    • The purpose of the European Stroke Organisation-Karolinska Stroke Update Conference is to provide updates on recent stroke therapy research and to give an opportunity for the participants to discuss how these results may be implemented into clinical routine. The meeting started 22 years ago as Karolinska Stroke Update, but since 2014 it is a joint conference with European Stroke Organisation. Importantly, it provides a platform for discussion on the European Stroke Organisation guidelines process and on recommendations to the European Stroke Organisation guidelines committee on specific topics. By this, it adds a direct influence from stroke professionals otherwise not involved in committees and work groups on the guideline procedure. The discussions at the conference may also inspire new guidelines when motivated. The topics raised at the meeting are selected by the scientific programme committee mainly based on recent important scientific publications. This year's European Stroke Organisation-Karolinska Stroke Update Meeting was held in Stockholm on 11-13 November 2018. There were 11 scientific sessions discussed in the meeting including two short sessions. Each session except the short sessions produced a consensus statement (Full version with background, issues, conclusions and references are published as web-material and at www.eso-karolinska.org and http://eso-stroke.org) and recommendations which were prepared by a writing committee consisting of session chair(s), scientific secretary and speakers. These statements were presented to the 250 participants of the meeting. In the open meeting, general participants commented on the consensus statement and recommendations and the final document were adjusted based on the discussion from the general participants Recommendations (grade of evidence) were graded according to the 1998 Karolinska Stroke Update meeting with regard to the strength of evidence. Grade A Evidence: Strong support from randomised controlled trials and statistical reviews (at least one randomised controlled trial plus one statistical review). Grade B Evidence: Support from randomised controlled trials and statistical reviews (one randomised controlled trial or one statistical review). Grade C Evidence: No reasonable support from randomised controlled trials, recommendations based on small randomised and/or non-randomised controlled trials evidence.

  • Avslut och fortsättning : En studie om övergången från introduktionsprogrammet språkintroduktion till nationellt program vid gymnasieskolan

    • This thesis highlights elements of the schooling for newly arrived students aged 16-19 within the Swedish school system in six different municipalities, where the transition between the language introduction programme and national programme at upper secondary school is central. The main purpose is to provide knowledge of pupils' experiences of the aforementioned transition. In this thesis, the concept of transition has a broader and more abstract meaning than just the actual move from one activity to another, and the thesis also focuses on the time spent in the language introduction programme and the first months in a national program at upper secondary school.Three studies are included in this thesis, where discourse analysis, curriculum theory and organizational theory of the school are used as a theoretical framework. The first study is an analysis of syllabuses from 1980 to 2011 for the subject Swedish as a second language within primary and secondary schools. The second is a study that interviews teachers in the language introduction programme, and the third study is based on interviews with pupils who have studied language introduction programme, but at the time of the interviews were studying in their first semester on a national programme.The analyses show that the transition is governed by curriculum and regulations that teachers and students must adhere to. The teachers carry out their everyday work on the basis of curriculum and regulations, such as the Education Act, but also on the basis of the individual school's organization. From an organizational perspective, this gives teachers limited personal room for manoeuvre. It also appears that the choice of national programmes and other choices that students can make within the framework of their schooling are sometimes obvious to the students and made by the students themselves. Nevertheless, other times teachers and study counsellors make the choices without the students’ knowledge. An additional result shows that the students say they experience a discrepancy when it comes to teaching of the various school subjects in the language introduction programme and the national programme at upper secondary school. The linguistic support that the students received during the time at the language introduction programme is not perceived by the students to be at the same level as the national programme. A development of linguistic support in various subjects within the national programme could, from a didactic perspective, lead to a less abrupt transition for the students

  • Volatile Emissions from Compressed Tissue

    • Since almost every fifth patient treated in hospital care develops pressure ulcers, early identification of risk is important. A non-invasive method for the elucidation of endogenous biomarkers related to pressure ulcers could be an excellent tool for this purpose. We therefore found it of interest to determine if there is a difference in the emissions of volatiles from compressed and uncompressed tissue. The ultimate goal is to find a non-invasive method to obtain an early warning for the risk of developing pressure ulcers for bed-ridden persons. Chemical analysis of the emissions, collected in compresses, was made with gas-chromatography – mass spectrometry and with a chemical sensor array, the so called electronic nose. It was found that the emissions from healthy and hospitalized persons differed significantly irrespective of the site. Within each group there was a clear difference between the compressed and uncompressed site. Peaks that could be certainly deemed as markers of the compression were, however, not identified. Nonetheless, different compounds connected to the application of local mechanical pressure were found. The results obtained with GC-MS reveal the complexity of VOC composition, thus an array of non-selective chemical sensors seems to be a suitable choice for the analysis of skin emission from compressed tissues; it may represent a practical instrument for bed side diagnostics. Results show that the adopted electronic noses are likely sensitive to the total amount of the emission rather than to its composition. The development of a gas sensor-based device requires then the design of sensor receptors adequate to detect the VOCs bouquet typical of pressure. This preliminary experiment evidences the necessity of studies where each given person is followed for a long time in a ward in order to detect the insurgence of specific VOCs pattern changes signalling the occurrence of ulcers.

  • The population genomic legacy of the second plague pandemic

    • Human populations have been shaped by catastrophes that may have left long-lasting signatures in their genomes. One notable example is the second plague pandemic that entered Europe in ca. 1,347 CE and repeatedly returned for over 300 years, with typical village and town mortality estimated at 10%–40%.1 It is assumed that this high mortality affected the gene pools of these populations. First, local population crashes reduced genetic diversity. Second, a change in frequency is expected for sequence variants that may have affected survival or susceptibility to the etiologic agent (Yersinia pestis).2 Third, mass mortality might alter the local gene pools through its impact on subsequent migration patterns. We explored these factors using the Norwegian city of Trondheim as a model, by sequencing 54 genomes spanning three time periods: (1) prior to the plague striking Trondheim in 1,349 CE, (2) the 17th–19th century, and (3) the present. We find that the pandemic period shaped the gene pool by reducing long distance immigration, in particular from the British Isles, and inducing a bottleneck that reduced genetic diversity. Although we also observe an excess of large FST values at multiple loci in the genome, these are shaped by reference biases introduced by mapping our relatively low genome coverage degraded DNA to the reference genome. This implies that attempts to detect selection using ancient DNA (aDNA) datasets that vary by read length and depth of sequencing coverage may be particularly challenging until methods have been developed to account for the impact of differential reference bias on test statistics.

  • Temporal Variation in Sindbis Virus Antibody Prevalence in Bird Hosts in an Endemic Area in Sweden

    • Sindbis virus (SINV) is a mosquito-borne bird virus that occasionally causes human disease in Fennoscandia, suggested to have cyclic 7-year intervals between outbreaks. Reliable data on human infections in Sweden is however lacking. Here we investigated the SINV antibody prevalence among birds in a Swedish area endemic to SINV to scrutinize if a cyclic variation in antibody prevalence is present in the natural host of SINV. Serum from birds were sampled in the summers of 2002-2004 and 2009 in the floodplains of the River Dalalven in central Sweden, with 2002 and 2009 representing hypothesized years of SINV outbreaks. A total of 963 birds from 52 species (mainly passerines) were tested for the presence of SINV antibodies using a plaque reduction neutralization test. The highest SINV antibody prevalence was found in Turdidae species, specifically Fieldfare, Redwing and Song thrush in which more than 70% of sampled individuals had antibodies to SINV in 2009. The SINV antibody prevalence significantly varied between years with 2% in 2002, 8% in 2003, 14% in 2004 and 37% in 2009. Antibodies were found equally often in hatchlings and in adults and increased from early to late in the season. Clearly, the SINV antibody prevalence was not elevated in the bird hosts in the predicted outbreak year 2002, thus solid evidence of a cyclic occurrence of SINV in Sweden is still lacking.

    • In this article, drift correction algorithms were used in order to remove linear drift in multivariate spaces of two data sets obtained by an electronic tongue based on voltammetry. The electronic tongue consisted of various metal electrodes (Au, Ir, Pt, Rh) combined with pattern recognition tools, such as principal component analysis. The first data set contained different types of liquid, from well defined to more complex solutions. The second data set contained different black and green teas. Component correction (CC) was compared to a simple additive correction. In CC, the drift direction of measured reference solutions in a multivariate space was subtracted from other types of solution. In additive correction, responses from reference samples were subtracted from other samples. CC showed similar or better performance in reducing drift compared to additive correction for the two data sets. The additive correction method was dependent on the fact that the differences in between samples of a reference solution were similar to the changes in between samples of other liquids, which was not the case with CC.

    • An electronic tongue based on voltammetry and a multichannel lipid membrane taste sensor based on potentiometry are compared using two aqueous examples: detergents and teas. The electronic tongue consists of four electrodes of different metals, a reference electrode and a counter electrode. The measurement principle is based on pulse voltammetry in which current is measured during the change of the amplitude of the applied potential. The taste sensor is based on eight different lipid/polymer membranes. The voltage difference between the electrodes and an Ag/AgCl reference electrode is measured when the current is close to zero. The responses from the two sensors systems are treated separately with multivariate data analysis based on principal component analysis and then merged to examine if further information could be extracted. It is shown that although the two sensor systems are about equal in separation ability in the two cases, extra information can be gained by combination of the two sensor systems. © 2001 Elsevier Science B.V.
