    • As the lipidomics field continues to advance, self-evaluation within the community is critical. Here, we performed an interlaboratory comparison exercise for lipidomics using Standard Reference Material (SRM) 1950-Metabolites in Frozen Human Plasma, a commercially available reference material. The interlaboratory study comprised 31 diverse laboratories, with each laboratory using a different lipidomics workflow. A total of 1,527 unique lipids were measured across all laboratories and consensus location estimates and associated uncertainties were determined for 339 of these lipids measured at the sum composition level by five or more participating laboratories. These evaluated lipids detected in SRM 1950 serve as community-wide benchmarks for intra-and interlaboratory quality control and method validation. These analyses were performed using nonstandardized laboratory-independent workflows. The consensus locations were also compared with a previous examination of SRM 1950 by the LIPID MAPS consortium.jlr While the central theme of the interlaboratory study was to provide values to help harmonize lipids, lipid mediators, and precursor measurements across the community, it was also initiated to stimulate a discussion regarding areas in need of improvement.

  • C1QA and COMP: plasma-based biomarkers for early diagnosis of pancreatic neuroendocrine tumors

    • Pancreatic Neuroendocrine tumors (PanNET) are challenging to diagnose and often detected at advanced stages due to a lack of specific and sensitive biomarkers. This study utilized proteomics as a valuable approach for cancer biomarker discovery; therefore, mass spectrometry-based proteomic profiling was conducted on plasma samples from 12 subjects (3 controls; 5 Grade I, 4 Grade II PanNET patients) to identify potential proteins capable of effectively distinguishing PanNET from healthy controls. Data are available via ProteomeXchange with the identifier PXD045045. 13.2% of proteins were uniquely identified in PanNET, while 60% were commonly expressed in PanNET and controls. 17 proteins exhibiting significant differential expression between PanNET and controls were identified with downstream analysis. Further, 5 proteins (C1QA, COMP, HSP90B1, ITGA2B, and FN1) were selected by pathway analysis and were validated using Western blot analysis. Significant downregulation of C1QA (p = 0.001: within groups, 0.03: control vs. grade I, 0.0013: grade I vs. grade II) and COMP (p = 0.011: within groups, 0.019: control vs grade I) were observed in PanNET Grade I & II than in controls. Subsequently, ELISA on 38 samples revealed significant downregulation of C1QA and COMP with increasing disease severity. This study shows the potential of C1QA and COMP in the early detection of PanNET, highlighting their role in the search for early-stage (Grade-I and Grade-II) diagnostic markers and therapeutic targets for PanNET.

  • Artificial synaptic characteristics of PVA:ZnO nanocomposite memristive devices

    • Computational efficiency is significantly enhanced using artificial neural network-based computing. A two-terminal memristive device is a powerful electronic device that can mimic the behavior of a biological synapse in addition to storing information and performing logic operations. This work focuses on the fabrication of a memristive device that utilizes a resistive switching layer composed of polyvinyl alcohol infused with ZnO nanoparticles. By incorporating ZnO nanoparticles into the polymer film, the fabricated memristive devices exhibit functionalities that closely resemble those of biological synapses, including short-term and long-term plasticity, paired-pulse facilitation, and spike time-dependent plasticity. These findings establish the ZnO nanoparticle-polymer nanocomposite as a highly promising material for future neuromorphic systems. © 2023 Author(s).

  • Hierarchically Assembled Two-dimensional Hybrid Nanointerfaces: A Platform for Bioelectronic Applications

    • We introduce a hybrid two-dimensional nanointerface structure for bioelectronic systems. We fabricated smart hierarchically self-assembled 2D electrobiocatalytic interface system based on the combination of gold nanoparticles doped graphene oxide (GO)-molybdenum disulfide (MoS2) layered nanohybrid, conjugated with poly (N-isopropylacrylamide, PNIPAAm) resulting in GO/AuNPs/MoS2/PNIPAAm interface. Horseradish peroxidase (HRP) was subsequently immobilized on the GO/AuNPs/MoS2/PNIPAAm interface through electrostatic interactions giving GO/AuNPs/MoS2/PNIPAAm/Peroxidase electrobiocatalytic interface system as a platform for electrobiocatalysis reactions for biosensing and bioelectronic applications. The electrobiocatalytic activity of the nanohybrid interface structure was studied using hydrogen peroxide (H2O2) as a model analyte. Cyclic Voltammetry showed diffusion controlled electron transfer properties at the interface. The fabricated bioelectrode exhibits a wide linear response to the detection of H2O2 from 1.57 to 11.33mM, with a detection limit of 3.34mM (S/N=3) and a capacitance of 8.6F/cm(2).
