  • Tillstånd och trender för arter och deras livsmiljöer – rödlistade arter i Sverige 2015

    • 2015 års upplaga av den svenska rödlistan är den fjärde i ordningen. Den är baserad på IUCN:s rödlistningskriterier och revideras vart femte år. I rödlistan bedöms risken som enskilda arter av djur, växter och svampar löper att försvinna från Sverige. Bedömningen utförs av ArtDatabankens medarbetare i samverkan med över 100 externa experter, indelade i 14 expertkommittéer för olika organismgrupper. Under arbetet med 2015 års rödlista har tillstånd och trender bedömts för 21 600 arter och 1 318 lägre taxa (apomiktiska arter, underarter och varieteter), sammanlagt ca 22 900 taxa. Av de bedömda arterna klassificerades 2 029 som hotade (kategorierna CR, EN och VU) och 4 273 som rödlistade (inkluderar även kategorierna NT, RE och DD). Förhållandet mellan antalet rödlistade och antalet bedömda arter ar 19,8 %, vilket är ungefär samma värde som 2010 och 2005. I denna rapport jämförs antalet och andelen rödlistade arter mellan olika organismgrupper, biotoper, substrat och påverkansfaktorer. Texten ar indelad i en allmän del och åtta kapitel inriktade på olika landskapstyper. Landskapstyperna utgör en grov indelning av landets miljöer enligt följande kategorier: Skog, Jordbrukslandskap, Urbana miljöer, Fjäll, Våtmarker, Sötvatten, Havsstränder och Havsmiljöer. Skogen och jordbrukslandskapet är de artrikaste landskapstyperna med 1 800 respektive 1 400 arter som har en stark anknytning dit, och ytterligare flera hundra arter som förekommer där mer sporadiskt. De faktorer som påverkar flest rödlistade arter i Sverige är skogsavverkning och igenväxning, som båda utgör ett hot mot vardera ca 30 % av de rödlistade arterna. Avverkning minskar arealen av skog där naturliga strukturer och naturlig dynamik upprätthålls, och den orsakar därmed förlust av livsmiljöer. Igenväxning orsakas av ett antal faktorer, bland annat upphörande hävd (bete och slåtter), gödsling, trädplantering och brist på naturliga störningsregimer som t.ex. regelbundna översvämningar kring vattendrag och sjöar. Andra viktiga påverkansfaktorer är fiske, torrläggning av våtmarker, tillbakagång hos värdarter (främst alm och ask som drabbats av invasiva svampsjukdomar), klimatförändringar och konkurrens från invasiva arter. IUCN:s rödlisteindex beräknas för ett urval av de bedömda organismgrupperna. Rödlisteindex visar att skillnaderna mellan rödlistorna från 2000, 2005, 2010 och 2015 är små. Ett par undantag finns dock. Groddjur och stora däggdjur har fått en något förbättrad situation sedan 2000. Totalt förefaller det ändå som att trycket mot Sveriges artstock har förblivit relativt konstant under de senaste 15 åren.

  • A field investigation of the size, morphology and chemical composition of airborne particles in rail transport

    • The health effects of inhalable airborne particles are well documented. In the European Union the European Council mandates that the level of airborne particles with a diameter smaller than 10 µm (PM10) must not exceed an annual average of 40 µg/m3. Examples of possible sources from rail transport are mechanical brakes, wheel rail contact, current collectors, ballast, sleepers and masonry structures. In this regard, a series of field tests have been conducted on a regular Swedish track using a regional train instrumented with: particle measurement devices, temperature sensors in brake pads and sensors to measure the magnitude of train speed and a GPS.Two sampling points for airborne particles were designated in the train under frame. One of the sampling points was near a pad to rotor disc brake contact and a second global sampling point was chosen under the frame, but not near a mechanical brake or the wheel-rail contact. The first one was highly influenced by brake pad wear debris and the other one was influenced by all of the brake pads, wheel and rail wear debris as well as re-suspension. In each sampling points, three tubes were linked to three particle measurement devices. Two sets of Ptrak, Dustrak and Grimm devices were used. The Ptrak 8525 was an optical particle measurement device which could measure particle diameter in the size interval of 20 nm up to 1 micrometer. The Dustrak was used to measure particle mass concentration. The Grimm 1.109 was an aerosol spectrometer which counted number of particles from 0.25 micrometer to 32 micrometer in 31 intervals. These two Grimm devices were equipped with Millipore filters in the devices outlets to capture particles for further studies on morphology and matter of particles.The total number and size distribution of the particles for these two sampling points were registered and evaluated in different situations such as activating and deactivating electrical brake or train curve negotiating.During braking, three peaks of 250 nm, 350 nm and 600 nm in diameter, with the 350 nm peak dominating were identified in the fine particle region. In the coarse particle region, a peak of around 3-6 µm in diameter was discovered. The brake pad temperature effects on particle size distribution were also investigated and the results showed that the peak around 250 nm increased. Furthermore, the activation of electrical braking significantly reduced the number of airborne particles.A SEM was used to capture the images from collected particles on filters. Furthermore, an ICP-Ms method was used to investigate the elemental contents of the particulates on the filter.  In this case the main contribution belonged to Fe, Si, Al, Ca, Cu, Zn. The higher amount of some elements weights such as calcium, silicon, sodium and aluminum in the global sampling point filters revealed that ballast and concrete sleepers were the main sources for these particles although some of them originated from rail, wheel, brake disc and brake pad as well.

    • Brake pads on wheel-mounted disc brakes are often used in rail transport due to their good thermal properties and robustness. During braking, both the disc and the pads are worn. This wear process generates particles that may become airborne and thus affect human health. The long term purpose of ‘Airborne particles in Rail transport’ project is to gain knowledge on the wear mechanisms in order to find means of controlling the number and size distribution of airborne particles. In this regard, a series of full-scale field tests and laboratory tests with a pin-on-disc machine have been conducted. The morphology and the matter of particles, along with their size distribution and concentration, have been studied. The validity of results from the pin-on-disc simulation has been verified by the field test results. Results show an ultra-fine peak for particles with a diameter size around 100 nm in diameter, a dominant fine peak for particles with a size of around 350 nm in diameter, and a coarse peak with a size of 3-7 μm in diameter. Materials such as iron, copper, aluminium, chromium, cobalt, antimony, and zinc have been detected in the nano-sized particles.

    • Inhalable airborne particles have inverse health affect. In railways, mechanical brakes, the wheel–rail contact, current collectors, ballast, sleepers, and masonry structures yield particulate matter. Field tests examined a Swedish track using a train instrumented with particle measurement devices, brake pad temperature sensors, and speed and brake sensors. The main objective of this field test was to study the characteristics of particles generated from disc brakes on a running train with an on-board measuring set-up.Two airborne particle sampling points were designated, one near a pad–rotor disc brake contact and a second under the frame, not near a mechanical brake or the wheel–rail contact; the numbers and size distributions of the particles detected were registered and evaluated under various conditions (e.g. activating/deactivating electrical brakes or negotiating curves). During braking, three speed/temperature-dependent particle peaks were identified in the fine region, representing particles 280 nm, 350 nm, and 600 nm in diameter. In the coarse region, a peak was discerned for particles 3–6 μm in diameter. Effects of brake pad temperature on particle size distribution were also investigated. Results indicate that the 280 nm peak increased with increasing temperature, and that electrical braking significantly reduced airborne particle numbers. FESEM images captured particles sizing down to 50 nm. The ICP-MS results indicated that Fe, Cu, Zn, Al, Ca, and Mg were the main elements constituting the particles.

  • Effect of General Adiposity and Central Body Fat Distribution on the Circulating Metabolome : A Multi-Cohort Nontargeted Metabolomics Observational and Mendelian Randomization Study

    • Obesity is associated with adverse health outcomes, but the metabolic effects have not yet been fully elucidated. We aimed to investigate the association between adiposity with circulating metabolites and to address causality with Mendelian randomization (MR). Metabolomics data was generated by non-targeted ultra-performance liquid-chromatography coupled to time-of-flight mass-spectrometry in plasma and serum from three population-based Swedish cohorts: ULSAM (N=1,135), PIVUS (N=970), and TwinGene (N=2,059). We assessed associations between general adiposity measured as body mass index (BMI) and central body fat distribution measured as waist-to-hip ratio adjusted for BMI (WHRadjBMI) with 210 annotated metabolites. We employed MR analysis to assess causal effects. Lastly, we attempted to replicate the MR findings in the KORA and TwinsUK cohorts (N=7,373), the CHARGE consortium (N=8,631), the Framingham Heart Study (N=2,076) and the DIRECT consortium (N=3,029). BMI was associated with 77 metabolites, while WHRadjBMI was associated with 11 and 3 metabolites in women and men, respectively. The MR analyses in the Swedish cohorts suggested a causal association (p-value <0.05) of increased general adiposity and reduced levels of arachidonic acid, dodecanedioic acid and lysophosphatidylcholine (P-16:0) as well as with increased creatine levels. The replication effort provided support for a causal association of adiposity on reduced levels of arachidonic acid (p-value 0.03). Adiposity is associated with variation of large parts of the circulating metabolome, however causality needs further investigation in well-powered cohorts.

    • Antibiotic resistance is a major worldwide concern, and new drugs with mechanistically novel modes of action are urgently needed. Here, we report the structure-based drug design, synthesis, and evaluation in vitro and in cellular systems of sialic acid derivatives able to inhibit the bacterial sialic acid symporter SiaT. We designed and synthesized 21 sialic acid derivatives and screened their affinity for SiaT by a thermal shift assay and elucidated the inhibitory mechanism through binding thermodynamics, computational methods, and inhibitory kinetic studies. The most potent compounds, which have a 180-fold higher affinity compared to the natural substrate, were tested in bacterial growth assays and indicate bacterial growth delay in methicillin-resistant Staphylococcus aureus. This study represents the first example and a promising lead in developing sialic acid uptake inhibitors as novel antibacterial agents.

    • A 1 yr study was conducted along a brackish-water production gradient to enhance the understanding of factors governing pelagic food web function. This was achieved by measuring carbon transfer efficiency (TE) from the basal resource to an intermediate trophic level. TE was defined as mesozooplankton carbon consumption rate divided by production at the basal trophic level, which is composed of phytoplankton and bacteria. A north–south transect in the Baltic Sea was used as a model system, with 2 stations each in the Bothnian Bay, Bothnian Sea and Baltic Proper being sampled 5 to 8× during 2006. In addition, data from monitoring programmes were used, which comprised 10 to 22 samplings stn–1. TE was expected to be governed by the size distribution of phytoplankton and due to the nutrient gradient, we expected to find an optimal cell size, and thus also a high TE, in the intermediate-productive Bothnian Sea. The basal production during summer/autumn increased 5-fold from north to south, while the mesozooplankton carbon consumption rate exhibited a peak inthe Bothnian Sea, being ~3× higher than in both Bothnian Bay and Baltic Proper. TE was found to be intermediate in the Bothnian Bay (average: 0.8), highest in the Bothnian Sea (1.6), and lowest in the Baltic Proper (0.2). We suggest that the variation in carbon transfer efficiency can be explained by the composition of the phytoplankton community, the abundance balance between copepods and cladocerans, as well as the species composition of mesozooplankton in relation to the size structure of phytoplankton.

  • Identification of the BCAR1-CFDP1-TMEM170A Locus as a Determinant of Carotid Intima-Media Thickness and Coronary Artery Disease Risk

    • Background-Carotid intima-media thickness (cIMT) is a widely accepted marker of subclinical atherosclerosis. To date, large-scale investigations of genetic determinants of cIMT are sparse. Methods and Results-To identify cIMT-associated genes and genetic variants, a discovery analysis using the Illumina 200K CardioMetabochip was conducted in 3430 subjects with detailed ultrasonographic determinations of cIMT from the IMPROVE (Carotid Intima Media Thickness [IMT] and IMT-Progression as Predictors of Vascular Events in a High Risk European Population) study. Segment-specific IMT measurements of common carotid, bifurcation, and internal carotid arteries, and composite IMT variables considering the whole carotid tree (IMTmean, IMTmax, and IMTmean-max), were analyzed. A replication stage investigating 42 single-nucleotide polymorphisms for association with common carotid IMT was undertaken in 5 independent European cohorts (total n=11 590). A locus on chromosome 16 (lead single-nucleotide polymorphism rs4888378, intronic in CFDP1) was associated with cIMT at significance levels passing multiple testing correction at both stages (array-wide significant discovery P=6.75x10(-7) for IMTmax; replication P=7.24x10(-6) for common cIMT; adjustments for sex, age, and population substructure where applicable; minor allele frequency 0.43 and 0.41, respectively). The protective minor allele was associated with lower carotid plaque score in a replication cohort (P=0.04, n=2120) and lower coronary artery disease risk in 2 case-control studies of subjects with European ancestry (odds ratio [95% confidence interval] 0.83 [0.77-0.90], P=6.53x10(-6), n=13 591; and 0.95 [0.92-0.98], P=1.83x10(-4), n= 82 297, respectively). Queries of human biobank data sets revealed associations of rs4888378 with nearby gene expression in vascular tissues (n=126-138). Conclusions-This study identified rs4888378 in the BCAR1-CFDP1-TMEM170A locus as a novel genetic determinant of cIMT and coronary artery disease risk in individuals of European descent.
