    • C3 is common to all pathways of complement activation augmenting ischemia/reperfusion (I/R)-induced myocardial injury and cardiac dysfunction. Complement inhibition with the complement regulatory protein, C1 inhibitor (C1INH), obviously exerts cardioprotective effects. Here, we examine whether C1INH regulates C3 activity in the ischemic myocardial tissue. C1INH markedly suppressed C3 mRNA expression and protein synthesis in both a model of I/R-induced rat acute myocardial infarction (AMI) and the cultured rat H9c2 heart myocytes. At least, this regulation was at the transcriptional level in response to oxygen tension. In vitro, C3 deposition on, and binding to, the surface of rat myocardial cells were significantly blocked by C1INH treatment. C1INH could inhibit classical complement-mediated cell lysis via suppressing the biological activity of C3. Therefore, C1INH, in addition to inhibition of the systemic complement activation, prevents myocardial cell injury via a direct inhibitory role in the local myocardial C3 activity.

    • Humidity measurement has been of extreme importance in both conventional environment monitoring and emerging digital health management. State-of-the-art flexible humidity sensors with combined structures, however, lack sensing reliability when they subject to high humidity with condensation and/or liquid water invasion. Here, we report a free-standing humidity sensor by creating a stable open porous graphite structure and controlling the number of oxygen-related groups at the molecular level. The sensor indicates high sensing reliability against water liquid wetting and machine washing as well as many mechanical deformations. We demonstrate the sensor has wide potential applications in challenge environmental monitoring and wearable body area sensing networks. Particularly, the concept of the humidity sensing strategy is applicative to not only cellulose-based materials such as cotton and linen, but also protein-based materials such as silk, paving a new route for producing high-performance and cost-effective humidity sensors.

    • Complement activation augments myocardial cell injury and apoptosis during ischemia/reperfusion (I/R), whereas complement system inhibition with C1 inhibitor (C1INH), a serine protease inhibitor, exerts markedly cardioprotective effects. Our recent data demonstrate that C1INH prevents vascular endothelial cell apoptosis and a "modified" form of the reactive center loop-cleaved, inactive C1INH (iC1INH) plays an anti-inflammatory role in endotoxin shock. The aim of this study was to determine whether C1INH protects against myocardial cell injury via an anti-apoptotic activity or anti-inflammatory effect. In a rat model of acute myocardial infarction (AMI) induced by I/R, administration of C1INH protected against cardiomyocytic apoptosis via normalization of ratio of the Bcl-2/Bax expression in the myocardial infarct area. C1INH improved parameters of cardiac function and hemodynamics and reduced myocardial infarct size (MIS). In addition, myocardial and blood myeloperoxidase (MPO) activity, a marker of neutrophil infiltration, was decreased by treatment of C1INH. In cultured H9c2 rat cardiomyocytic cells, C1INH blocked hypoxia/reoxygenation-induced apoptosis in the absence of sera associated with inhibition of cytochrome c translocation and suppression of caspase-3 activation. The proportion of Bcl-2/Bax expression induced by hypoxia/reoxygenation was reversed by C1INH. Importantly, iC1INH also revealed these similar effects, indicating that C1INH has a direct anti-apoptotic activity. Therefore, these studies support the hypothesis that C1INH, in addition to inhibition of activation of the complement and contact systems, improves outcome in I/R-mediated myocardial cell injury via an anti-apoptotic activity independent of serine protease inhibitory activity.

  • Construction of an Iodine Diffusion Barrier Using Network Structure Silicone Resin for Stable Perovskite Solar Cells

    • Long-term stability of organic-inorganic hybrid perovskite solar cells (PSCs) is inhibited by ion diffusion. Herein, we introduce a thermally stable and hydrophobic silicone resin layer with a network structure as an interfacial layer between the perovskite and the hole-transporting layer ( HTL). Experimental and theoretical investigations confirm that the small Si-O-Si unit in the network forms both Si-I and Pb-O bonds with the perovskite surface, which physically and chemically inhibit the diffusion and self-release of iodine. Besides, the silicone resin layer suppresses the thermal crystallization of spiro-OMeTAD and optimizes the interfacial energy level alignment for hole extraction. The power conversion efficiency (PCE) of a perovskite solar cell with a silicone resin layer is improved to 21.11%. The device maintains more than 90.1% of its original PCE after 1200 h under 85 degrees C thermal stress, 99.6% after 2000 h under RH similar to 55 +/- 5%, and 83% of its original PCE after light soaking in air for 1037 h.

  • The selection strategy of ammonium-group organic salts in vapor deposited perovskites: From dimension regulation to passivation

    • Dimension regulation and defect passivation are two key strategies for highly efficient and stable perovskite solar cell. Vapor deposition of perovskite is a toxic-solvent-free method for large-scale fabrication. However, without the assistance of solvent for crystal optimization, effective structural regulation and defect passivation become challenging. Here, detailed investigations on the structural evolution of perovskite thin film are carried out in sequential vapor deposition using mixed-vapor (R-NH3I/MAI). Correlation between electron donating ability of R-NH3I (BAI, PEAI, PMAI and ALI) molecule and the way of structural transition is established. It is found that RNH3I with stronger electron-donating ability promoted the phase transition from three-dimensional (3D) to twodimensional (2D) perovskite. Typically, the n value from 1 to 5 can be tuned by reaction time or component control using BAI with the strongest electron donating ability. R-NH3I with weak electron-donating ability suppresses the 3D to 2D transition, but enhances the defect passivation effect. The ALI with the weakest electron donating ability shows the best passivation effect, leading to the best device performance than that of the control 3D device, with PCE of 18.23% (0.045 cm2) and 15.48% (1 cm2) and the significantly improved stability. This study provides the evidence that the concept of Lewis acid-base reaction is applicable in vapor deposition, which provides us with the selection guide of R-NH3I molecules for structural design in vapor fabrication of perovskite thin film.

  • C1 inhibitor-mediated protection from sepsis.

    • C1 inhibitor (C1INH) protects mice from lethal Gram-negative bacterial LPS-induced endotoxin shock and blocks the binding of LPS to the murine macrophage cell line, RAW 264.7, via an interaction with lipid A. Using the cecal ligation and puncture (CLP) model for sepsis in mice, treatment with C1INH improved survival in comparison with untreated controls. The effect was not solely the result of inhibition of complement and contact system activation because reactive center-cleaved, inactive C1INH (iC1INH) also was effective. In vivo, C1INH and iC1INH both reduced the number of viable bacteria in the blood and peritoneal fluid and accelerated killing of bacteria by blood neutrophils and peritoneal macrophages. In vitro, C1INH bound to bacteria cultured from blood or peritoneal fluid of mice with CLP-induced sepsis, but had no direct effect on bacterial growth. However, both C1INH and iC1INH enhanced the bactericidal activity of blood neutrophils and peritoneal exudate leukocytes. C1INH-deficient mice (C1INH-/- mice) subjected to CLP had a higher mortality than did wild-type littermate mice. Survival of C1INH-/- mice was significantly increased with two doses of C1INH, one given immediately following CLP, and the second at 6 h post-CLP. C1INH may be important in protection from sepsis through enhancement of bacterial uptake by, and/or bactericidal capacity of, phagocytes. Treatment with C1INH may provide a useful additional therapeutic approach in some patients with peritonitis and/or sepsis.

    • Reaction resonances, or transiently stabilized transition-state structures, have proven highly challenging to capture experimentally. Here, we used the highly sensitive H atom Rydberg tagging time-of-flight method to conduct a crossed molecular beam scattering study of the F + H2 → HF + H reaction with full quantum-state resolution. Pronounced forward-scattered HF products in the v′ = 2 vibrational state were clearly observed at a collision energy of 0.52 kcal/mol; this was attributed to both the ground and the first excited Feshbach resonances trapped in the peculiar HF(v′ = 3)-H′ vibrationally adiabatic potential, with substantial enhancement by constructive interference between the two resonances.

  • Over 13% Efficient Organic Solar Cells Based on Low-Cost Pentacyclic A-DA D-A-Type Nonfullerene Acceptor

    • Recent studies have almost focused on finding active layer materials with extended pi-conjugation structures for high-performance organic solar cells (OSCs). However, with the extension of conjugate length, the synthesis difficulty and cost of materials will increase. Achieving high efficiency while reducing material costs is a prerequisite for the commercialization of OSCs. Herein, two low-cost A-DA D-A-type (where A and D represent an electron-withdrawing unit and an electron-donating unit, respectively) nonfullerene acceptors (Y25,Y26) are synthesized with pentacyclic fused backbone as the DA D electron-deficient core and 5,6-difluoro-3-(dicyandiamethyl) indigo as the end groups. Compared with classical Y series acceptors with heptacyclic backbone, although Y25 and Y26 own the reduced conjugated length, they still show moderate performance (11.65% and 13.34%), and the cost of synthesis is significantly reduced. Therefore, we provide a new molecular design idea for commercially efficient nonfullerene OSCs acceptors. We also find that adding alkyl chains to the beta site of thiophenes is beneficial to obtaining the reduced energetic disorder, dominant molecular stacking, and desirable morphology, which can facilitate charge carrier transport and prompt higher short-circuit current density (J(sc)) as well as fill factor.

    • Increased expression of adhesion molecules by activated endothelium is a critical feature of vascular inflammation associated with the several diseases such as endotoxin shock and sepsis/septic shock. Our data demonstrated complement regulatory protein C1 inhibitor (C1INH) prevents endothelial cell injury. We hypothesized that C1INH has the ability of an anti-endothelial activation associated with suppression of expression of adhesion molecule(s). C1INH blocked leukocyte adhesion to endothelial cell monolayer in both static assay and flow conditions. In inflammatory condition, C1INH reduced vascular cell adhesion molecule (VCAM-1) expression associated with its cytoplasmic mRNA destabilization and nuclear transcription level. Studies exploring the underlying mechanism of C1INH-mediated suppression in VCAM-1 expression were related to reduction of NF-kappaB activation and nuclear translocation in an IkappaBalpha-dependent manner. The inhibitory effects were associated with reduction of inhibitor IkappaB kinase activity and stabilization of the NF-kappaB inhibitor IkappaB. These findings indicate a novel role for C1INH in inhibition of vascular endothelial activation. These observations could provide the basis for new therapeutic application of C1INH to target inflammatory processes in different pathologic situations.
