  • Characterization of cell-free breast cancer patient-derived scaffolds using liquid chromatography-mass spectrometry/mass spectrometry data and RNA sequencing data

    • Patient-derived scaffolds (PDSs) generated from primary breast cancer tumors can be used to model the tumor microenvironment in vitro . Patient-derived scaffolds are generated by repeated detergent washing, removing all cells. Here, we analyzed the protein composition of 15 decellularized PDSs using liquid chromatography-mass spectrometry/mass spectrometry. One hundred forty-three proteins were detected and their relative abundance was calculated using a reference sample generated from all PDSs. We performed heatmap analysis of all the detected proteins to display their expression patterns across different PDSs together with pathway enrichment analysis to reveal which processes that were connected to PDS protein composition. This protein dataset together with clinical information is useful to investigators studying the microenvironment of breast cancers. Further, after repopulating PDSs with either MCF7 or MDA-MB-231 cells, we quantified their gene expression profiles using RNA sequencing. These data were also compared to cells cultured in conventional 2D conditions, as well as to cells cultured as xenografts in immune-deficient mice. We investigated the overlap of genes regulated between these different culture conditions and performed pathway enrichment analysis of genes regulated by both PDS and xenograft cultures compared to 2D in both cell lines to describe common processes associated with both culture conditions. Apart from our described analyses of these systems, these data are useful when comparing different experimental model systems.     • Tumor cells interact with the microenvironment that specifically supports and promotes tumor development. Key components in the tumor environment have been linked to various aggressive cancer features and can further influence the presence of subpopulations of cancer cells with specific functions, including cancer stem cells and migratory cells. To model and further understand the influence of specific microenvironments we have developed an experimental platform using cell-free patient-derived scaffolds (PDSs) from primary breast cancers infiltrated with standardized breast cancer cell lines. This PDS culture system induced a series of orchestrated changes in differentiation, epithelial-mesenchymal transition, stemness and proliferation of the cancer cell population, where an increased cancer stem cell pool was confirmed using functional assays. Furthermore, global gene expression profiling showed that PDS cultures were similar to xenograft cultures. Mass spectrometry analyses of cell-free PDSs identified subgroups based on their protein composition that were linked to clinical properties, including tumor grade. Finally, we observed that an induction of epithelial-mesenchymal transition-related genes in cancer cells growing on the PDSs were significantly associated with clinical disease recurrences in breast cancer patients. Patient-derived scaffolds thus mimics in vivo-like growth conditions and uncovers unique information about the malignancy-inducing properties of tumor microenvironment. © 2019 The Authors

  • Erfarenheter, lärdomar och effekter med gränsöverskridande arbete för utbytesstudier

    • I Högskolan i Skövdes (HS) nuvarande utvecklingsplan beskrivs att samtliga utbildningsprogram som ges vid lärosätet skall erbjuda möjligheter till studier utomlands. Som en konsekvens har en strategisk satsning för att stimulera en ökad mobilitet vid HS initierats genom att Institutionen för informationsteknologi (IIT) tillsammans med verksamhetsstödet har deltagit i ett UHR-projekt med fokus på vägledningsprocessen i samband med mobilitet.Projektet har pågått under 18 månader och ett 10-tal deltagare som representerar de flesta roller som är involverade i internationaliseringsarbetet på IIT och verksamhetsstödet har ingått. Projektet har bedrivits främst som en serie av workshoppar där parallell datainsamling har skett med hjälp av intervjuer och enkätstudier.I detta bidrag vill vi visa några av de mål som projektet fokuserat på samt syftet med dessa. Projektet har haft följande mål:Identifiera minst tre partnerlärosäten som passar varje utbildningsprogram - Målet syftar till att kartlägga både existerande och nya partnerlärosäte för att på så sätt sänka tröskeln för studenter som är intresserade av utbyte, men som har svårt att hitta lämpliga alternativ.Tydliggöra roll- och ansvarsfördelning i mobilitetsprocessen - Detta mål syftar till att utforma processbeskrivningar för att tydliggöra roll- och ansvarsfördelning kring utresandeprocessen för programansvarig, ämnesföreträdare, internationell koordinator, studie- och karriärvägledaren, studenten och partnerlärosätet. Även kommunikationsaspekter och studentperspektiv beaktas i detta mål.Skapa adekvat vägledning och informationsinsatser gentemot studenterna - Syftet med målet är att utveckla, strukturera och systematisera informationsvägar och kommunikation mellan vägledning, programansvarig, partnerlärosäte och studenter.Identifiera och utvärdera nyckelfaktorerna för att förbättra stödet till studenterna, undanröja hinder i mobilitetsprocessen samt underlätta programansvarigas och studie- och karriärvägledarnas arbete med utresande studenter - Målet är att identifiera och utvärdera nyckelfaktorer som hindrar mobilitet som kan spridas internt på HS samt externt för att i förlängningen öka mobiliteten bland Sveriges studenter.Se till att samtliga kandidatprogram på IIT har en termin avsatt för utlandsstudier och undanröja de hinder som finns i befintliga programstrukturer - Syftet är att göra det enklare för studenter att under sin ordinarie studietid genomföra utbytesstudier utan att deras studier vid HS blir drabbade av förkunskapsstrukturer som hindrar fortsatta studier vid hemkomst.I studien inkludera de studenter som har varit intresserade eller sökt utbytesstudier men som inte kommit iväg på utbyte - Syftet är att skapa en god översikt över vilka skäl denna studentkategori har haft för att avstå utbytesstudier för att på så vis kunna förbättra existerande processer för utbytesstudier och därmed minimera risken att intresserade studenter väljer att avstå från utbytesstudier.Vid projektets avslut analyserades projektets direkta och indirekta interorganisatoriska effekter tillsammans med aktuell statistik för de studenter som under 2017 nominerats för utbytesstudier. En uppenbar effekt av projektets arbete är att antalet studenter som 2017 vid IIT nominerats för utbytesstudier ökat kraftigt. Likaså observeras ökade kunskaper om processen kring utbytesstudier och ett förbättrat studentperspektiv och bättre kunskaper om studenternas upplevelse av nomineringsprocessen.Under presentation vill vi visa upp fler detaljer från vår analys samt hur vi planerar att arbeta vidare med de resultat vi fått fram i projektet.

  • Branding ‘progressive’ security : The case of Sweden

    • Contemporary research on so-called Nordic branding has provided crucial insights into the social power of states and how various actors use and circulate ‘progressive’ nation brand tropes for political and commercial goals. Hitherto, the literature on Nordic branding has focused on a wide range of substantive issues, among other things, human rights, gender equality, social welfare and foreign aid, but considerably less attention has been paid to the topic of security. The present article adds to a small but established literature on how the security sphere is increasingly entangled with nation branding. In the Nordic region, we argue, the latter is particularly evident in the case of Sweden – one of the world’s largest per-capita arms exporters in the post-Cold War era but also a country known and often revered for its peaceful and progressive image. Focusing on the case of Sweden, the article contributes to knowledge of how defence industry-related actors (both public and private) draw on and frame nation branding tropes to sell and legitimise their products and services to both insiders (domestic constituents) and outsiders (the global security market).

    • In this work, the application of roller dies as an alternative to conventional dies in the wire drawing process – claimed in earlier works to offer many advantageous improvements – is evaluated. To this end, experimental procedures using a single stage drawing machine were conducted, drawing low and medium carbon steel wires using both roller dies and conventional dies. The two reduction processes were compared with respect to drawing force and the resulting wire temperature. Also the mechanical properties of the drawn wires were investigated. The obtained results have not shown any large differences between the two processes, and the main improvement using the roller die method was a better wire surface. This advantage was counteracted with downsides including higher wire temperature and lower strain at fracture.Moreover, a formula to theoretically calculate drawing force for the conventional drawing process was modified and used to calculate drawing force for the two reduction steps in the roller die cassette. The results showed that the proportion of inhomogeneous deformation was much higher in the rolling process as compared to the conventional process.In view of the outcomes in this work, the alternative of drawing wire through roller dies was not considered to be offering more improvements with respect to the conventional drawing method.

  • Identification of breast cancer stem cell related genes using functional cellular assays combined with single-cell RNA sequencing in MDA-MB-231 cells

    • Breast cancer tumors display different cellular phenotypes. A growing body of evidence points toward a population of cancer stem cells (CSCs) that is important for metastasis and treatment resistance, although the characteristics of these cells are incomplete. We used mammosphere formation assay and label-retention assay as functional cellular approaches to enrich for cells with different degree of CSC properties in the breast cancer cell line MDA-MB-231 and performed single-cell RNA sequencing. We clustered the cells based on their gene expression profiles and identified three subpopulations, including a CSC-like population. The cell clustering into these subpopulations overlapped with the cellular enrichment approach applied. To molecularly define these groups, we identified genes differentially expressed between the three subpopulations which could be matched to enriched gene sets. We also investigated the transition process from CSC-like cells into more differentiated cell states. In the CSC population we found 14 significantly upregulated genes. Some of these potential breast CSC markers are associated to reported stem cell properties and clinical survival data, but further experimental validation is needed to confirm their cellular functions. Detailed characterization of CSCs improve our understanding of mechanisms for tumor progression and contribute to the identification of new treatment targets. © 2019 Jonasson, Ghannoum, Persson, Karlsson, Kroneis, Larsson, Landberg and Ståhlberg.   • Additively manufactured conformal cooling tool holder for wire drawing utilizing triply periodic minimal surfaces

    • Wire is all around us and it forms joints in our structures, stabilizes our tires andtransports electricity. In almost every complex product there are components made fromwire. In wire drawing hot rolled material is drawn through a single or a series of tools calleddrawing dies, reducing the cross-section and enhancing the mechanical properties of thematerial. The tribological conditions in wire drawing are quite extreme and the high frictionbetween the wire and the die which leads to plenty of heat going into the die, resulting in hightool temperatures. Previous studies have shown that by reducing the tool temperature it ispossible to increase the productivity without risking an increased tool wear, this makes thecooling of the tool of high importance for the wire drawing process.Triply periodic minimal surfaces (TPMS) which have lately been enabled to be manufacturedby the use of additive manufacturing (AM) have shown great potential to be used for coolingapplications with demand on high efficiency. In this study a tool holder for wire drawing wasdesigned utilizing TPMS and was manufactured using laser powder bed fusion (LPBF). Thecooling efficiency of the manufactured tool holder was evaluated and compared to a con-ventional tool holder in an industrial wire drawing process. The study shows promisingresults on improving the cooling efficiency in the process by using the TPMS AM toolholder.

    • Components made from wire can be found in almost all complex products. In the wire drawing process, hot rolled wire is drawn through a single or a series of tools, which reduces the cross section of the wire and enhances the mechanical properties of the material. The tribological conditions in the process are extreme and the high frictional forces between the wire and the die results in high tool temperatures. Previous studies have shown that by reducing the temperature of the drawing tool it is possible to decrease the tool wear rate. Hence, cooling of the tool is of high importance in the wire drawing process.In this study, the possibilities to decrease the tool temperature by introducing conformal cooling in the drawing tool was investigated. Drawing tools made of cemented carbide like material were designed to utilize conformal cooling and manufactured by additive manufacturing. Results on cooling efficiency in an industrial-like wire drawing process are presented and discussed.

    • Wire drawing is a manufacturing process in which metal rods or wires are drawn through a single or a series of dies, reducing the wire cross-section and enhancing the mechanical properties of the wire. The tribological conditions in wire drawing are quite extreme and high friction between the wire and the die results in an increased die temperature. Previous studies have shown that by reducing the die temperature the lifetime of the die increases and thus efficient cooling of the die is of high importance.Additive manufacturing enables fabrication of tools with advanced conformal cooling channels with high cooling efficiency. This technique may, therefore, be of high importance in the design of the cooling system of drawing dies. In the present study, the effect of conformal cooling design of die holder on the die temperature, and thus die performance, was investigated. A die holder was manufactured by means of laser powder bed fusion (LPBF) in an EOS M290 machine using atomized corrosion resistant steel (Corrax). The cooling efficiency of the manufactured tool holder was evaluated in an industrial wire drawing process and further analysed using FEM modelling. This study shows promising results on improved cooling efficiency for die holder designed and manufactured by additive manufacturing.
