    • The origins and Pleistocene evolution of plateau landscapes along passive continental margins of the North Atlantic have been debated for more than a century. A key question in this debate concerns whether glacial and periglacial surface processes have substantially eroded plateau areas during late Cenozoic climatic cooling or whether the plateaus have mainly been protected from erosion by cold-based and largely nonerosive ice sheets. Here we investigate the Pleistocene evolution of a prominent plateau landscape in Reinheimen National Park, southern Norway. We estimate erosion rates across the plateau via inverse modeling of 141 new cosmogenic Be-10 and Al-26 measurements in regolith profiles and bedrock. We combine these results with sedimentological analyses of the regolith. In the vicinity of Reinheimen's regolith-covered summits, the combination of uniformly slow erosion (<10m/Myr) and near-parabolic slope geometry suggests long-term equilibrium with the presently active periglacial mass-wasting processes. Outside summit areas, erosion is faster (up to >50m/Myr), possibly due to episodic glacial erosion. Despite some indications of chemical alteration, such as grusic saprolite and small amounts of secondary minerals, the fine regolith comprises low clay/silt ratios and is dominated by primary minerals with no sign of dissolution. Together with our modeled erosion rates, this indicates that the regolith cover formed, and continues to develop, during the cold climate of the Late Pleistocene. Plain Language Summary Plateaus dissected by steep-sided valleys and fjords are common landscape elements within the mountains bordering the North Atlantic. Most of these plateaus have likely experienced millions of years of near-freezing temperatures and were repeatedly covered by ice sheets during recent glacial periods. Yet the imprint of cold-climate erosion processes on the plateau landscape evolution remains poorly understood. Here we investigate the Pleistocene evolution of an extensive Scandinavian plateau landscape in Reinheimen National Park, southern Norway. We measure cosmogenic nuclides within the surficial layers of rock and sediment on the plateau. The concentration of these cosmogenic nuclides reflects the erosion of the plateau landscape and thereby the impact of recent cold-climate surface processes. We find that erosion has influenced the plateaus within the latest glacial cycles. In the vicinity of the highest, sediment-clad summits, the plateau shape is determined by processes related to freezing and thawing of rocks and sediment, while the influence of erosion by glaciers and streams increases further downslope.

  • Dissecting unique and common variance across body and brain health indicators using age prediction

    • Ageing is a heterogeneous multisystem process involving different rates of decline in physiological integrity across biological systems. The current study dissects the unique and common variance across body and brain health indicators and parses inter-individual heterogeneity in the multisystem ageing process. Using machine-learning regression models on the UK Biobank data set (N = 32,593, age range 44.6-82.3, mean age 64.1 years), we first estimated tissue-specific brain age for white and gray matter based on diffusion and T1-weighted magnetic resonance imaging (MRI) data, respectively. Next, bodily health traits, including cardiometabolic, anthropometric, and body composition measures of adipose and muscle tissue from bioimpedance and body MRI, were combined to predict 'body age'. The results showed that the body age model demonstrated comparable age prediction accuracy to models trained solely on brain MRI data. The correlation between body age and brain age predictions was 0.62 for the T1 and 0.64 for the diffusion-based model, indicating a degree of unique variance in brain and bodily ageing processes. Bayesian multilevel modelling carried out to quantify the associations between health traits and predicted age discrepancies showed that higher systolic blood pressure and higher muscle-fat infiltration were related to older-appearing body age compared to brain age. Conversely, higher hand-grip strength and muscle volume were related to a younger-appearing body age. Our findings corroborate the common notion of a close connection between somatic and brain health. However, they also suggest that health traits may differentially influence age predictions beyond what is captured by the brain imaging data, potentially contributing to heterogeneous ageing rates across biological systems and individuals. A 'body age' model trained on health traits demonstrated comparable age prediction accuracy to models trained solely on brain MRI data.     • Background: Second malignant neoplasms and cardiotoxicity are among the most serious and frequent adverse health outcomes experienced by childhood and adolescent cancer survivors (CCSs) and contribute significantly to their increased risk of premature mortality. Owing to differences in health-care systems, language and culture across the continent, Europe has had limited success in establishing multi-country collaborations needed to assemble the numbers of survivors required to clarify the health issues arising after successful cancer treatment. PanCareSurFup (PCSF) is the first pan-European project to evaluate some of the serious long-term health risks faced by survivors. This article sets out the overall rationale, methods and preliminary results of PCSF. Methods: The PCSF consortium pooled data from 13 cancer registries and hospitals in 12 European countries to evaluate subsequent primary malignancies, cardiac disease and late mortality in survivors diagnosed between ages 0 and 20 years. In addition, PCSF integrated radiation dosimetry to sites of second malignancies and to the heart, developed evidence-based guidelines for long-term care and for transition services, and disseminated results to survivors and the public. Results: We identified 115,596 individuals diagnosed with cancer, of whom 83,333 were 5-year survivors and diagnosed from 1940 to 2011. This single data set forms the basis for cohort analyses of subsequent malignancies, cardiac disease and late mortality and case-control studies of subsequent malignancies and cardiac disease in 5-year survivors. Conclusions: PCSF delivered specific estimates of risk and comprehensive guidelines to help survivors and care-givers. The expected benefit is to provide every European CCS with improved access to care and better long-term health.

  • Biotransport of organic pollutants to an inland Alaska Lake by migrating Sockeye Salmon (Oncorhynchus nerka)

    • Persistent organic pollutants such as polychlorinated biphenyls (PCBs) and the pesticide DDT, known to harm wildlife, have been shown to reach pristine Subarctic and Arctic areas by global atmospheric transport. Another transport route for pollutant entry into these ecosystems is provided by migrating salmon. Pollutant transport was studied in a population of sockeye salmon (Oncorhynchus nerka) in the Copper River, Alaska during their 410 km spawning migration. Pollutants accumulated by the salmon during their ocean life stage were not eliminated during migration, but were transported to the spawning lakes and accumulated in the freshwater food web there. The influence of the biotransported pollutants was investigated by comparing pollutant levels and compositions in atmospheric deposition as well as in two different populations of arctic grayling (Thymallus arcticus). One grayling population was in the salmon spawning lake and the other in a nearby lake not hosting anadromous fish, but receiving pollutants only via atmospheric deposition. The grayling in the salmon spawning lake were found to have concentrations of organic pollutants more than two times higher than those of the grayling in the salmon-free lake, and the pollutant composition resembled that found in salmon. Thus, in the studied Alaska river system, biotransport was found to have a far greater influence than atmospheric input on the PCB and DDT levels in lake biota.

    • Objectives: . SLE is an autoimmune disease with increased cardiovascular morbidity and platelet activation. In the general population, increased platelet size predicts platelet reactivity and cardiovascular disease. The aim of this study was to investigate whether platelet size related to platelet activation and cardiovascular disease in SLE.Methods: . Fresh blood samples from SLE patients ( n = 148), healthy volunteers ( n = 79) and disease controls ( n = 40) were analysed for platelet size and activation by flow cytometry, ELISA and cell count. Associations to manifest cardiovascular disease, venous thrombosis and APS were adjusted for traditional cardiovascular risk factors using logistic regression analysis.Results: . SLE patients had decreased platelet size as compared with healthy controls ( P = 0.003). In SLE, decreased platelet size was related to increased platelet activation, in particular microparticle formation ( P < 0.0001, r = -0.46) and release of serotonin from dense granules ( P < 0.001, r = 0.57). SLE patients with aCL had decreased platelet size ( P = 0.02) and aCL decreased platelet size in vitro ( P = 0.007). In contrast to the general population, increased platelet size was not associated with cardiovascular disease. Instead, decreased platelet size was associated with secondary APS, even after adjusting for traditional cardiovascular risk factors ( P = 0.01, odds ratio 3.58).Conclusion: . Platelet size is decreased in SLE patients and associated with microparticle formation and APS. Future studies are needed to determine the underlying mechanism(s) as well as the potential predictive value of small platelets for disease complications in SLE.

    • BackgroundExcess muscle fat is observed in obesity and associated with greater burden of cardiovascular risk factors and higher risk of mortality. Liraglutide reduces total body weight and visceral fat but its effect on muscle fat and adverse muscle composition is unknown.MethodsThis is a pre-specified secondary analysis of a randomized, double-blind, placebo-controlled trial that examined the effects of liraglutide plus a lifestyle intervention on visceral adipose tissue and ectopic fat among adults without diabetes with body mass index >= 30 kg/m2 or >= 27 kg/m2 and metabolic syndrome. Participants were randomly assigned to a once-daily subcutaneous injection of liraglutide (target dose 3.0 mg) or matching placebo for 40 weeks. Body fat distribution and muscle composition was assessed by magnetic resonance imaging at baseline and 40-week follow-up. Muscle composition was described by the combination of thigh muscle fat and muscle volume. Treatment difference (95% confidence intervals [CI]) was calculated by least-square means adjusted for baseline thigh muscle fat. The association between changes in thigh muscle fat and changes in body weight were assessed using Spearman correlation coefficients. The effect of liraglutide versus placebo on adverse muscle composition, denoted by high thigh muscle fat and low thigh muscle volume, was explored.ResultsAmong the 128 participants with follow-up imaging (92.2% women, 36.7% Black), median muscle fat at baseline was 7.8%. The mean percent change in thigh muscle fat over median follow-up of 36 weeks was -2.87% among participants randomized to liraglutide (n = 73) and 0.05% in the placebo group (absolute change: -0.23% vs. 0.01%). The estimated treatment difference adjusted for baseline thigh muscle fat was -0.24% (95% CI, -0.41 to -0.06, P-value 0.009). Longitudinal change in thigh muscle fat was significantly associated with change in body weight in the placebo group but not the liraglutide group. The proportion of participants with adverse muscle composition decreased from 11.0% to 8.2% over follow-up with liraglutide, but there was no change with placebo.ConclusionsIn a cohort of predominantly women with overweight or obesity in the absence of diabetes, once-daily subcutaneous liraglutide was associated with a reduction in thigh muscle fat and adverse muscle composition compared with placebo. The contribution of muscle fat improvement to the cardiometabolic benefits of liraglutide requires further study.
