  • Managing Traceability Information Models: Not Such a Simple Task After All?

    • Practitioners are poorly supported by the scientific literature when managing traceability information models (TIMs), which capture the structure and semantics of trace links. In practice, companies manage their TIMs in very different ways, even in cases where companies share many similarities. We present our findings from an in-depth focus group about TIM management with three different systems engineering companies. We find that the concrete needs of the companies as well as challenges such as scale and workflow integration are not considered by existing scientific work. We thus issue a call-to-arms for the requirements engineering and software and systems traceability communities, the two main communities for traceability research, to refocus their work on these practical problems.   • Potentiating and depressive effects of ouabain and potassium-free solutions on rat mesenteric resistance vessels.

    • We have investigated the in vitro effects of ouabain and K-free solutions on some pharmacological and electrophysiological properties of rat mesenteric resistance vessels (internal diameter approximately 190 micrometers). Vessels were mounted as ring preparations on a myograph capable of measuring their isometric wall tension. In normal saline solutions, vessels did not exhibit any tone and had a membrane potential of -54 mV. Both 1 mM ouabain and K-free solutions caused a transient depolarization of 5-8 mV; thereafter the membrane slowly depolarized to about -45 mV after 30 minutes. There was no mechanical response to ouabain, but K-free solutions caused a transient development of tension which could be inhibited by phentolamine (1 microM). In norepinephrine-activated vessels, exposure to ouabain or K-free solutions caused a small depolarization and an increase in tension. Long-term (30-minute) exposure to 1 mM ouabain or K-free solutions reduced the amplitude of norepinephrine responses and, for the lower (but not the higher) norepinephrine concentrations, the membranes were about 14 mV more depolarized than control. The mechanical responses to a cocktail of norepinephrine in a high potassium solution were, however, unaffected. Re-exposure to normal saline solution produced a transient hyperpolarization and transiently eliminated the norepinephrine response, but thereafter the membrane potential and response returned to normal. The results indicate that ouabain and K-free solutions can have both short-term potentiating and long-term depressive effects on the mechanical response of rat mesenteric resistance vessels to norepinephrine.

    • The pneumolysin (ply) gene is widely used as a target in PCR assays for Streptococcus pneumoniae in respiratory secretions. However, false-positive results with conventional ply-based PCR have been reported. The aim here was to study the performance of a quantitative ply-based PCR for the identification of pneumococcal lower respiratory tract infection (LRTI). In a prospective study, fibreoptic bronchoscopy was performed in 156 hospitalized adult patients with LRTI and 31 controls who underwent bronchoscopy because of suspicion of malignancy. Among the LRTI patients and controls, the quantitative ply-based PCR applied to bronchoalveolar lavage (BAL) fluid was positive at >/=10(3) genome copies/mL in 61% and 71% of the subjects, at >/=10(5) genome copies/mL in 40% and 58% of the subjects, and at >/=10(7) genome copies/mL in 15% and 3.2% of the subjects, respectively. Using BAL fluid culture, blood culture, and/or a urinary antigen test, S. pneumoniae was identified in 19 LRTI patients. As compared with these diagnostic methods used in combination, quantitative ply-based PCR showed sensitivities and specificities of 89% and 43% at a cut-off of 10(3) genome copies/mL, of 84% and 66% at a cut-off of 10(5) genome copies/mL, and of 53% and 90% at a cut-off of 10(7) genome copies/mL, respectively. In conclusion, a high cut-off with the quantitative ply-based PCR was required to reach acceptable specificity. However, as a high cut-off resulted in low sensitivity, quantitative ply-based PCR does not appear to be clinically useful. Quantitative PCR methods for S. pneumoniae using alternative gene targets should be evaluated.

  • Spatial distribution of erosion and deposition during a glacier surge: Bruarjokull, Iceland

    • Time-series of digital elevation models (DEMs) of the forefield of the Bruarjokull surge-type glacier in Iceland were used to quantify the volume of material that was mobilized by the 1963-1964 surge. The DEMs were produced by stereophotogrammetry on aerial photographs from before the surge (1961) and after (1988 and 2003). The analysis was performed on two DEMs of Difference (DoDs), i.e., a 1961-2003 DoD documenting the impact of the surge and a 1988-2003 DoD documenting the post-surge modification of the juvenile surging glacier landsystem. Combined with a digital geomorphological map, the DoDs allow us to quantify the impact of the surge on a landsystem scale down to individual landforms. A total of 34.2 +/- 11.3 x 10(6) m(3) of material was mobilized in the 30.7-km(2) study area as a result of the most recent surge event Of these, 17.4 +/- 6.6 x 10(6) m(3) of the material were eroded and 16.8 +/- 4.7 x 10(6) m(3) were deposited. More than half of the deposited volume was ice-cored landforms. This study demonstrates that although the total mobilized mass volume is high, the net volume gain of ice and sediment deposited as landforms in the forefield caused by the surge is low. Furthermore, deposition of new dead-ice from the 1963-1964 surge constitutes as much as 64% of the volume gain in the forefield. The 1988-2003 DoD is used to quantify the melt-out of this dead-ice and other paraglacial modification of the recently deglaciated forefield of Bruarjokull. (C) 2015 Elsevier B.V.     • OBJECTIVE: The aim of this study was to give a collective overview on all available data sources on bladder cancer patients in the Nordic countries including the amount of detail and coverage.METHODS: National representatives from five Nordic countries were asked to fill out a questionnaire on available information regarding bladder cancer patients from databases in their respective countries. Additional information was retrieved from descriptions of the relevant registries.RESULTS: : Information on overall survival was available in all countries whereas recurrence-free survival and cancer-specific survival were available for some but not all patients depending on treatment modality.CONCLUSIONS: Despite limitations, we found that it was possible to retrieve detailed information on diagnostics, treatment, and outcome for most aspects of bladder cancer in the Nordic countries on a population based, non-selected patient cohort.
