    • The level scheme of 157Er has been extended from a spin region where the nucleus behaves as a prolate rotor to a region where the spin is produced by the alignment of all or most of the available valence nucleons along the symmetry axis of a weakly deformed oblate shape. The level scheme was established at high spin using up to four-fold gamma -ray coincidences detected in the Eurogam spectrometer following the reaction 114Cd( 48Ca,5n)157Er at a bombarding energy of 210 MeV. Particularly favoured states have been established at IK=69/2+, 81/2+, 71/2+, 77/2-, 87-/2 and 89-/2. Specific single-particle configurations are assigned to these special states by comparison with cranked Nilsson-Strutinsky calculations. These states are related to structures observed in the neighbouring nuclei 158Er and 157Ho. These data provide the spectrum of single-particle states for the lowest lying valence orbitals above the 146Gd closed core.

  • High-spin studies of the neutron deficient nuclei In-103, In-105, In-107, and In-109

    • High-spin states of the isotopes 103,105,107,109In have been investigated using in-beam γ-ray spectroscopic methods. Results from three different experiments are presented. Targets of 54Fe, 50Cr, and 92Mo were bombarded by a 270 and 261 MeV 58Ni beam and by a 95 MeV 19F beam, respectively. Reaction channel separation was achieved with a charged-particle detector array and in the first two experiments also with a 1π neutron detector system. As a result of these experiments the level schemes of 103,105,107,109In were significantly extended. Excited states of these odd-A indium isotopes are discussed within the framework of the nuclear shell model and the hole-core coupling scheme. The systematics of excited states of light odd-A indium isotopes is also discussed.

    • No contemporary sources mention Erik Jedvardsson, Sweden's king saint. The only account of his life is the saint's legend, in its preserved form written in the late 13th century, and legends are notoriously untrustworthy. It says that in 1160, in the tenth year of Erik's reign, he was killed by a throne claimant. His remains have rested in a reliquary in Uppsala Cathedral since 1257 at the latest and survived the Reformation. A thorough investigation was made in 1946, and the development of new methods motivated a new investigation in 2014. 23 bones remain that apparently belong to the same individual. (They are accompanied in the reliquary by an unrelated shinbone.) Radiocarbon values are consistent with a death in 1160. The bones belong to a man, 35-40 years old, about 171 cm tall, without any discernible medical conditions. Bone density indicates a life of good nourishment and abundant exercise. The skull has one or two healed wounds that may have been due to weapons. Isotope analysis points to a diet rich in freshwater fish. Stable isotopes also imply that he did not spend his last decade in the expected Uppsala area but rather in Västergötland further south. Insufficient reference materials however make this a very preliminary conclusion. Samples for DNA analysis were collected, but the results are not expected for another year. The saint's legend says that in the king's final battle, the enemy swarmed him, and when he fell to the ground they gave him wound after wound until he lay half dead. They then taunted him and finally cut off his head. The remaining bones have at least nine cuts inflicted in connection with death, seven of them on the legs. No wounds have been found on the ribs or the remaining arm bone, which probably means that the king wore a hauberk but had less protected legs. Both shin bones have cuts inflicted from the direction of the feet, indicating that the victim lay on his front. A neck vertebra has been cut through, which could not have been done without removing the hau berk, i.e. not during battle. This confirms that there was an interlude, as described by the taunting in the legend, between battle and decapitation. At no point do the documented wounds gainsay the account of the fight given by the much later legend.

  • Development of a D-xylose fermenting and inhibitor tolerant industrial Saccharomyces cerevisiae strain with high performance in lignocellulose hydrolysates using metabolic and evolutionary engineering

    • Background: The production of bioethanol from lignocellulose hydrolysates requires a robust, D-xylose-fermenting and inhibitor-tolerant microorganism as catalyst. The purpose of the present work was to develop such a strain from a prime industrial yeast strain, Ethanol Red, used for bioethanol production. Results: An expression cassette containing 13 genes including Clostridium phytofermentans XylA, encoding D-xylose isomerase (XI), and enzymes of the pentose phosphate pathway was inserted in two copies in the genome of Ethanol Red. Subsequent EMS mutagenesis, genome shuffling and selection in D-xylose-enriched lignocellulose hydrolysate, followed by multiple rounds of evolutionary engineering in complex medium with D-xylose, gradually established efficient D-xylose fermentation. The best-performing strain, GS1.11-26, showed a maximum specific D-xylose consumption rate of 1.1 g/g DW/h in synthetic medium, with complete attenuation of 35 g/L D-xylose in about 17 h. In separate hydrolysis and fermentation of lignocellulose hydrolysates of Arundo donax (giant reed), spruce and a wheat straw/hay mixture, the maximum specific D-xylose consumption rate was 0.36, 0.23 and 1.1 g/g DW inoculum/h, and the final ethanol titer was 4.2, 3.9 and 5.8% (v/v), respectively. In simultaneous saccharification and fermentation of Arundo hydrolysate, GS1.11-26 produced 32% more ethanol than the parent strain Ethanol Red, due to efficient D-xylose utilization. The high D-xylose fermentation capacity was stable after extended growth in glucose. Cell extracts of strain GS1.11-26 displayed 17-fold higher XI activity compared to the parent strain, but overexpression of XI alone was not enough to establish D-xylose fermentation. The high D-xylose consumption rate was due to synergistic interaction between the high XI activity and one or more mutations in the genome. The GS1.11-26 had a partial respiratory defect causing a reduced aerobic growth rate. Conclusions: An industrial yeast strain for bioethanol production with lignocellulose hydrolysates has been developed in the genetic background of a strain widely used for commercial bioethanol production. The strain uses glucose and D-xylose with high consumption rates and partial cofermentation in various lignocellulose hydrolysates with very high ethanol yield. The GS1.11-26 strain shows highly promising potential for further development of an all-round robust yeast strain for efficient fermentation of various lignocellulose hydrolysates.

  • An approach for manganese biomonitoring using a manganese carrier switch in serum from transferrin to citrate at slightly elevated manganese concentration

    • After high-dose-short-term exposure (usually from occupational exposure) and even more under low-dose long term exposure (mainly environmental) manganese (Mn) biomonitoring is still problematic since these exposure scenarios are not necessarily reflected by a significant increase of total Mn in blood or serum. Usually, Mn concentrations of exposed and unexposed persons overlap and individual differentiation is often not possible. In this paper Mn speciation on a large sample size (n=180) was used in order to be able to differentiate between highly Mn-exposed or low or unexposed individuals at low total Mn concentration in serum (Mn(S)). The whole sample set consisted of three subsets from Munich, Emilia Romagna region in Italy and from Sweden. It turned out that also at low total Mn(S) concentrations a change in major Mn carriers in serum takes place from Mn-transferrin (Mn-Tf(S)) towards Mn-citrate (Mn-Cit(S)) with high statistical significance (p < 0.000002). This carrier switch from Mn-Tf(S) to Mn-Cit(S) was observed between Mn(S) concentrations of 1.5 mu g/L to ca. 1.7 mu g/L. Parallel to this carrier change, for sample donors from Munich where serum and cerebrospinal fluid were available, the concentration of Mn beyond neural barriers analysed as Mn in cerebrospinal fluid (Mn(C)) positively correlates to Mn-Cit(S) when Mn(S) concentration was above 1.7 mu g/L. The correlation between Mn-Cit(S) and Mn(C) reflects the facilitated Mn transport through neural barrier by means of Mn-citrate. Regional differences in switch points from Mn-Tf(S) to Mn-Cit(S) were observed for the three sample subsets. It is currently unknown whether these differences are due to differences in location, occupation, health status or other aspects. Based on our results, Mn-Cit(S) determination was considered as a potential means for estimating the Mn load in brain and CSF, i.e., it could be used as a biomarker for Mn beyond neural barrier. For a simpler Mn-Cit(S) determination than size exclusion chromatography inductively coupled plasma mass spectrometry (SEC-ICP-MS), ultrafiltration (UF) of serum samples was tested for suitability, the latter possibly being a preferred choice for routine occupational medicine laboratories. Our results revealed that UF could be an alternative if methodical prerequisites and limitations are carefully considered. These prerequisites were determined to be a thorough cleaning procedure at a minimum Mn(S) concentration >1.5 mu g/L, as at lower concentrations a wide scattering of the measured concentrations in comparison to the standardized SEC-ICP-MS results were observed.
