    • This work demonstrates a novel method for the synthesis of large pore mesoporous silica nanoparticles (MSNs) with a pore diameter of 10.3 nm and a particle diameter of ∼50 nm based on the incorporation of mixed anionic surfactants sodium dodecyl benzene sulfonate (SDBS) and sodium dodecyl sulphate (SDS) as the template in the synthesis process. The dispersity, morphology, pore structure and size of mesoporous nanoparticles were adjusted by changing the molar ratio of two anionic surfactants, the concentration of the co-structure-directing agent (3-aminopropyltrimethoxysilane) and the reaction temperature. The results of synthesis experiments suggested that the formation of large pore MSNs involved a nucleation and growth process. MSNs were post-grafted with a Schiff base moiety for fluorescence sensing of Fe3+ in water. The applicability of functionalized MSNs was demonstrated by selective fluorescence detection of Fe3+ in aqueous media.

    • This work demonstrated the synthesis of (4E)-4-(4-(diphenylamino)benzylideneamino)-1,2-dihydro-1,5- dimethyl-2-phenylpyrazol-3-one (DBDDP) for Fe (III) detection in aqueous media and in the core of silica cross-linked micellar nanoparticles in living cells. The free DBDDP performed fluorescence enhancement due to Fe (III)-promoted hydrolysis in a mixed aqueous solution, while the DBDDP-doped silica cross-linked micellar nanoparticles (DBDDP-SCMNPs) performed an electron-transfer based fluorescence quenching of Fe (III) in living cells. The quenching fluorescence of DBDDP-SCMNPs and the concentration of Fe (III) exhibited a linear correlation, which was in accordance with the Stern-Volmer equation. Moreover, DBDDP-SCMNPs showed a low limit of detection (LOD) of 0.1 ppm and an excellent selectivity against other metal ions. Due to the good solubility and biocompatibility, DBDDP-SCMNPs could be applied as fluorescence quenching nanosensors in living cells.

  • p21-activated kinase 4 phosphorylation of integrin beta5 Ser-759 and Ser-762 regulates cell migration

    • Modulation of integrin alphavbeta5 regulates vascular permeability, angiogenesis, and tumor dissemination. In addition, we previously found a role for p21-activated kinase 4 (PAK4) in selective regulation of integrin alphavbeta5-mediated cell motility (Zhang, H., Li, Z., Viklund, E. K., and Strömblad, S. (2002) J. Cell Biol. 158, 1287-1297). This report focuses on the molecular mechanisms of this regulation. We here identified a unique PAK4-binding membrane-proximal integrin beta5-SERS-motif involved in controlling cell attachment and migration. We also mapped the integrin beta5-binding site within PAK4. We found that PAK4 binding to integrin beta5 was not sufficient to promote cell migration, but that PAK4 kinase activity was required for PAK4 promotion of cell motility. Importantly, PAK4 specifically phosphorylated the integrin beta5 subunit at Ser-759 and Ser-762 within the beta5-SERS-motif. Point mutation of these two serine residues abolished the PAK4-induced cell migration, indicating a functional role for these phosphorylations in migration. Our results may give important leads to the functional regulation of integrin alphavbeta5, with implications for vascular permeability, angiogenesis, and cancer dissemination.

  • An Extra-Large-Pore Zeolite with 24 x 8 x 8-Ring Channels Using a Structure-Directing Agent Derived from Traditional Chinese Medicine

    • Extra-large-pore zeolites have attracted much interest because of their important applications for processing larger molecules. Although great progress has been made in academic science and industry, it is challenging to synthesize these materials. A new extra-large-pore zeolite SYSU-3 (Sun Yat-sen University no?) has been synthesized by using a novel sophoridine derivative as an organic structure-directing agent (OSDA). The framework structure was solved and refined using continuous rotation electron diffraction (cRED) data from nanosized crystals. SYSU-3 exhibits a new zeolite framework topology, which has the first 24 x 8 x 8-ring extra-large pore system and a framework density (FD) as low as 11.4 T/1000 angstrom(3). The unique skeleton of the OSDA plays an essential role in the formation of the distinctive zeolite structure. This work provides a new perspective for developing new zeolilic materials by using alkaloids as cost-effective OSDAs.
