  • SARS-CoV-2 brainstem encephalitis in human inherited DBR1 deficiency.

    • Inherited deficiency of the RNA lariat-debranching enzyme 1 (DBR1) is a rare etiology of brainstem viral encephalitis. The cellular basis of disease and the range of viral predisposition are unclear. We report inherited DBR1 deficiency in a 14-year-old boy who suffered from isolated SARS-CoV-2 brainstem encephalitis. The patient is homozygous for a previously reported hypomorphic and pathogenic DBR1 variant (I120T). Consistently, DBR1 I120T/I120T fibroblasts from affected individuals from this and another unrelated kindred have similarly low levels of DBR1 protein and high levels of RNA lariats. DBR1 I120T/I120T human pluripotent stem cell (hPSC)-derived hindbrain neurons are highly susceptible to SARS-CoV-2 infection. Exogenous WT DBR1 expression in DBR1 I120T/I120T fibroblasts and hindbrain neurons rescued the RNA lariat accumulation phenotype. Moreover, expression of exogenous RNA lariats, mimicking DBR1 deficiency, increased the susceptibility of WT hindbrain neurons to SARS-CoV-2 infection. Inborn errors of DBR1 impair hindbrain neuron-intrinsic antiviral immunity, predisposing to viral infections of the brainstem, including that by SARS-CoV-2.

    • The frequency of human papilloma virus (HPV) and its influence on clinical outcome was analyzed retrospectively in pre-treatment paraffin embedded biopsies from 110 patients with tongue cancer. The presence of HPV DNA was examined in 85 mobile tongue tumors and 25 base of tongue tumors by a polymerase chain reaction (PCR) with 2 general primer pairs, GP5+/6+ and CPI/IIG. When HPV-DNA was found, HPV-type specific primers and direct sequencing were used for HPV sub-type verification. Twelve of 110 (10.9%) samples were HPV-positive; 9 for HPV-16, 1 for HPV-33, 1 for HPV-35 and 1 could not be analyzed because of shortage of DNA. HPV was significantly more common in base of tongue tumors (10/25, 40.0%) compared to tumors of the mobile tongue (2/85, 2.3%). The influence of HPV on clinical outcome in mobile tongue cancer could not be studied, due to that HPV was present in too few cases. Of the 19 patients with base of tongue cancer that were included in the survival analysis, however, 7 patients with HPV-positive base of tongue cancer had a significantly favorable 5-year survival rate compared to the 12 HPV-negative patients. In conclusion, HPV is significantly more common in base of tongue cancer than in mobile tongue cancer, and has a positive impact on disease-specific survival in patients with base of tongue cancer.

  • Främmande arter invaderar våra sötvatten

    • På senare år har allt fler främmande, invasiva arter börjat breda ut sig i våra sötvatten. En främmande art är en organism som har spridits utanför sitt naturliga utbredningsområde med människans hjälp. Detta kan ha skett avsiktligt eller av misstag. En del av dessa nya arter ställer till problem, andra gör det inte. Här beskrivs övervakningen av några invasiva arter samt hur man kan arbeta för att begränsa dem. Ullhandskrabba, vandrarmussla och sjögull kommer av allt att döma att öka sin utbredning i Sverige de närmaste åren. Det finns idag ett stort antal främmande arter rapporterade från svenska sötvatten och i vår närhet finns ett femtiotal nya arter som kan tänkas etablera sig här. Alla dessa arter kommer inte nödvändigtvis att skapa problem men kunskap om deras biologi och observationer från allmänheten kommer att betyda mycket även i det framtida övervakningsarbetet.

  • Parenting a child with cancer and maintaining a healthy couple relationship : Findings from the Family Talk Intervention

    • Background Despite the challenges that caring for a child with cancer brings for parents, little knowledge is available concerning the effects on the parents’ couple relationship. Furthermore, few interventions have been designed to support parents in their couple relationship. The aim of this paper was, therefore, to explore parents’ experiences of their couple relationship while having a dependent child with cancer and the support they received from a family-based intervention, the Family Talk Intervention (FTI).Methods Data for this paper were taken from semi-structured interviews performed in a pilot study of the FTI in the context of pediatric oncology. In total, 22 couples were interviewed after participating in the FTI. The interviews were transcribed and analyzed using qualitative content analysis.Results Parents described how maintaining a couple relationship while living with childhood cancer could be very challenging and was not given the highest priority. The FTI was considered a way of providing important support to the couple and a chance for them to sit down together and listen to each other's perspectives on the situation. Parents described that the FTI had helped them gain an increased mutual understanding, sometimes also helping them to realize that they needed more extensive professional support in their relationship.Conclusions Living with childhood cancer and upholding a healthy couple relationship is challenging for parents. The FTI has the potential to support couples, mainly by providing opportunities for parents to communicate with each other. However, some couples may be in need of a tailored clinical intervention.

  • Studying the uptake of cell-penetrating peptides

    • More than a decade ago, it was discovered that cationic peptides could traverse the cellular plasma membrane without specific transporter proteins or membrane damage. Subsequently, it was found that these peptides, known as cell-penetrating peptides (CPPs), were also capable of delivering cargos into cells, hence the great potential of these vectors was acknowledged. Today, many different research groups are working with CPPs, which necessitates efforts to develop unified assays enabling the comparison of data. Here we contribute three protocols for evaluation of CPPs which, if used in conjunction, provide complementary data about the amount and mechanism of uptake (fluorometric analysis and confocal microscopy, respectively), as well as the extent of degradation (HPLC analysis of cell lysates). All three protocols are based on the use of fluorescently labeled peptides and can be performed on the same workday.

  • Lower LPS responsiveness in Estonian than Swedish infants associates with less allergy development and high endotoxin exposure

    • Background: Allergic diseases have increased in the last decades, particularly in countries with an affluent lifestyle, possibly due to a reduced or altered microbial exposure during infancy. Objective: The aim of this study was to follow lipopolysaccharide (LPS) induced immune responses prospectively in infants from Estonia and Sweden, i.e. two countries with different frequencies of allergic disease and a different domestic endotoxin exposure. ' Methods: The study included 14 Estonian and 36 Swedish infants who were followed prospectively from birth up to two years of age regarding development of allergy using questionnaires, clinical examinations and skin prick tests. Isolated cord blood mononuclear cells (birth) and peripheral blood mononuclear cells (obtained at 3, 6, 12 and 24 months) were cultured for 24h with LPS in combination with IFN-γ. The secretion of IL-6, CXCL8 (IL-8), IL-10, IL12p70, IL-17, IL-1β, CCL2 (MCP-1), CCL4 (MIP-1β) and TNF was analysed with a Luminex assay. Results: The Swedish, as compared to Estonian children, had higher levels of LPS/IFN-γ induced CCL4 and IL-6 at birth and of IL-1β, IL-12p70, IL-17 and TNF at 3 months as well as IL-6 at 6 months. Also, the levels of CCL2 at 3 and 6 months of age and CCL4 and TNF at 6 months of age were higher in Swedish than Estonian infants in unstimulated cultures. Sensitised Swedish infants had higher levels of LPS/IFN-γ induced IL-10 at 3 and 12 months of age compared to non-sensitised Swedish infants. Conclusion: The enhanced LPS/IFN-γ induced cytokine and chemokine secretion in Swedish, compared to Estonian infants may support a less rapid induction of immune regulation in an affluent society. This could possibly be due to a reduced microbial pressure on Swedish children during early childhood.

  • Exploatering av stränder 2013 - 2018

    • Strandområden är viktiga miljöer för många djur och växter, men de är även attraktiva för bebyggelse och friluftsliv. Det finns behov av att följa trender i strändernas exploateringsgrad för kommuner, län och i hela landet. I denna rapport presenteras en landstäckande kartering av exploatering i strandzonen. Karteringen följer upp exploatering av strandzonen på nationell, länsoch kommunnivå med cirka fem års intervall.Metoden som använts går ut på att använda befintliga geografiska data och lägga samman dessa för att skapa en indikator som ger ett mått på hur stor arealandel av strandzonen som är exploaterad. Motsvarande karteringar har utförts 2013 för kust- och havsöstränder samt för sötvattenstränder, vilket har redovisats av Länsstyrelsen Norrbotten (Engdahl A & Nilsson T, 2014) och Länsstyrelsen Gävleborg (Kellner O, 2016).Karteringen redovisar ytor och statistik för stränder vid två olika kategorier av havsvatten (fastlandskust och havsöar) samt tre olika kategorier av sötvatten (sjöar, breda vattendrag och smala vattendrag). För varje vattenkategori/strandtyp finns tre olika bredder på strandzoner, 30 meter, 100 meter respektive 300 meter.Inom strandzonerna karteras påverkansytor vid byggnader, vägar och järnvägar med hjälp av buffertzoner. Andelen påverkad areal inom de olika strandzonskategorierna har beräknats länsoch kommunvis.Då metoden justerats sedan 2013 vad gäller underlagsdata för strandtyperna så ingår analyser av strandexploatering för både 2018 och 2013 i denna studie. Resultatet av analyserna för 2018 och 2013 visar en liten ökning i strandexploatering. De största exploateringsökningarna återfinns inom fastlandskustzonen.
