  • Epigenetic–smoking interaction reveals histologically heterogeneous effects of TRIM27 DNA methylation on overall survival among early-stage NSCLC patients

    • Tripartite motif containing 27 (TRIM27) is highly expressed in lung cancer, including non-small-cell lung cancer (NSCLC). Here, we profiled DNA methylation of lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC) tumours from 613 early-stage NSCLC patients and evaluated associations between CpG methylation of TRIM27 and overall survival. Significant CpG probes were confirmed in 617 samples from The Cancer Genome Atlas. The methylation of the CpG probe cg05293407TRIM27 was significantly associated with overall survival in patients with LUSC (HR = 1.65, 95% CI: 1.30–2.09, P = 4.52 × 10−5), but not in patients with LUAD (HR = 1.08, 95% CI: 0.87–1.33, P = 0.493). As incidence of LUSC is associated with higher smoking intensity compared to LUAD, we investigated whether smoking intensity impacted on the prognostic effect of cg05293407TRIM27 methylation in NSCLC. LUSC patients had a higher average pack-year of smoking (37.49LUAD vs 54.79LUSC, P = 1.03 × 10−19) and included a higher proportion of current smokers than LUAD patients (28.24%LUAD vs 34.09%LUSC, P = 0.037). cg05293407TRIM27 was significantly associated with overall survival only in NSCLC patients with medium–high pack-year of smoking (HR = 1.58, 95% CI: 1.26–1.96, P = 5.25 × 10−5). We conclude that cg05293407TRIM27 methylation is a potential predictor of LUSC prognosis, and smoking intensity may impact on its prognostic value across the various types of NSCLC.

  • Expression of the Shrimp wap gene in Drosophila elicits defense responses and protease inhibitory activity

    • The wap gene encodes a single whey acidic protein (WAP) domain-containing peptide from Chinese white shrimp (Fenneropenaeus chinensis), which shows broad-spectrum antimicrobial activities and proteinase inhibitory activities in vitro. To explore the medical applications of the WAP peptide, a wap gene transgenic Drosophila melanogaster was constructed. In wap-expressing flies, high expression levels of wap gene (> 100 times) were achieved, in contrast to those of control flies, by qRT-PCR analysis. The wap gene expression was associated with increased resistance to microbial infection and decreased bacterial numbers in the flies. In addition, the WAP protein extract from wap-expressing flies, compared with control protein extract from control flies, showed improved antimicrobial activities against broad Gram-positive and Gram-negative bacteria, including the clinical drug resistant bacterium of methicillin-resistant S. aureus (MRSA), improved protease inhibitor activities against crude proteinases and commercial proteinases, including elastase, subtilis proteinase A, and proteinase K in vitro, and improved growth rate and microbial resistance, as well as wound-healing in loach and mouse models. These results suggest that wap-expressing flies could be used as a food additive in aquaculture to prevent infections and a potential antibacterial for fighting drug-resistant bacteria.

  • Local Excitation of Whistler Mode Waves and Associated Langmuir Waves at Dayside Reconnection Regions

    • In the Earth's dayside reconnection boundary layer, whistler mode waves coincide with magnetic field openings and the formation of the resultant anisotropic electrons. Depending on the energy range of anisotropic electrons, whistlers can grow at frequencies in the upper and/or lower band. Observations show that whistler mode waves modulate Langmuir wave amplitude as they propagate toward the X line. Observations of whistler mode wave phase and Langmuir waves packets, as well as coincident electron measurements, reveal that whistler mode waves can accelerate electrons via Landau resonance at locations where E(parallel to)is antiparallel to the wave propagation direction. The accelerated electrons produce localized beams, which subsequently drive the periodically modulated Langmuir waves. The close association of those two wave modes reveals the microscale electron dynamics in the exhaust region, and the proposed mechanism could potentially be applied to explain the modulation events observed in planetary magnetospheres and in the solar wind. Plain Language Summary The Sun's and Earth's magnetic field can merge and reconnect on dayside magnetopause. Using measurements from NASA's MMS spacecraft, we report that a class of electromagnetic wave, named whistler mode wave, coincides with the reconnected magnetic field lines. Besides, those whistlers are observed to modulate the electric field oscillations, known as Langmuir waves. Using high-resolution wave and particle measurements, we explain that the whistlers are locally excited when electrons from both sides of the magnetopause mix and form an unstable distribution. The modulated Langmuir waves are generated due to localized electron acceleration, which occurs when the velocity of electrons matches that of whistlers in the direction along the magnetic field. The whistler mode waves and associated Langmuir waves can be used as an additional tool to remotely sense the occurrence of magnetic reconnections.

    • Humans rely increasingly on sensors to address grand challenges and to improve quality of life in the era of digitalization and big data. For ubiquitous sensing, flexible sensors are developed to overcome the limitations of conventional rigid counterparts. Despite rapid advancement in bench-side research over the last decade, the market adoption of flexible sensors remains limited. To ease and to expedite their deployment, here, we identify bottlenecks hindering the maturation of flexible sensors and propose promising solutions. We first analyze challenges in achieving satisfactory sensing performance for real-world applications and then summarize issues in compatible sensor-biology interfaces, followed by brief discussions on powering and connecting sensor networks. Issues en route to commercialization and for sustainable growth of the sector are also analyzed, highlighting environmental concerns and emphasizing nontechnical issues such as business, regulatory, and ethical considerations. Additionally, we look at future intelligent flexible sensors. In proposing a comprehensive roadmap, we hope to steer research efforts towards common goals and to guide coordinated development strategies from disparate communities. Through such collaborative efforts, scientific breakthroughs can be made sooner and capitalized for the betterment of humanity.

  • A Large-Scale Genome-Wide Gene-Gene Interaction Study of Lung Cancer Susceptibility in Europeans With a Trans-Ethnic Validation in Asians

    • Introduction: Although genome-wide association studies have been conducted to investigate genetic variation of lung tumorigenesis, little is known about gene-gene (G × G) interactions that may influence the risk of non-small cell lung cancer (NSCLC).Methods: Leveraging a total of 445,221 European-descent participants from the International Lung Cancer Consortium OncoArray project, Transdisciplinary Research in Cancer of the Lung and UK Biobank, we performed a large-scale genome-wide G × G interaction study on European NSCLC risk by a series of analyses. First, we used BiForce to evaluate and rank more than 58 billion G × G interactions from 340,958 single-nucleotide polymorphisms (SNPs). Then, the top interactions were further tested by demographically adjusted logistic regression models. Finally, we used the selected interactions to build lung cancer screening models of NSCLC, separately, for never and ever smokers.Results: With the Bonferroni correction, we identified eight statistically significant pairs of SNPs, which predominantly appeared in the 6p21.32 and 5p15.33 regions (e.g., rs521828C6orf10 and rs204999PRRT1, ORinteraction = 1.17, p = 6.57 × 10−13; rs3135369BTNL2 and rs2858859HLA-DQA1, ORinteraction = 1.17, p = 2.43 × 10−13; rs2858859HLA-DQA1 and rs9275572HLA-DQA2, ORinteraction = 1.15, p = 2.84 × 10−13; rs2853668TERT and rs62329694CLPTM1L, ORinteraction = 0.73, p = 2.70 × 10−13). Notably, even with much genetic heterogeneity across ethnicities, three pairs of SNPs in the 6p21.32 region identified from the European-ancestry population remained significant among an Asian population from the Nanjing Medical University Global Screening Array project (rs521828C6orf10 and rs204999PRRT1, ORinteraction = 1.13, p = 0.008; rs3135369BTNL2 and rs2858859HLA-DQA1, ORinteraction = 1.11, p = 5.23 × 10−4; rs3135369BTNL2 and rs9271300HLA-DQA1, ORinteraction = 0.89, p = 0.006). The interaction-empowered polygenetic risk score that integrated classical polygenetic risk score and G × G information score was remarkable in lung cancer risk stratification.Conclusions: Important G × G interactions were identified and enriched in the 5p15.33 and 6p21.32 regions, which may enhance lung cancer screening models.

  • Dynamic Channel Modeling for Indoor Millimeter-Wave Propagation Channels Based on Measurements

    • In this contribution, a recently conducted measurement campaign for indoor millimeter-wave (mm-wave) propagation channels is introduced. A vector network analyzer (VNA)-based channel sounder was exploited to record the channel characteristics at the frequency band from 28-30 GHz. A virtual uniform circular array (UCA) with a radius of 0.25 m was formed using a rotator with 360 steps. Moreover, by taking advantage of fiber-optic technique applied in the channel sounder, measurements at 50 positions were performed from an indoor hall to an indoor corridor along a long pre-defined route. A low-complexity high-resolution propagation estimation (HRPE) algorithm is exploited to estimate the propagation parameters of multipath components (MPCs). Based on the HRPE estimation results, a novel clustering identification and tracking algorithm is proposed to trace clusters. Composite channel characteristics, cluster-level characteristics and dynamic (or birth-death) behaviours of the clusters are investigated, which constitute a dynamic model for the indoor mm-wave channel.

    • Millimeter-Wave (mmWave) massive multiple-input multiple-output (MIMO) has been considered as a key enabler for the fifth-generation (5G) communications. It is essential to design and test mmWave 5G devices under various realisticscenarios since the radio propagation channels pose intrinsic limitations on the performance. This requires emulating realistic dynamic mmWave channels in a reproducible manner in laboratories, which is the goal of this paper. In this contribution, we first illustrate the dominant-cluster(s) concept, where thenon-dominant clusters in the mmWave channels are pruned, for mmWave 5G devices applying massive MIMO beamforming. This demonstrates the importance and necessity to accurately emulate the mmWave channels at a cluster level rather than the composite-channel level. Thus, an over-the-air (OTA) emulationstrategy for dynamic mmWave channels is proposed based on the concept of dominant-cluster(s) in a sectored multiprobe anechoic chamber (SMPAC). The key design parameters including the probe number and the angular spacing of probes are investigated through comprehensive simulations. A cost-effective switch circuit is also designed for this purpose and validated in the simulation. Furthermore, a dynamic mmWave channel measured in an indoor scenario at 28-30 GHz is presented, where the proposed emulation strategy is also validated by reproducing the measured reality.

  • Structural Deformation of Sm@C88under High Pressure

    • We have studied the structural transformation of Sm@C88 under pressure up to 18 GPa by infraredspectroscopy combined with theoretical simulations. The infrared-active vibrational modes of Sm@C88 at ambient conditions have been assigned for the first time. Pressure-induced blue and red shiftsof the corresponding vibrational modes indicate an anisotropic deformation of the carbon cage uponcompression. We propose that the carbon cage changes from ellipsoidal to approximately sphericalaround 7 GPa. A smaller deformation of the carbon bonds in the area close to the Sm atom in thecage suggests that the trapped Sm atom plays a role in minimizing the compression of the adjacentbonds. Pressure induced a significant reduction of the band gap of the crystal. The HOMO-LUMOgap of the Sm@C88 molecule decreases remarkably at 7 GPa as the carbon cage is deformed. Also,compression enhances intermolecular interactions and causes a widening of the energy bands. Botheffects decrease the band gap of the sample. The carbon cage deforms significantly above 7 GPa,from spherical to a peanut-like shape and collapses at 18 GPa.

    • Stretchable electronics and soft robotics have shown unsurpassed features, inheriting remarkable functions from stretchable and soft materials. Electrically conductive and mechanically stretchable materials based on composites have been widely studied for stretchable electronics as electrical conductors using various combinations of materials. However, thermally tunable and stretchable materials, which have high potential in soft and stretchable thermal devices as interface or packaging materials, have not been sufficiently studied. Here, a mechanically stretchable and electrically insulating thermal elastomer composite is demonstrated, which can be easily processed for device fabrication. A liquid alloy is embedded as liquid droplet fillers in an elastomer matrix to achieve softness and stretchability. This new elastomer composite is expected useful to enhance thermal response or efficiency of soft and stretchable thermal devices or systems. The thermal elastomer composites demonstrate advantages such as thermal interface and packaging layers with thermal shrink films in transient and steady-state cases and a stretchable temperature sensor.
