    • Bone marrow-derived mesenchymal stem cells (MSCs) are dominant seed cell sources for bone regeneration. Bone morphogenetic proteins (BMPs) initiate cartilage and bone formation in a sequential cascade. Vascular endothelial growth factor (VEGF) is an essential coordinator of extracellular matrix remodeling, angiogenesis and bone formation. In the present study, the effects of the vascular endothelial growth factor 165 (VEGF165) and bone morphogenetic protein 2 (BMP2) genes on bone regeneration were investigated by the lentivirus-mediated cotransfection of the two genes into rat bone marrow-derived MSCs. The successful co-expression of the two genes in the MSCs was confirmed using quantitative polymerase chain reaction (qPCR) and western blot analysis. The results of alizarin red and alkaline phosphatase (ALP) staining at 14 days subsequent to transfection showed that the area of staining in cells transfected with BMP2 alone was higher than that in cells transfected with BMP2 and VEGF165 or untransfected control cells, while the BMP2 + VEGF165 group showed significantly more staining than the untransfected control. This indicated that BMP2 alone exhibited a stronger effect in bone regeneration than BMP2 in combination with VEGF165. Similarly, in inducing culture medium, the ALP activity of the BMP2 + VEGF165 group was notably suppressed compared with that of the BMP2 group. The overexpression of VEGF165 inhibited BMP2-induced MSC differentiation and osteogenesis in vitro. Whether or not local VEGF gene therapy is likely to affect bone regeneration in vivo requires further investigation.

    • Knowing the multi-level influences of determinants on medical-service resumptions is of great benefits to the policymaking for medical-service recovery at different levels of study units during the post-COVID-19 pandemic era. This article evaluated the hospital- and city-level resumptions of medical services in mainland China based on the data of location-based service (LBS) requests of mobile devices during the two time periods (December 2019 and from February 21 to March 18, 2020). We selected medical-service capacity, human movement, epidemic severity, and socioeconomic factors as the potential determinants on medical-service resumptions and then explicitly assessed their multi-level explanatory powers and the interactive effects of paired determinants using the geographical detector method. The results indicate that various determinants had different individual explanatory powers and interactive relationships/effects at different levels of medical-service resumptions. The current study provides a novel multi-level insight for assessing work resumption and individual/interactive influences of determinants, and considerable implications for regionalized recovery strategies of medical services.

    • OBJECTIVE: A disturbed energy transfer involving the adenine nucleotide translocator across the inner mitochondrial membrane has been suggested to be one specific pathogenetic mechanism in dilated cardiomyopathy. Pretranslational steady state expression of this protein in dilated cardiomyopathy was investigated. METHODS: Concentrations of adenine nucleotide translocator were quantified by solution hybridisation. The enzyme or protein expressions of citrate synthase, lactate dehydrogenase, and creatine kinase with isozymes were determined. Analysis was performed on specimens from the left and right ventricles from six organ donor hearts, six explanted hearts with dilated cardiomyopathy, two explanted hearts with ischaemic cardiomyopathy, and from papillary muscles from seven patients operated on for mitral regurgitation. RESULTS: The ejection fraction in patients with mitral regurgitation was 50(10)%, significantly higher (p < 0.001) than in patients with dilated cardiomyopathy (23(5))%. In mitral regurgitation and in ischaemic cardiomyopathy left ventricular adenine nucleotide translocator mRNA concentrations did not differ from those in donor hearts. In dilated cardiomyopathy, adenine nucleotide translocator mRNA concentrations were significantly increased (p < 0.001). Upregulation was more pronounced in right ventricular than in left ventricular myocardium (p < 0.01). The lactate dehydrogenase M subunit fraction was increased to a similar degree in dilated cardiomyopathy and in mitral regurgitation (p < 0.05). Citrate synthase activity was significantly decreased only in dilated cardiomyopathy (p < 0.005). On the other hand, the creatine kinase B subunit content was significantly higher in mitral regurgitation than in dilated cardiomyopathy (p < 0.001). CONCLUSIONS: Despite signs of increased anaerobic and depressed oxidative capacities, dilated cardiomyopathy was specifically characterised by pretranslational upregulation of adenine nucleotide translocator.

  • Boron-Rich Molybdenum Boride with Unusual Short-Range Vacancy Ordering, Anisotropic Hardness, and Superconductivity

    • Determination of the structures of materials involving more light elements such as boron-rich compounds is challenging and technically important in understanding their varied compositions and superior functionalities. Here we resolve the long-standing uncertainties in structure and composition about the highest boride (termed MoB4, Mo1-xB3, or MoB3) through the rapid formation of large sized boron-rich molybdenum boride under pressure. Using high-quality single-crystal X-ray diffraction analysis and aberration-corrected scanning transmission electron microscopy, we reveal that boron-rich molybdenum boride with a composition of Mo0.757B3 exhibits P6(3)/mmc symmetry with a partial occupancy of 0.514 in 211 Mo sites (Mol), and direct observations reveal the short-range ordering of cation vacancies in (010) crystal planes. Large anisotropic Young's moduli and Vickers hardness are seen for Mo0.757B3, which may be attributed by its two-dimensional boron distributions. Mo0.757B3 is also found to be superconducting with a transition temperature (T-c) of 2.4 K, which was confirmed by measurements of resistivity and magnetic susceptibility. Theoretical calculations suggest that the partial occupancy of Mo atoms plays a crucial role in the emergence of superconductivity.
