Household responsiveness to residential demand response strategies - Results and policy implications from a Swedish field study

- To realize the benefits of smart grids, residential demand response (DR) aims to increase demand flexibility by influence household electricity consumption. Although price-based DR programs have shown potential, there is a need to further investigate the effectiveness of DR in energy strategy and policy development. The evaluation of DR has focused on the impact on overall power demand, assuming that consumers are economically rational decision-maker. However, recent findings suggest that consumer responses have been insufficient and calls have been made to identify novel evaluation approaches that better reflect the human dimension of energy consumption. Continuing this line of enquiry, this paper aims to investigate the effectiveness of DR and explore the potential of environmental incentives for increased consumer engagement. We propose an interdisciplinary evaluation framework to understand variations in household responsiveness to DR strategies, which is tested in a Swedish DR field trial covering 136 households during 2017. Results suggest that the effectiveness of DR varies widely across household type; ranging from substantial reductions in overall consumption and during peak periods, to increases in consumption during peak periods. Furthermore, a clear favor of price incentives, compared to environmental incentives, as the most efficient strategy to increase demand flexibility was observed.

The globalizability of temporal discounting

- Economic inequality is associated with preferences for smaller, immediate gains over larger, delayed ones. Such temporal discounting may feed into rising global inequality, yet it is unclear whether it is a function of choice preferences or norms, or rather the absence of sufficient resources for immediate needs. It is also not clear whether these reflect true differences in choice patterns between income groups. We tested temporal discounting and five intertemporal choice anomalies using local currencies and value standards in 61 countries (N = 13,629). Across a diverse sample, we found consistent, robust rates of choice anomalies. Lower-income groups were not significantly different, but economic inequality and broader financial circumstances were clearly correlated with population choice patterns. Ruggeri et al. find in a study of 61 countries that temporal discounting patterns are globally generalizable. Worse financial environments, greater inequality and high inflation are associated with extreme or inconsistent long-term decisions.

Chromosomal Abnormalities and Prognosis in NPM1-Mutated Acute Myeloid Leukemia : A Pooled Analysis of Individual Patient Data From Nine International Cohorts

- PURPOSE: Nucleophosmin 1 (NPM1) mutations are associated with a favorable prognosis in acute myeloid leukemia (AML) when an internal tandem duplication (ITD) in the fms-related tyrosine kinase 3 gene (FLT3) is absent (FLT3-ITDneg) or present with a low allelic ratio (FLT3-ITDlow). The 2017 European LeukemiaNet guidelines assume this is true regardless of accompanying cytogenetic abnormalities. We investigated the validity of this assumption.METHODS: We analyzed associations between karyotype and outcome in intensively treated patients with NPM1(mut)/FLT3-ITDneg/low AML who were prospectively enrolled in registry databases from nine international study groups or treatment centers.RESULTS: Among 2,426 patients with NPM1(mut)/FLT3-ITDneg/low AML, 2,000 (82.4%) had a normal and 426 (17.6%) had an abnormal karyotype, including 329 patients (13.6%) with intermediate and 83 patients (3.4%) with adverse-risk chromosomal abnormalities. In patients with NPM1(mut)/FLT3-ITDneg/low AML, adverse cytogenetics were associated with lower complete remission rates (87.7%, 86.0%, and 66.3% for normal, aberrant intermediate, and adverse karyotype, respectively; P < .001), inferior 5-year overall (52.4%, 44.8%, 19.5%, respectively; P < .001) and event-free survival (40.6%, 36.0%, 18.1%, respectively; P < .001), and a higher 5-year cumulative incidence of relapse (43.6%, 44.2%, 51.9%, respectively; P = .0012). These associations remained in multivariable mixed-effects regression analyses adjusted for known clinicopathologic risk factors (P < .001 for all end points). In patients with adverse-risk chromosomal aberrations, we found no significant influence of the NPM1 mutational status on outcome.CONCLUSION: Karyotype abnormalities are significantly associated with outcome in NPM1(mut)/FLT3-ITDneg/low AML. When adverse-risk cytogenetics are present, patients with NPM1(mut) share the same unfavorable prognosis as patients with NPM1 wild type and should be classified and treated accordingly. Thus, cytogenetic risk predominates over molecular risk in NPM1(mut)/FLT3-ITDneg/low AML.

- This paper argues that the present dominant economic system rests upon unquestioned beliefs that are in a deep contradiction with the pursuit of sustainable development. The economics of conventions is used as an analytical framework through which to demonstrate the conflict between the dominant conventions underpinning societal development and the objectives of sustainable development. It suggests that a trajectory towards the objectives of sustainable development should be managed through a reflexion concerning the conventional principles required to be adopted in order to favour the emergence of a new conventional regime. The principles of proximity, the increase in individual and collective capabilities, and participative democracy are presented as possible principles that could be adopted in order to favour the emergence of a new conventional regime.

- This chapter presents a case study of how Accus, a small Swedish company, worked on developing a circular business model for light sign production and installation to become more sustainable. Drawing on Actor Network Theory (ANT), the Accus case shows that circular business model development is a cooperative endeavor that rests on bringing together a large and changing array of human, as well as non-human, actors in a development process that is hesitant, imprecise, provisory, contingent, and reversible. The case exemplifies the difficulties for a small company to enroll other actors in a transition to circular business, and initiate a shift toward a parsimonious material order that challenges, transforms, and replaces worked-in linear supplier-customer relationships. Good intentions only reach up to a certain point. If more than exceptional firms are to successfully find partners to translate their circular ambitions into circular business practices, the competitive strength of linear solutions needs to be drastically reduced and delegitimized.

Claudin-3-deficient C57BL/6J mice display intact brain barriers

- The tight junction protein claudin-3 has been identified as a transcriptional target of the Wnt/beta-catenin signaling pathway regulating blood-brain barrier (BBB) maturation. In neurological disorders loss of claudin-3 immunostaining is observed at the compromised BBB and blood-cerebrospinal fluid barrier (BCSFB). Although these observations support a central role of claudin-3 in regulating brain barriers' tight junction integrity, expression of claudin-3 at the brain barriers has remained a matter of debate. This prompted us to establish claudin-3-/-C57BL/6J mice to study the role of claudin-3 in brain barrier integrity in health and neuroinflammation. Bulk and single cell RNA sequencing and direct comparative qRT-PCR analysis of brain microvascular samples from WT and claudin-3-/- mice show beyond doubt that brain endothelial cells do not express claudin-3 mRNA. Detection of claudin-3 protein at the BBB in vivo and in vitro is rather due to junctional reactivity of anti-claudin-3 antibodies to an unknown antigen still detected in claudin-3-/- brain endothelium. We confirm expression and junctional localization of claudin-3 at the BCSFB of the choroid plexus. Our study clarifies that claudin-3 is not expressed at the BBB and shows that absence of claudin-3 does not impair brain barrier function during health and neuroinflammation in C57BL/6J mice.
