  • MTH1 inhibition eradicates cancer by preventing sanitation of the dNTP pool

    • Cancers have dysfunctional redox regulation resulting in reactive oxygen species production, damaging both DNA and free dNTPs. The MTH1 protein sanitizes oxidized dNTP pools to prevent incorporation of damaged bases during DNA replication. Although MTH1 is non-essential in normal cells, we show that cancer cells require MTH1 activity to avoid incorporation of oxidized dNTPs, resulting in DNA damage and cell death. We validate MTH1 as an anticancer target in vivo and describe small molecules TH287 and TH588 as first-in-class nudix hydrolase family inhibitors that potently and selectively engage and inhibit the MTH1 protein in cells. Protein co-crystal structures demonstrate that the inhibitors bindin the active site of MTH1. The inhibitors cause incorporation of oxidized dNTPs in cancer cells, leading to DNA damage, cytotoxicity and therapeutic responses in patient-derived mouse xenografts. This study exemplifies the non-oncogene addiction concept for anticancer treatment and validates MTH1 as being cancer phenotypic lethal.

Därför stödjer vi deras krav, skriver 270 klimatforskare.

  • IgG Endopeptidase in Highly Sensitized Patients Undergoing Transplantation.

    • BACKGROUND: Donor-specific antibodies create an immunologic barrier to transplantation. Current therapies to modify donor-specific antibodies are limited and ineffective in the most highly HLA-sensitized patients. The IgG-degrading enzyme derived from Streptococcus pyogenes (IdeS), an endopeptidase, cleaves human IgG into F(ab')2 and Fc fragments inhibiting complement-dependent cytotoxicity and antibody-dependent cellular cytotoxicity, which suggests that IdeS might be useful for desensitization. We report on the combined experience of two independently performed open-label, phase 1-2 trials (conducted in Sweden and the United States) that assessed the efficacy of IdeS with regard to desensitization and transplantation of a kidney from an HLA-incompatible donor.METHODS: We administered IdeS to 25 highly HLA-sensitized patients (11 patients in Uppsala or Stockholm, Sweden, and 14 in Los Angeles) before the transplantation of a kidney from an HLA-incompatible donor. Frequent monitoring for adverse events, outcomes, donor-specific antibodies, and renal function was performed, as were renal biopsies. Immunosuppression after transplantation consisted of tacrolimus, mycophenolate mofetil, and glucocorticoids. Patients in the U.S. study also received intravenous immune globulin and rituximab after transplantation to prevent antibody rebound.RESULTS: Recipients in the U.S. study had a significantly longer cold ischemia time (the time elapsed between procurement of the organ and transplantation), a significantly higher rate of delayed graft function, and significantly higher levels of class I donor-specific antibodies than those in the Swedish study. A total of 38 serious adverse events occurred in 15 patients (5 events were adjudicated as being possibly related to IdeS). At transplantation, total IgG and HLA antibodies were eliminated. A total of 24 of 25 patients had perfusion of allografts after transplantation. Antibody-mediated rejection occurred in 10 patients (7 patients in the U.S. study and 3 in the Swedish study) at 2 weeks to 5 months after transplantation; all these patients had a response to treatment. One graft loss, mediated by non-HLA IgM and IgA antibodies, occurred.CONCLUSIONS: IdeS reduced or eliminated donor-specific antibodies and permitted HLA-incompatible transplantation in 24 of 25 patients.     • Epilepsi är den vanligaste neurologiska sjukdomen efter stroke och huvudvärk, ca 60 000 personer har aktiv epilepsi i Sverige. Vi har idag tillgång till en rad mycket effektiva läkemedel, bara sedan 1989 har åtta nya epilepsimediciner registrerats. Trots den utvidgade behandlingsarsenalen bli bara två av tre patienter anfallsfria av läkemedel, för en tredjedel fortsätter anfallen att komma. Vid farmakologiskt terapiresistent epilepsi finns andra behandlingsalternativ.

  • An experimental rabbit model for jaw-bone healing.

    • The purpose of this investigation was to study the structural and topographical bone anatomy of the right and left edentulous areas between the incisors and molars in the rabbit maxilla with regard to the symmetry of the bone, and to assess the degree of spontaneous healing of surgical defects. Anatomical and radiographic examinations together with analysis of serial histological ground sections in ten rabbits disclosed no statistically significant differences between the two sides regarding the different bone-tissue structures, i.e. they exhibit a sufficient degree of symmetry to serve as a useful bilateral test-control model. Surgical defects were made on one side of the jaw (test side) in a group of eight rabbits. This resulted in an average loss of 17% of the total bone volume after a healing period of four weeks as compared to the untreated control side. It was concluded that surgically-created defects do not show completely spontaneous healing. From a histological section of the test side, it was possible to redraw the original bone contour by interpolation between unaffected areas of bone, coronal and apical to the defect. This means that the test side of this model can also serve as its own control with regard to the amount of regenerated tissue, given that there is unaffected bone, coronal and apical to the defect.

    • The aim of this experimental investigation was to compare the effect of using autologous particulate bone grafts with and without a bioresorbable barrier covering for augmentation of the rabbit skull bone. For this purpose, bilateral, circular, 8 mm wide and 1 mm deep skull bone defects were prepared and overfilled with particulate bone grafts. The grafts placed in the test sites were covered with a bioresorbable barrier (Guidor Matrix Barrier). The grafts placed in the control sites were covered only by the repositioned, cutaneous flap. 12 weeks later, the animals were sacrificed, the experimental sites were defleshed and the height and volume of the augmented bone in the test and control sites were measured clinically. Histologically, morphometrical measurements of the bone tissue were performed in decalcified vertical cross-sections of the experimental sites. Statistically significant differences were found in favour of the coverage of the bone graft particles with the barrier, both with respect to the height and the volume of the augmented bone.
