  • Consumption of flea beetles (Phyllotreta, Coleoptera: Chrysomelidae) by spiders in field habitats detected by molecular analysis

    • Flea beetles, Phyllotreta Chevrolat (Coleoptera: Chrysomelidae) species, are often found in oilseed rape (OSR), Brassica napus Linnaeus (Brassicaceae). Among predators in the generalist predator complex present in agricultural fields, wolf spiders (Araneae: Lycosidae) are found on the ground and cobweb spiders (Araneae: Theridiidae) build webs in the foliage. We present group-specific primers developed for five flea beetle species within the genus Phyllotreta and study the incidence of predation of flea beetles by these spider groups using DNA-based gut-content analysis. Wolf spiders of the genus Pardosa Koch and the cobweb spider, Phylloneta impressa (Koch), were collected in three winter OSR fields. Flea beetle densities as well as the occurrence of predators and alternative prey were monitored. In total 19.4% of the collected Pardosa tested positive for flea beetle DNA in the polymerase chain reaction analyses, whereas 10% P. impressa were positive. Pardosa were more likely to be positive for flea beetle DNA when Pardosa activity density was low. Phylloneta impressa were more likely to be positive for flea beetle DNA if they were positive for pollen beetle DNA. Implications of these results for conservation biological control and future studies of food webs in OSR are discussed.

    • This article documents the addition of 411 microsatellite marker loci and 15 pairs of Single Nucleotide Polymorphism (SNP) sequencing primers to the Molecular Ecology Resources Database. Loci were developed for the following species: Acanthopagrus schlegeli, Anopheles lesteri, Aspergillus clavatus, Aspergillus flavus, Aspergillus fumigatus, Aspergillus oryzae, Aspergillus terreus, Branchiostoma japonicum, Branchiostoma belcheri, Colias behrii, Coryphopterus personatus, Cynogolssus semilaevis, Cynoglossus semilaevis, Dendrobium officinale, Dendrobium officinale, Dysoxylum malabaricum, Metrioptera roeselii, Myrmeciza exsul, Ochotona thibetana, Neosartorya fischeri, Nothofagus pumilio, Onychodactylus fischeri, Phoenicopterus roseus, Salvia officinalis L., Scylla paramamosain, Silene latifo, Sula sula, and Vulpes vulpes. These loci were cross-tested on the following species: Aspergillus giganteus, Colias pelidne, Colias interior, Colias meadii, Colias eurytheme, Coryphopterus lipernes, Coryphopterus glaucofrenum, Coryphopterus eidolon, Gnatholepis thompsoni, Elacatinus evelynae, Dendrobium loddigesii Dendrobium devonianum, Dysoxylum binectariferum, Nothofagus antarctica, Nothofagus dombeyii, Nothofagus nervosa, Nothofagus obliqua, Sula nebouxii, and Sula variegata. This article also documents the addition of 39 sequencing primer pairs and 15 allele specific primers or probes for Paralithodes camtschaticus.

    • Species in northern Europe re-colonized the region after the last glacial maximum via several routes, which could have lingering signatures in current intraspecific trait variation. The spruce bark beetle, Ips typographus, occurs across Europe, and biological differences have been found between southern and northern Scandinavian populations. However, the postglacial history of I. typographus in Scandinavia has not been previously studied at a fine geographical scale. Therefore, we collected specimens across northern Europe and analysed the genetic variation in a quite large mitochondrial fragment (698 bp). A high genetic diversity was found in some of the most northern populations, in the Baltic States, Gotland and central Europe. Detected genetic and phylogeographic structures suggest that I. typographus re-colonized Scandinavia via two pathways, one from the northeast and one from the south. These findings are consistent with the re-colonization history of its host plant, Picea abies. However, we observed low haplotype and nucleotide diversity in southern Scandinavian populations of I. typographus, indicating that (unlike P. abies) it did not disperse across the Baltic Sea in multiple events. Further, the divergence among Scandinavian populations was shallow, conflicting with a scenario where I. typographus expanded concurrently with its host plant from a 'cryptic refugium' in the northwest.

    • The scarce heath (Coenonympha hero) is an internationally threatened butterfly in Western Europe, where it occurs primarily on hay fields and abandoned arable land in a small-scale agricultural landscape of south-central Scandinavia. Due to afforestation, this habitat is becoming increasingly fragmented in Sweden, and it can be expected that the scarce heath will decline abruptly when threshold conditions for metapopulation persistence are no longer met. We used stepwise polychotomous logistic regression to compare habitat characteristics and isolation measures for patches that harbour large, small or no populations, respectively, in an area of south-western Sweden. We found that patch area, distance to the nearest large population and amount of Galium spp. explained a significant part of the variation in relative abundance among patches. Distance to nearest large population resulted in a better model to predict occupancy than both distance to the nearest inhabited patch and connectivity, which suggests that primarily large populations act as sources for small satellite populations. Today, sites of three of the eight larger populations in the study area have been planted with spruce or pine and will disappear within 20 years. We argue that the disappearance of these patches may very well lead to rapid extinction of the whole metapopulation system.

    • Dispersal between habitat patches may be important for the long-term persistence of populations. We conducted a mark-release-recapture study and analysed the dispersal pattern in the scarce heath butterfly inhabiting a network of suitable habitat patches using stepwise logistic regression (SLR) and the Virtual Migration (VM) model. We also analysed the influence of different types of matrices. We found that the majority of the recaptured butterflies remained within the patch where they were originally caught. However, dispersal between patches did occur and both the SLR analysis and the VM model indicated that the migration pattern was significantly associated with patch area and its level of isolation. The SLR model also showed that there was a positive association between immigration rate and tree density, supporting earlier observations that this species prefers semi-open habitat. We discuss the use of SLR versus the VM model to analyse recapture data in dispersal studies. This system is not at equilibrium, as a number of the most important patches in the network are continuously being lost due to afforestation and a number of populations are facing deterministic extinction. This increases the risk of a chain reaction of local extinctions, which may cause a collapse of the whole system.

    • In this study, we examined the evolutionary outcome of and interplay between historic isolation and current selection pressures on traits more or less closely connected to fitness in the Pearly Heath butterfly (Coenonympha arcania) across its range in Europe. We hypothesized that a trait mean is more related to historic events if it has low connection to fitness, while a trait more closely connected with fitness is expected to have a mean that relates more to current selection pressures. In order to test this, we collected 322 butterflies from across the species range in Europe and measured five wing traits relating to size and color patterns. To infer a phylogeographic history for each individual, we sequenced a 594 bp fragment of the COI gene. The morphological data were then analyzed in relation to selected climatic variables and the history of individuals to disentangle which factors best correlated with morphological variation. The results supported our hypothesis in that wing sizes correlated with summer precipitation but not with its inferred location during the last glaciation. Eyespot position, on the other hand, correlated with the history of individuals but not with the analyzed climatic indicators. The sizes of the black spot and the white band, two traits that were expected to have intermediate selection pressure, were associated with both history and current conditions. Thus, this study illustrates the fascinating interplay between events and processes that lead to a specific evolutionary outcome.

  • Bayesian species delimitation reveals generalist and specialist parasitic wasps on Galerucella beetles (Chrysomelidae) : sorting by herbivore or plant host

    • Background: To understand the ecological and evolutionary consequences of species interactions in food webs necessitates that interactions are properly identified. Genetic analyses suggest that many supposedly generalist parasitoid species should rather be defined as multiple species with a more narrow diet, reducing the probability that such species may mediate indirect interactions such as apparent competition among hosts. Recent studies showed that the parasitoid Asecodes lucens mediate apparent competition between two hosts, Galerucella tenella and G. calmariensis, affecting both interaction strengths and evolutionary feedbacks. The same parasitoid was also recorded from other species in the genus Galerucella, suggesting that similar indirect effects may also occur for other species pairs. Methods: To explore the possibility of such interactions, we sequenced mitochondrial and nuclear genetic markers to resolve the phylogeny of both host and parasitoid and to test the number of parasitoid species involved. We thus collected 139 Galerucella larvae from 8 host plant species and sequenced 31 adult beetle and 108 parasitoid individuals. Results: The analysis of the Galerucella data, that also included sequences from previous studies, verified the five species previously documented as reciprocally monophyletic, but the Bayesian species delimitation for A. lucens suggested 3-4 cryptic taxa with a more specialised host use than previously suggested. The gene data analyzed under the multispecies coalescent model allowed us to reconstruct the species tree phylogeny for both host and parasitoid and we found a fully congruent coevolutionary pattern suggesting that parasitoid speciation followed upon host speciation. Conclusion: Using multilocus sequence data in a Bayesian species delimitation analysis we propose that hymenopteran parasitoids of the genus Asecodes that infest Galerucella larvae constitute at least three species with narrow diet breath. The evolution of parasitoid Asecodes and host Galerucella show a fully congruent coevolutionary pattern. This finding strengthens the hypothesis that the parasitoid in host search uses cues of the host rather than more general cues of both host and plant.

  • A clue to invasion success: genetic diversity quickly rebounds after introduction bottlenecks

    • One of the fundamental questions in invasion biology is to understand the genetic mechanisms behind success or failure during the establishment of a species. However, major limitations to understanding are usually a lack of spatiotemporal population data and information on the populations' colonisation history. In a large-scale, detailed study on the bush-cricket Metrioptera roeselii 70 groups of founders were introduced in areas outside the species' distribution range. We examined how (1) the number of founders (2-32 individuals), (2) the time since establishment (7 or 15 years after introduction) and (3) possible gene flow affected establishment success and temporal genetic changes of the introduced populations. We found higher establishment success in introductions with larger propagule sizes but genetic diversity indices were only partly correlated to propagule size. As expected, introduced populations were more similar to their founder population the larger the propagule size was. However, even if apparent at first, most of the differentiation in the small propagule introductions disappeared over time. Surprisingly, genetic variability was regained to a level comparable to the large and outbreeding founder population only 15 generations after severe demographic bottlenecks. We suggest that the establishment of these populations could be a result of several mechanisms acting in synergy. Here, rapid increase in genetic diversity of few introductions could potentially be attributed to limited gene flow from adjacent populations, behavioural adaptations and/or even increased mutation rate. We present unique insights into genetic processes that point towards traits that are important for understanding species' invasiveness.
