively involved in learning processes for students in higher education. However, after completing the institutional period entering the professional arena, the professional person is still actually a student at the workplace as s/he never stops learning. Learning within the organization is not only linked to the formal training and education of the employees, but also to informal processes of sharing knowledge. The knowledge is often embedded in organizational activities, practices and norms, as well as in social ties among individuals in the organization. • Multi-step enzymatic synthesis of 1,9-nonanedioic acid from a renewable fatty acid and its application for the enzymatic production of biopolyesters
  • 2019
  • Ingår i: Polymers. - : MDPI AG. - 2073-4360. ; 11:10
  • Tidskriftsartikel (refereegranskat)abstract
    • 1,9-Nonanedioic acid is one of the valuable building blocks for producing polyesters and polyamides. Thereby, whole-cell biosynthesis of 1,9-nonanedioic acid from oleic acid has been investigated. A recombinant Corynebacterium glutamicum, expressing the alcohol/aldehyde dehydrogenases (ChnDE) of Acinetobacter sp. NCIMB 9871, was constructed and used for the production of 1,9-nonanedioic acid from 9-hydroxynonanoic acid, which had been produced from oleic acid. When 9-hydroxynonanoic acid was added to a concentration of 20 mM in the reaction medium, 1,9-nonanedioic acid was produced to 16 mM within 8 h by the recombinant C. glutamicum. The dicarboxylic acid was isolated via crystallization and then used for the production of biopolyester by a lipase. For instance, the polyesterification of 1,9-nonanedioic acid and 1,8-octanediol in diphenyl ether by the immobilized lipase B from Candida antarctica led to formation of the polymer product with the number-average molecular weight (Mn) of approximately 21,000. Thereby, this study will contribute to biological synthesis of long chain dicarboxylic acids and their application for the enzymatic production of long chain biopolyesters.

  • A simple pharmacokinetic model of alendronate developed using plasma concentration and urine excretion data from healthy men
  • 2014
  • Ingår i: Drug Development and Industrial Pharmacy. - : Informa UK Limited. - 0363-9045 .- 1520-5762. ; 40:10, s. 1325-1329
  • Tidskriftsartikel (refereegranskat)abstract
    • The study of pharmacokinetics of alendronate has been hampered by difficulties in accurately and reproducibly determining their concentrations in serum and urine. Thus, pharmacokinetic characteristics of alendronate have been described in many reports based on urinary excretion data; and plasma pharmacokinetics and the simultaneous pharmacokinetic models of alendronate in plasma and urine are not available. The aims of this study were to measure alendronate concentration in plasma and excretion in urine concurrently and to develop compartmental pharmacokinetic model using urine data. In open-label, single-dose pharmacokinetic study, 10 healthy male volunteers received oral dose of alendronate (70mg tablet). Blood and urine alendronate concentrations were determined using validated high-performance liquid chromatography method. Non-compartmental analysis was performed using WinNonlin program (Pharsight Inc., Apex, NC). A one-compartment pharmacokinetic model was applied to describe pharmacokinetics of alendronate. A peak plasma alendronate concentration of 33.10 +/- 14.32 ng/mL was attained after 1.00 +/- 0.16 h. The cumulative amount of alendronate excreted in urine and peak excretion rate were 731.28 +/- 654.57 mu g and 314.68 +/- 395.43 mg/h, respectively. The model, which included first-order absorption rate for oral dosing, showed good fit to alendronate data obtained from plasma and urine. The absorption rate constant was 2.68 +/- 0.95 h(-1). The elimination rate constants K-urine and Knon-ur were 0.005 +/- 0.004 h(-1) and 0.42 +/- 0.08 h(-1), respectively. The pharmacokinetics of alendronate in plasma and urine of healthy men can be predicted using one-compartment model, and thus the behavior of drug in plasma can be estimated from urinary excretion data.
