    • Progression through mitosis is balanced by the timely regulation of phosphorylation and dephosphorylation events ensuring the correct segregation of chromosomes before cytokinesis. This balance is regulated by the opposing actions of CDK1 and PP2A, as well as the Greatwall kinase/MASTL. MASTL is commonly overexpressed in cancer, which makes it a potential therapeutic anticancer target. Loss of Mastl induces multiple chromosomal errors that lead to the accumulation of micronuclei and multilobulated cells in mitosis. Our analyses revealed that loss of Mastl leads to chromosome breaks and abnormalities impairing correct segregation. Phospho-proteomic data for Mastl knockout cells revealed alterations in proteins implicated in multiple processes during mitosis including double-strand DNA damage repair. In silico prediction of the kinases with affected activity unveiled NEK2 to be regulated in the absence of Mastl. We uncovered that, RAD51AP1, involved in regulation of homologous recombination, is phosphorylated by NEK2 and CDK1 but also efficiently dephosphorylated by PP2A/B55. Our results suggest that MastlKO disturbs the equilibrium of the mitotic phosphoproteome that leads to the disruption of DNA damage repair and triggers an accumulation of chromosome breaks even in noncancerous cells.

    • Metabolic engineering of heterologous biosynthetic pathways in microbial hosts is an alternative way for producing high-value biochemicals. Here, heterologous mevalonate (MVA) and prenyl pyrophosphate (IPP) pathways of a new isolate Enterococcus kingsejongensis Gf1 were redesigned and reconstructed in Escherichia coli. Two MVA upstream enzymes of E. kingsejongensis Gf1 were functionally expressed and overexpression of the two enzymes led to a yield of 52.5 g/L MVA after 65 h of fed-batch fermentation. Next, the functions of three MVA downstream enzymes were verified by addition of exogenous MVA. Finally, whole MVA pathway enzymes and IPP pathway enzymes were combined in E. coli in order to overproduce squalene and β-carotene. The E. coli strains coexpressing entire MVA and IPP pathway enzymes enhanced squalene production by 15-fold and β-carotene production by 2.5-fold.

    • The tailoring of the chromaticity of white organic light-emitting diodes (WOLEDs) has presented a significant challenge in their application in smart lighting sources to improve the quality of life and human performance. Here, a new microcavity WOLED (M-WOLED) structure to modulate the chromaticity of the emitted light is demonstrated by only adjusting the thickness of the white light-emitting layer. By introducing a polymer-metal hybrid electrode that functions both as a partially reflective mirror and a transparent electrode, a very simple microcavity architecture that does not require additional outer mirrors, such as distributed Bragg reflectors is developed. The resulting M-WOLEDs exhibit reddish-, greenish-, and bluish-white colors with different thicknesses of the single white light-emitting layer.

    • Correct identification of the origins of herbal medical products is becoming increasingly important in tandem with the growing interest in alternative medicine. However, visual inspection of raw material is still the most widely used method, and newer scientific approaches are needed. To develop a more objective and efficient tool for discriminating herbal origins, particularly Korean and Chinese, we employed a nuclear magnetic resonance (NMR)-based metabolomics approach combined with an orthogonal projections to latent structure-discriminant analysis (OPLS-DA) multivariate analysis. We first analyzed the constituent metabolites of Scutellaria baicalensis through NMR studies. Subsequent holistic data analysis with OPLS-DA yielded a statistical model that could cleanly discriminate between the sample groups even in the presence of large structured noise. An analysis of the statistical total correlation spectroscopy (STOCSY) spectrum identified citric acid and arginine as the key discriminating metabolites for Korean and Chinese samples. As a validation of the discrimination model, we performed blind prediction tests of sample origins using an external test set. Our model correctly predicted the origins of all of the 11 test samples, demonstrating its robustness. We tested the wider applicability of the developed method with three additional herbal medicines from Korea and China and obtained very high prediction accuracy. The solid discriminatory power and statistical validity of our method suggest its general applicability for determining the origins of herbal medicines.

    • There are contradictory reports in the literature regarding the anti-bacterial activity of graphene, graphene oxide (GO) and reduced graphene oxide (rGO). This controversy is mostly due to variations in key parameters of the reported experiments, like: type of substrate, form of graphene, number of layers, type of solvent and most importantly, type of bacteria. Here, we present experimental data related to bacterial response to GO and rGO integrated in solid agar-based nutrient plates-a standard set-up for bacterial growth that is widely used by microbiologists. Bacillus subtilis and Pseudomonas aeruginosa strains were used for testing bacterial growth. We observed that plate-integrated rGO showed strong anti-bacterial activity against both bacterial species. By contrast, plate-integrated GO was harmless to both bacteria. These results reinforce the notion that the response of bacteria depends critically on the type of graphene material used and can vary dramatically from one bacterial strain to another, depending on bacterial physiology.

    • We demonstrate a ligand-free green chemical method for the rapid growth of nanoporous Ca0.35CoO2 thin films on sapphire and mica substrates from a water-based precursor ink, formulated by dissolving the precursor solid, composed of in situ prepared Ca2+-DMF and Co2+-DMF complexes. Mica serves as the flexible substrate as well as the sacrificial layer for the film transfer. Despite the presence of nanopores, the power factor of the flexible film Ca0.35CoO2-on-mica is above 0.50 x 10(-4) W m(-1) K-2 at around room temperature. The present technique is simple and cost-effective.
