  • What is a “unimodal” cell population? Using statistical tests as criteria for unimodality in automated gating and quality control

    • Many automated gating algorithms for flow cytometry data are based on the concept of unimodal cell populations. However, in this article, we show that criteria previously used to make decisions on unimodality cannot adequately distinguish unimodal from bimodal densities. We show that dip and bandwidth tests for unimodality, taken from the statistics literature, can do this with consistent and low error rates. These tests also have the possibility to adjust the significance level to handle the trade-off between failing to detect a second mode and seeing a second mode when there is none. The differences between the dip and bandwidth tests are elucidated using real data from the FlowCAP I challenge, also guidelines for flow cytometry data preprocessing are given.

  • Family history of cancer as a risk factor for second malignancies after Hodgkin's lymphoma

    • This study estimated the risk of second primary malignancies after Hodgkin's lymphoma (HL) in relation to family history of cancer, age at diagnosis and latency, among 6946 patients treated for HL in Sweden in 1965–1995 identified through the Swedish Cancer Register (SCR). First-degree relatives (FDRs) to the HL patients and their malignancies were then ascertained together with their malignancies through the Multi-Generation Registry and SCR. The HL patient cohort was stratified on the number of FDRs with cancer, and standardised incidence ratios (SIRs) of developing SM were analysed. In the HL cohort, 781 SM were observed 1 year or longer after HL diagnosis. The risk for developing SM increased with the number of FDRs with cancer, SIRs being 2.26, 3.01, and 3.45 with 0, 1, or ≥2 FDRs with cancer, respectively. Hodgkin's lymphoma long-term survivors treated at a young age with a family history of cancer carry an increased risk for developing SM and may represent a subgroup where standardised screening for the most common cancer sites could be offered in a stringent surveillance programme.

  • Resource Decline in Higher Education – Perceptions and Consequences

    • Teaching students is one of the main tasks of LTH, as thousands of students engage on their engineering studies at the faculty every year. The funding for teaching these students is an essential source of income for LTH, but in recent years there has been an increased worry for diminishing financing for lecturing at the faculty.This study aims to investigate how lecturers perceive the development and what practical measures they have taken in their courses to counteract it. Interviews with ten lecturers at the department of Building and Environmental technology were performed. Many different practical strategies for dealing with the situation were documented during the interviews. The strategies are mainly focusing on making the students do more of the work on their own, e.g. working with the course literature. But the approaches to the problem varied greatly, as some lecturers tried to change as little as possible in order to save time, and others who saw a need for restructuring the entire educational program.

    • Robots used in manufacturing today are tailored to their tasks by system integration based on expert knowledge concerning both production and machine control. For upcoming new generations of even more flexible robot solutions, in applications such as dexterous assembly, the robot setup and programming gets even more challenging. Reuse of solutions in terms of parameters, controls, process tuning, and of software modules in general then gets increasingly important. There has been valuable progress within reuse of automation solutions when machines comply with standards and behave according to nominal models. However, more flexible robots with sensor-based manipulation skills and congnitive functions for human inteaction are far too complex to manage, and solutions are rarely reusable since knowledge is either implicit in imperative software or not captured in machine readable form. We propose techniques that build on existing knowledge by converting structured data into an RDF-based knowledge base. By enhancements of industrial control systems and available engineering tools, such knowledge can be gradually extended as part of the interaction during the definition of the robot task.

    • Virtuella karaktärer, det vill säga mer eller mindre människoliknande och interaktiva datorgenererade figurer, har sedan länge förekommit i datorspel. På senare år har de även blivit allt vanligare i digitala lärmiljöer. De uppträder där i form av virtuella instruktörer, coacher, rollspelskaraktärer, lärkompanjoner och avatarer (alter egos) i pedagogiskt material från förskola till högskola likväl som i specialistutbildningar. I bredare pedagogiska sammanhang träffar man på dem i egenskap av till exempel virtuella rådgivare och informatörer. Skälen till att använda virtuella pedagogiska karaktärer kan variera och i Gulz (2004) ges en översikt samt kritisk diskussion av olika möjliga pedagogiska vinster. Veletsianos Scharber och Doering (2008) fördjupar den kritiska diskussionen i en artikel med titeln ”När sex, droger och våld kommer in i klassrummet: Konversation mellan ungdomar och en kvinnlig pedagogisk agent”. Den analys de presenterar av loggade konversationer visar en kraftig diskrepans mellan pedagogisk intention, å ena sidan, och vardaglig praktik där elever interagerar med virtuella pedagogiska karaktärer, å andra sidan. Former av interaktion, kommunikation och språkbruk, som normalt är otillåtna och förekom sparsamt i de aktuella klassrummen, gjorde här ett inträde på bred front. I denna text kommer vi emellertid att titta närmare på pedagogiska möjligheter. Specifikt fokuserar vi på följande fråga: ”Kan virtuella pedagogiska karaktärer vara ett redskap för att erbjuda alternativa kulturella bilder inom områden med starka genusassociationer som till exempel ingenjörsutbildning och datorteknik?”. Innan vi går in på huvudtemat inleder vi med att presentera en begreppslig och temamässig bakgrund till den kommande analysen och diskussionen kring kulturella bilder, rollmodeller och genusproblematik.

  • Therapy using implanted organic bioelectronics

    • Many drugs provide their therapeutic action only at specific sites in the body, but are administered in ways that cause the drug’s spread throughout the organism. This can lead to serious side effects. Local delivery from an implanted device may avoid these issues, especially if the delivery rate can be tuned according to the need of the patient. We turned to electronically and ionically conducting polymers to design a device that could be implanted and used for local electrically controlled delivery of therapeutics. The conducting polymers in our device allow electronic pulses to be transduced into biological signals, in the form of ionic and molecular fluxes, which provide a way of interfacing biology with electronics. Devices based on conducting polymers and polyelectrolytes have been demonstrated in controlled substance delivery to neural tissue, biosensing, and neural recording and stimulation. While providing proof of principle of bioelectronic integration, such demonstrations have been performed in vitro or in anesthetized animals. Here, we demonstrate the efficacy of an implantable organic electronic delivery device for the treatment of neuropathic pain in an animal model. Devices were implanted onto the spinal cord of rats, and 2 days after implantation, local delivery of the inhibitory neurotransmitter g-aminobutyric acid (GABA) was initiated. Highly localized delivery resulted in a significant decrease in pain response with low dosage and no observable side effects. This demonstration of organic bioelectronics-based therapy in awake animals illustrates a viable alternative to existing pain treatments, paving the way for future implantable bioelectronic therapeutics. 2015 © The Authors.

  • No excess long-term mortality in stage I-IIA Hodgkin lymphoma patients treated with ABVD and limited field radiotherapy

    • When treating limited stage classical Hodgkin lymphoma (cHL), balancing treatment efficacy and toxicity is important. Toxicities after extended-field radiotherapy are well documented. Investigators have aimed at reducing toxicity without compromising efficacy, mainly by using combined modality treatment (CMT), i.e. chemotherapy and limited-field radiotherapy. In some clinical trials, radiotherapy has been omitted. We evaluated 364 patients with stage I-IIA cHL treated between 1999 and 2005. Patients were treated with two or four cycles of doxorubicin, bleomycin, vinblastine and dacarbazine (ABVD) according to presence of risk factors, followed by 30 Gy limited-field (reduced compared to involved-field) radiotherapy. After a median follow-up of 16 years for survival, freedom from progression at five and ten years was 93% and overall survival at 5 and 10 years was 98% and 96%, respectively. Only two relapses, out of 27, occurred after more than 5 years. There was no excess mortality compared to the general population. Of the analysed subgroups, only patients with progression within five years showed significant excess mortality. The absence of excess mortality questions the concept of omitting radiotherapy after short-term chemotherapy, a strategy that has been associated with an elevated risk of relapse but not yet with a proven reduced long-term excess mortality.

    • The rapid advancement of artificial intelligence (AI) in education necessitates a shared understanding of its intended purpose and societal implications. This paper underscores the significance of societal perspectives in AI and education, often overshadowed by technological aspects. At the same time, policy guidelines for the integration of AI technology within educational systems are playing a pivotal role in shaping the future of education. What we as society imagine AI and education to be, will in some shape or form lead the development of suggested fixes. The aim is to aid the understanding of why and how visions of learning and education are framed in relation to developments in Educational Technology (EdTech) and their introduction in education. It thereby contributes to the ongoing discussion on the integration of AI in education and its potential societal impacts.
