    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

  • Identification of Novel Loci and New Risk Variant in Known Loci for Colorectal Cancer Risk in East Asians

    • Background: Risk variants identified so far for colorectal cancer explain only a small proportion of milial risk of this cancer, particularly in Asians.Methods: We performed a genome-wide association study (GWAS) of colorectal cancer in East Asians, cluding 23,572 colorectal cancer cases and 48,700 controls. To identify novel risk loci, we selected 60 omising risk variants for replication using data from 58,131 colorectal cancer cases and 67,347 controls European descent. To identify additional risk variants in known colorectal cancer loci, we performed nditional analyses in East Asians.Results: An indel variant, rs67052019 at 1p13.3, was found to be associated with colorectal cancer risk P = 3.9 x 10(-8) in Asians (OR per allele deletion = 1.13, 95% confidence interval = 1.08-1.18). This sociation was replicated in European descendants using a variant (rs2938616) in complete linkage sequilibrium with rs67052019 (P = 7.7 x 10(-3)). Of the remaining 59 variants, 12 showed an association P < 0.05 in the European-ancestry study, including rs11108175 and rs9634162 at P < 5 x 10(-8) and o variants with an association near the genome-wide significance level (rs60911071, P = 5.8 x 10(-8); 62558833, P = 7.5 x 10(-8)) in the combined analyses of Asian- and European-ancestry data. In addition, ing data from East Asians, we identified 13 new risk variants at 11 loci reported from previous GWAS.Conclusions: In this large GWAS, we identified three novel risk loci and two highly suggestive loci for lorectal cancer risk and provided evidence for potential roles of multiple genes and pathways in the iology of colorectal cancer. In addition, we showed that additional risk variants exist in many colorectal ncer risk loci identified previously.Impact: Our study provides novel data to improve the understanding of the genetic basis for colorectal ncer risk.

  • Genome-wide association studies in East Asians identify new loci for waist-hip ratio and waist circumference

    • Sixty genetic loci associated with abdominal obesity, measured by waist circumference (WC) and waist-hip ratio (WHR), have been previously identified, primarily from studies conducted in Europeanancestry populations. We conducted a meta-analysis of associations of abdominal obesity with approximately 2.5 million single nucleotide polymorphisms (SNPs) among 53,052 (for WC) and 48,312 (for WHR) individuals of Asian descent, and replicated 33 selected SNPs among 3,762 to 17,110 additional individuals. We identified four novel loci near the EFEMP1, ADAMTSL3, CNPY2, and GNAS genes that were associated with WC after adjustment for body mass index (BMI); two loci near the NID2 and HLA-DRB5 genes associated with WHR after adjustment for BMI, and three loci near the CEP120, TSC22D2, and SLC22A2 genes associated with WC without adjustment for BMI. Functional enrichment analyses revealed enrichment of corticotropin-releasing hormone signaling, GNRH signaling, and/or CDK5 signaling pathways for those newly-identified loci. Our study provides additional insight on genetic contribution to abdominal obesity.

  • Lead-Free Cs2BiAgBr6 Double Perovskite-Based Humidity Sensor with Superfast Recovery Time

    • Lead halide perovskites have demonstrated outstanding achievements in photoelectric applications owing to their unique properties. However, the moisture sensitivity of lead halide perovskite has rarely been developed into an applicable humidity sensor due to the intrinsic instability and toxicity issue. Herein, as a highly stable lead-free perovskite, a Cs2BiAgBr6 thin film is chosen to be the active material for humidity sensor due to its extraordinary humidity-dependent electrical properties and good stability. This Cs2BiAgBr6 thin film humidity sensor demonstrates a superfast response time (1.78 s) and recovery time (0.45 s). The superfast response and recovery properties can be attributed to the reversible physisorption of water molecules, which can be easily adsorbed onto or desorbed from the thin film surface. Moreover, the sensor also shows an excellent reliability and stability properties as well as logarithmic linearity in a relative humidity's range of 15% to 78%. The lead-free Cs2BiAgBr6 perovskite possesses great potential for application in real-time humidity sensing.

  • Decompression-Induced Diamond Formation from Graphite Sheared under Pressure

    • Graphite is known to transform into diamond under dynamic compression or under combined high pressure and high temperature, either by a concerted mechanism or by a nucleation mechanism. However, these mechanisms fail to explain the recently reported discovery of diamond formation during ambient temperature compression combined with shear stress. Here we report a new transition pathway for graphite to diamond under compression combined with shear, based on results from both theoretical simulations and advanced experiments. In contrast to the known model for thermally activated diamond formation under pressure, the shear-induced diamond formation takes place during the decompression process via structural transitions. At a high pressure with large shear, graphite transforms into ultrastrong sp3 phases whose structures depend on the degree of shear stress. These metastable sp3 phases transform into either diamond or graphite upon decompression. Our results explain several recent experimental observations of low-temperature diamond formation. They also emphasize the importance of shear stress for diamond formation, providing new insight into the graphite-diamond transformation mechanism.

    • The polymerization of three typical aromatic solvent-doped fullerene materials with similar hexagonal closest packed (hcp) structures (mesitylene/C60, m-dichlorobenzene/C60 and m-xylene/C60 solvates) is studied under high pressure and high temperature (HPHT, 1.5 GPa, 573 K and 2 GPa, 700 K, respectively). Raman and photoluminescence spectroscopies reveal that the intercalated aromatic solvents play a crucial role in tailoring the extent of polymerization of C60 molecules. In the solvates, the solvents confine formation of covalent bonds between C60 molecules to the 001 direction and the (001) plane of the hcp lattices, leading to the formation of mixed polymeric phases of monomers, dimers, one-dimensional (1D) chainlike oligomers, and two-dimensional (2D) tetragonal phase polymers under suitable HPHT conditions. The type and number of substituent groups of the aromatic solvents are found to have significant influence, determining the amounts and types of polymeric phases formed. Our studies enrich the understanding of the formation mechanisms for controllably fabricating polymeric fullerenes and facilitate targeted design and synthesis of unique fullerene-based carbon materials.

  • Energy consumption prediction and optimization of industrial robots based on LSTM

    • Due to wide distribution and low energy efficiency, the energy-saving of industrial robots draws more and more attention, and a large number of methods have emerged to predict or optimize the energy consumption (EC) of robots. However, many dynamic and electrical parameters are unavailable due to the commercial limitations of industrial robots, which constrains the application of those model-based methods. Therefore, this paper proposes a data-driven method for the prediction and optimization of robot EC. Initially, the cause-and-effect relationship between robot EC and joint motion variables, such as the joint position, velocity, and acceleration, is qualitatively analyzed based on the influence of the capacitive and inductive components in the drive system. And a deep neural network based on long short-term memory (LSTM) is proposed to reveal the nonlinear mapping between the industrial robot EC and the joint motion variables, which can predict EC without the parameters of the industrial robot. Based on the proposed neural network, the adaptive genetic algorithm is adopted to optimize the time-variant scaling function, which can optimize the scaled trajectory to reduce EC without hardware modification. To validate the accuracy and efficacy of the proposed method, experiments are conducted on a KUKA KR60-3 six degree-of-freedom (DOF) industrial robot. The results demonstrate that the proposed neural network can predict EC with a mean absolute percentage error less than 4.21% and the proposed method reduces the EC by 22.35%.

  • Theory of photoinduced ultrafast switching to a spin-orbital ordered hidden phase

    • Photo-induced hidden phases are often observed in materials with intertwined orders. Understanding the formation of these non-thermal phases is challenging and requires a resolution of the cooperative interplay between different orders on the ultra-short timescale. In this work, we demonstrate that non-equilibrium photo-excitations can induce a state with spin-orbital orders entirely different from the equilibrium state in the three-quarter-filled two-band Hubbard model. We identify a general mechanism governing the transition to the hidden state, which relies on a non-thermal partial melting of the intertwined orders mediated by photoinduced charge excitations in the presence of strong spin-orbital exchange interactions. Our study theoretically confirms the crucial role played by orbital degrees of freedom in the light-induced dynamics of strongly correlated materials and it shows that the switching to hidden states can be controlled already on the fs timescale of the electron dynamics.

    • Background: Tendons are a crucial component of the musculoskeletal system and responsible for transmission forces derived from muscle to bone. Patients with tendon injuries are often observed with decreased collagen production and matrix degeneration, and healing of tendon injuries remains a challenge as a result of limited understanding of tendon biology. Recent studies highlight the contribution of miR-378a on the regulation gene expression during tendon differentiation. Methods: We examined the tendon microstructure and tendon repair with using miR-378a knock-in transgenic mice, and the tendon-derived stem cells were also isolated from transgenic mice to study their tenogenic differentiation ability. Meanwhile, the expression levels of tenogenic markers were also examined in mouse tendon-derived stem cells transfected with miR-378a mimics during tenogenic differentiation. With using online prediction software and luciferase reporter assay, the binding target of miR-378a was also studied. Results: Our results indicated miR-378a impairs tenogenic differentiation and tendon repair by inhibition collagen and extracellular matrix production both in vitro and in vivo. We also demonstrated that miR-378a exert its inhibitory role during tenogenic differentiation through binding at TGFβ2 by luciferase reporter assay and western blot. Conclusions: Our investigation suggests that miR-378a could be considered as a new potential biomarker for tendon injury diagnosis or drug target for a possible therapeutic approach in future clinical practice.

  • Ultrahard bulk amorphous carbon from collapsed fullerene

    • Amorphous materials inherit short- and medium-range order from the corresponding crystal and thus preserve some of its properties while still exhibiting novel properties1,2. Due to its important applications in technology, amorphous carbon with sp2 or mixed sp2–sp3 hybridization has been explored and prepared3,4, but synthesis of bulk amorphous carbon with sp3 concentration close to 100% remains a challenge. Such materials inherit the short-/medium-range order of diamond and should also inherit its superior properties5. Here, we successfully synthesized millimetre-sized samples—with volumes 103–104 times as large as produced in earlier studies—of transparent, nearly pure sp3 amorphous carbon by heating fullerenes at pressures close to the cage collapse boundary. The material synthesized consists of many randomly oriented clusters with diamond-like short-/medium-range order and possesses the highest hardness (101.9 ± 2.3 GPa), elastic modulus (1,182 ± 40 GPa) and thermal conductivity (26.0 ± 1.3 W m−1 K−1) observed in any known amorphous material. It also exhibits optical bandgaps tunable from 1.85 eV to 2.79 eV. These discoveries contribute to our knowledge about advanced amorphous materials and the synthesis of bulk amorphous materials by high-pressure and high-temperature techniques and may enable new applications for amorphous solids.
