    • Optical density (OD) is widely used to estimate the density of cells in liquid culture, but cannot be compared between instruments without a standardized calibration protocol and is challenging to relate to actual cell count. We address this with an interlaboratory study comparing three simple, low-cost, and highly accessible OD calibration protocols across 244 laboratories, applied to eight strains of constitutive GFP-expressing E. coli. Based on our results, we recommend calibrating OD to estimated cell count using serial dilution of silica microspheres, which produces highly precise calibration (95.5% of residuals <1.2-fold), is easily assessed for quality control, also assesses instrument effective linear range, and can be combined with fluorescence calibration to obtain units of Molecules of Equivalent Fluorescein (MEFL) per cell, allowing direct comparison and data fusion with flow cytometry measurements: in our study, fluorescence per cell measurements showed only a 1.07-fold mean difference between plate reader and flow cytometry data.

    • A SUMMARY There is an urgent demand for all-polymer organic solar cells (AP-OSCs) to gain higher efficiency. Here, we successfully improve the performance to 16.09% by introducing a small amount of BN-T, a B <- N-type polymer acceptor, into the PM6:PY-IT blend. It has been found that BN-T makes the active layer, based on the PM6:PY-IT:BN-T ternary blend, more crystalline but meanwhile slightly reduces the phase separation, leading to enhancement of both exciton harvesting and charge transport. From a thermodynamic viewpoint, BN-T prefers to reside between PM6 and PY-IT, and the fraction of this fine-tunes the morphology. Besides, a significantly reduced nonradiative energy loss occurs in the ternary blend, along with the coexistence of energy and charge transfer between the two acceptors. The progressive performance facilitated by these improved properties demonstrates that AP-OSCs can possibly comparably efficient with those based on small molecule acceptors, further enhancing the competitiveness of this device type.

    • Respiratory droplets are bioaerosols that originate from the respiratory tract. Knowing their deposition characteristics during exhalation would facilitate the understanding of the source of large respiratory droplets and their importance in the spread of respiratory infectious diseases. In this study, computational fluid dynamics is used to simulate the motion and deposition of droplets released from either trachea or bronchi in a realistic reconstruction of the human respiratory tract. Influences of airflow structures and locations of droplet generation on droplet deposition are studied, and droplets with diameters between 1 and 50 mu m are examined. The deposition of droplets is found to be influenced mainly by the droplet diameter and the flow rate of exhalation. The number of droplets released from the trachea or bronchi that can escape into the environment decreases as the flow rate increases. When the flow rate is low (10 L/min), the critical diameter of droplets generated in the lower respiratory system that can escape into the air is approximately 12 mu m, but this diameter is approximately 5 mu m when the flow rate is medium (30 to 60 L/min) or large (90 L/min). The larynx is the dominant site of deposition for droplets smaller than the critical diameter, while trachea and bronchus are more important locations that account for the deposition of larger droplets. This study indicates that the lower respiratory tract is an important source of fine droplets (<5 mu m) in indoor environments, and larger droplets probably originate from the upper respiratory tract, which needs further investigation.     • A growing body of evidence suggests interplay between the gut microbiota and the pathogenesis of nonalcoholic fatty liver disease (NAFLD). However, the role of the gut microbiome in early detection of NAFLD is unclear. Prospective studies are necessary for identifying reliable, microbiome markers for early NAFLD. We evaluated 2487 individuals in a community-based cohort who were followed up 4.6 years after initial clinical examination and biospecimen sampling. Metagenomic and metabolomic characterizations using stool and serum samples taken at baseline were performed for 90 participants who progressed to NAFLD and 90 controls who remained NAFLD free at the follow-up visit. Cases and controls were matched for gender, age, body mass index (BMI) at baseline and follow-up, and 4-year BMI change. Machine learning models integrating baseline microbial signatures (14 features) correctly classified participants (auROCs of 0.72 to 0.80) based on their NAFLD status and liver fat accumulation at the 4-year follow up, outperforming other prognostic clinical models (auROCs of 0.58 to 0.60). We confirmed the biological relevance of the microbiome features by testing their diagnostic ability in four external NAFLD case-control cohorts examined by biopsy or magnetic resonance spectroscopy, from Asia, Europe, and the United States. Our findings raise the possibility of using gut microbiota for early clinical warning of NAFLD development.

  • Unraveling the formation mechanism of graphitic nitrogen-doping in thermally treated graphene with ammonia

    • Nitrogen-doped graphene (N-graphene) has attractive properties that has been widely studied over the years. However, its possible formation process still remains unclear. Here, we propose a highly feasible formation mechanism of the graphitic-N doing in thermally treated graphene with ammonia by performing ab initio molecular dynamic simulations at experimental conditions. Results show that among the commonly native point defects in graphene, only the single vacancy 5-9 and divacancy 555-777 have the desirable electronic structures to trap N-containing groups and to mediate the subsequent dehydrogenation processes. The local structure of the defective graphene in combining with the thermodynamic and kinetic effect plays a crucial role in dominating the complex atomic rearrangement to form graphitic-N which heals the corresponding defect perfectly. The importance of the symmetry, the localized force field, the interaction of multiple trapped N-containing groups, as well as the catalytic effect of the temporarily formed bridge-N are emphasized, and the predicted doping configuration agrees well with the experimental observation. Hence, the revealed mechanism will be helpful for realizing the targeted synthesis of N-graphene with reduced defects and desired properties.

    • Metallic nanowires with desired properties for molecular integrated circuits (MICs) are especially significant in molectronics, but preparing such wires at a molecular level still remains challenging. Here, we propose, from first principles calculations, experimentally realizable edge-nitrogen-doped graphene nanoribbons (N-GNRs) as promising candidates for nanowires. Our results show that edge N-doping has distinct effects on the electronic structures and transport properties of the armchair GNRs and zigzag GNRs (AGNRs, ZGNRs), due to the formation of pyridazine and pyrazole rings at the edges. The pyridazine rings raise the Fermi level and introduce delocalized energy bands near the Fermi level, resulting in a highly enhanced conductance in N-AGNRs at the stable nonmagnetic ground state. Especially for the family of AGNRs with widths of n = 3p + 2, their semiconducting characteristics are transformed to metallic characteristics via N-doping, and they exhibit perfectly linear current-voltage (I-V) behaviors. Such uniform and excellent features indicate bright application prospects of the N-AGNRs as nanowires and electrodes in molectronics.

  • Aligning nature-based solutions with ecosystem services in the urban century

    • In an increasingly urbanized world, the concepts of ecosystem services and nature-based solutions can help tackle grand challenges. However, ambiguity in their definitions and in the relationship between the two concepts complicates comprehensive research efforts as well as their effective application in policy and planning in urban systems. This paper presents a framework to clarify and explicitly relate the two concepts, enhancing their applicability in the management of urban challenges. Within the framework, addressing urban challenges serves as the starting point for the development and implementation of nature-based solutions. Nature-based solutions alter the flows of ecosystem services that are produced by an ecosystem by altering the performance of the ecosystem or by changing how people engage with the ecosystem. This results both in changes in the target ecosystem services, as well as non-targeted ecosystem services, leading to benefits. Using two illustrative case studies, we show how the framework can be applied to two urban challenges that are expected to increase in intensity in cities across the world: stormwater management and urban heat stress. Moreover, we highlight key research topics that will benefit from more integrated use of nature-based solutions and ecosystem services. The framework helps emphasize co-benefits, and can be used to help make co-benefits and multifunctionality explicit in urban decision-making and planning processes.

  • KIBRA controls exosome secretion via inhibiting the proteasomal degradation of Rab27a

    • Exosomes are nanosized membrane vesicles released from cells after fusion of multivesicular bodies (MVBs) with the plasma membrane (PM) and play important roles in intercellular communication and numerous biological processes. However, the molecular mechanisms regulating exosome secretion remain poorly understood. Here we identify KIBRA as an adaptor-like protein that stabilizes Rab27a, which in turn controls exosome secretion both in vitro and in vivo. Knockdown or overexpression of KIBRA in neuronal and podocyte cell lines leads to a decrease or increase of exosome secretion, respectively, and KIBRA depletion increases MVB size and number. Comparing protein profiles between KIBRA knockout and wild-type mouse brain showed significantly decreased Rab27a, a small GTPase that regulates MVB-PM docking. Rab27a is stabilized by interacting with KIBRA, which prevents ubiquitination and degradation via the ubiquitin-proteasome pathway. In conclusion, we show that KIBRA controls exosome secretion via inhibiting the proteasomal degradation of Rab27a.

    • Postalkoxysilylation with diethoxydimethylsilane has been carried out on the zeolitic lamellar precursors of various topologies such as MWW, FER, CDO and MCM-47 aiming to construct new crystalline structures with expanded pore apertures between the layers. The silylation process and the crystalline and pore structures of the resulting materials have been investigated with the techniques of XRD, IR, 13C and 29Si MAS NMR, ICP, SEM, HRTEM, elemental analyses, and N2 adsorption. In contrast to forming known three-dimensional zeolite structures after direct calcination of the lamellar precursors, the silylation led to new crystalline structures with opener pores, as evidenced by the shift of layer-related diffractions to the lower-angle region in XRD patterns and the enlarged interlayer pores found by HRTEM images. After optimizing the treatment conditions, particularly the amount of silane agent, a maximum and homogeneous silylation was realized, which guaranteed the phase purity in interlayer expanded zeolites. The expanded structures were well preserved after calcination at 823 K or reflux in water for 1 to 2 weeks, indicating a high thermal stability and also a hydrothermal stability. The interlayer expanded zeolites prepared from the metallosilicate precursors of MWW topology exhibited higher catalytic activities in the redox and solid acid-catalyzed reactions of bulky molecules than that of their counterparts with conventional MWW topology.
