    • We used findings from the Global Burden of Disease 2015 study to update our previous publication on the burden of diabetes and chronic kidney disease due to diabetes (CKD-DM) during 1990-2015. We extracted GBD 2015 estimates for prevalence, mortality, and disability-adjusted life years (DALYs) of diabetes (including burden of low vision due to diabetes, neuropathy, and amputations and CKD-DM for 22 countries of the EMR from the GBD visualization tools. In 2015, 135,230 (95% UI 123,034-148,184) individuals died from diabetes and 16,470 (95% UI 13,977-18,961) from CKD-DM, 216 and 179% increases, respectively, compared to 1990. The total number of people with diabetes was 42.3 million (95% UI 38.6-46.4 million) in 2015. DALY rates of diabetes in 2015 were significantly higher than the expected rates based on Socio-demographic Index (SDI). Our study showed a large and increasing burden of diabetes in the region. There is an urgency in dealing with diabetes and its consequences, and these efforts should be at the forefront of health prevention and promotion.

    • BackgroundStipagrostis pennata (Trin.) De Winter is an important species for fixing sand in shifting and semi-fixed sandy lands, for grazing, and potentially as a source of lignocellulose fibres for pulp and paper industry. The seeds have low viability, which limits uses for revegetation. Somatic embryogenesis offers an alternative method for obtaining large numbers of plants from limited seed sources.ResultsA protocol for plant regeneration from somatic embryos of S. pennata was developed. Somatic embryogenesis was induced on Murashige & Skoog (MS) medium supplemented with 3 mg.L-1 2,4-D subsequently shoots were induced on MS medium and supplemented with 5 mg.L-1 zeatin riboside. The highest shoots induction was obtained when embryogenic callus derived from mature embryos (96%) in combination with MS filter-sterilized medium was used from Khuzestan location. The genetic stability of regenerated plants was analysed using ten simple sequence repeats (SSR) markers from S. pennata which showed no somaclonal variation in regenerated plants from somatic embryos of S. pennata. The regenerated plants of S. pennata showed genetic stability without any somaclonal variation for the four pairs of primers that gave the expected amplicon sizes. This data seems very reliable as three of the PCR products belonged to the coding region of the genome.Furthermore, stable expression of GUS was obtained after Agrobacterium-mediated transformation using a super binary vector carried by a bacterial strain LBA4404.ConclusionTo our knowledge, the current work is the first attempt to develop an in vitro protocol for somatic embryogenesis including the SSR marker analyses of regenerated plants, and Agrobacterium-mediated transformation of S. pennata that can be used for its large-scale production for commercial purposes.

  • Relative Importance of Different Attributes of Graphic Health Warnings on Tobacco Packages in Viet Nam

    • Graphic health warnings (GHW) on tobacco packages have proven to be effective in increasing quit attempts among smokers and reducing initial smoking among adolescents. This research aimed to examine the relative importance of different attributes of graphic health warnings on tobacco packages in Viet Nam. A discrete choice experimental (DCE) design was applied with a conditional logit model. In addition, a ranking method was used to list from the least to the most dreadful GHW labels. With the results from DCE model, graphic type was shown to be the most important attribute, followed by cost and coverage area of GHW. The least important attribute was position of the GHW. Among 5 graphic types (internal lung cancer image, external damaged teeth, abstract image, human suffering image and text), the image of lung cancer was found to have the strongest influence on both smokers and non-smokers. With ranking method, the image of throat cancer and heart diseases were considered the most dreadful images. GHWs should be designed with these attributes in mind, to maximise influence on purchase among both smokers and non-smokers.

  • Colchicine Mutagenesis from Long-term Cultured Adventitious Roots Increases Biomass and Ginsenoside Production in Wild Ginseng (Panax ginseng Mayer)

    • Panax ginseng Mayer is a perennial herb that has been used as a medicinal plant in Eastern Asia for thousands of years. The aim of this study was to enhance root biomass and ginsenoside content in cultured adventitious roots by colchicine mutagenesis. AdventitiousP. ginsengroots were treated with colchicine at different concentrations (100, 200, and 300 mg.L-1) and for different durations (1, 2, and 3 days). Genetic variability of mutant lines was assessed using random amplification of polymorphic DNA (RAPD) analysis. Ginsenoside biosynthesis gene expression, ginsenoside content, enzyme activities, and performance in bioreactor culture were assessed in four mutant lines (100-1-2, 100-1-18, 300-1-16, and 300-2-8). The results showed that ginsenoside productivity was enhanced in all mutant lines, with mutant 100-1-18 exhibiting the most pronounced increase (4.8-fold higher than the control). Expression of some ginsenoside biosynthetic enzymes was elevated in mutant lines. Enzyme activities varied among lines, and lipid peroxidation activity correlated with root biomass. All four lines were suitable for bioreactor cultivation, with mutant 100-1-18 exhibiting the highest biomass after culture scale-up. The results indicated that colchicine mutagenesis ofP. ginsengroots increased biomass and ginsenosides production. This technique, and the root lines produced in this study, may be used to increase industrial yields ofP. ginsengbiomass and ginsenosides.

    • Key message The optimum condition for bioreactor culture of Polygonum multiflorum is full-strength MS medium supplemented with 2 mg center dot L-1 IBA and 5% sucrose after 4 weeks of culture. FT-IR analysis can rapidly discriminate among root samples based on total metabolite equivalence.Bioreactor cultures have been used for biomass production and bioactive compounds accumulation in adventitious root cultures of medicinal plants. In this study, we evaluated the effects of different auxin types [indole-3-butyric acid (IBA) and 1-naphthaleneacetic acid (NAA)] and concentrations (0.5, 1.0, 2.0 and 4.0 mg center dot L-1), Murashige and Skoog (MS) medium salt strength (0.25, 0.5, 0.75, 1.0, 1.5 and 2X), and sucrose concentrations (0, 1.5, 3, 5, 7 and 10%) on Polygonum multiflorum Thunb. adventitious root cultures in a 3-L balloon-type bubble bioreactor (BTBB). IBA (1, 2, and 4 mg center dot L-1) was more effective than NAA in promoting root growth. Additionally, low MS salt strength (0.25 and 0.5X MS) increased the accumulation of total phenolics and flavonoids but reduced biomass accumulation. Four weeks of culture in full-strength MS medium supplemented with 2 mg center dot L-1 IBA and 5% sucrose resulted in the highest root biomass [98.46 g center dot L-1 fresh weight (FW); 13.46 g center dot L-1 dry weight (DW)] and bioactive compounds accumulation (total phenolics compounds, 53.08 mg center dot g(-1) DW; total flavonoids, 25.10 mg center dot g(-1) DW). To determine whether metabolic fingerprinting of whole-cell extracts could be used to compare metabolic equivalence of P. multiflorum root samples, we treated adventitious roots with different culture conditions, and analyzed the treated adventitious roots and natural roots by Fourier transform infrared (FT-IR) spectroscopy. The results showed that the metabolic pattern of adventitious root samples was similar under different culture conditions; however, these samples could be discriminated from each other in pilot-scale bioreactors. Overall, our study provides useful information for industrial-scale cultivation of P. multiflorum adventitious roots.

  • Direct observation of aliphatic structures in soot particles produced in low-pressure premixed ethylene flames via online Raman spectroscopy

    • Raman spectra of soot particles were monitored in the gaseous flow extracted from the burning regions of two low-pressure premixed ethylene flames, for the first time. The flame conditions were chosen to explore the diversity of soot nanostructure in slightly sooting flames. Evaluation of the Raman spectral parameters revealed that the soot particles exhibited a strongly disordered structure and a large proportion of sp hybridization of the carbon. The appearance of sp carbon chains composing up to 30% of the total carbon content as well as an olefinic component may indicate their important role in soot nucleation and growth in low pressure ethylene flames. Hence, Raman spectroscopy of soot particles in the aerosol phase revealed that accretion and cyclization of the aliphatic carbon including sp carbon chains could thus be of importance for the initial soot growth and require more investigation.

    • Somatic embryogenesis (SE) is an in vitro clonal propagation method increasingly in use for large scale clonal propagation of conifer plants. However, asynchronous development of immature somatic embryos into mature embryos lowers the yield of plants and limits the benefits for industrial applications. Through the development of an automated harvesting system (SE Fluidics system) for singulation, imaging and selection of mature embryos at the correct developmental stages, yields can be improved. Image analysis software developed for use with the SE Fluidics system has been utilised to characterize and sort mature embryos according to their morphological measurements related to developmental capacity. Here a set of imaging data from the SE Fluidics system has been analysed with the goal of further refining the selection criteria for germination-competent mature embryos.(c) 2021 IAgrE.   • Polarization effects in Raman spectroscopy of light-absorbing carbon

    • Raman spectroscopy is widely used for characterization of carbon-based mate- rials including light-absorbing carbonaceous (LAC) matter. However, informa- tion on polarization properties of these materials are generally lacking, and often the polarization characteristics of experiments are not presented in the literature thereby giving uncertainty to presented data. In this work, the polari- zation properties of various LACs are studied using Raman spectroscopy. The Raman spectra have been compared while varying the relative polarization, particularly the relative direction of the electric field between the incident light and the detected scattered light. Specifically, the intensity ratio between the D1 and G peaks (ID1/IG) was analyzed in parallel and perpendicular polarization configurations for samples taken from a sooting flame, in the exhaust of a soot generator and carbon blacks. In this work, the parallel and perpendicular polarization configurations mean that the polarization of the detected scattered light is parallel and orthogonal, respectively, to the linearly polarized incident light. It was found that the (ID1/IG) ratio decreased in the more ordered carbo- naceous structures when changing the polarization configuration from parallel to perpendicular. Additionally, based on the depolarization ratios (Dp < 0.75), the prominent Raman peaks of LAC were found to originate from totally sym- metric vibrations. For the G peak, the depolarization ratio was measured to be in the range 0.46–0.56 for all LAC materials.

  • Raman spectroscopy of mini-CAST soot with various fractions of organic compounds: Structural characterization during heating treatment from 25 °C to 1000 °C

    • Soot particles undergo considerable changes in composition, morphology, as well as in internal structure during formation from the incipient particles to more mature aggregates, and still there is a lack of understanding of these mechanisms. In this study, soot produced by a mini-CAST soot generator was probed using Raman spectroscopy with focus on soot with large variation in size and fraction of organic compounds. The volatility of organic compounds and changes in internal bonding structures of the soot was surveyed by heating the samples from room temperature up to 1000 °C in a flow of inert N2 gas. The soot rich in organics showed more complex Raman spectra and stronger photoluminescence background. In particular, Raman signatures interpreted as the CH “out-of-plane” mode, ethers COC, and carbonyl CO groups were observed. During the heating treatment, these signatures disappeared, which was related to vaporization of the organics. Moreover, an enhancement of Raman band intensity of mature soot during heating treatment was observed for the first time. Our study thus brings new information on structural and compositional changes for soot during heating treatment in an inert atmosphere.
