    • Snabbväxande regioner riskerar att hämmas i sin utveckling på grund av en bostadsmarknad som fungerar dåligt. I samhällsdebatten framhålls ofta att problemen står att finna i ett för lågt bostadsbyggande. I den här rapporten framtonar ett något annorlunda och mycket intressant perspektiv. Författarnas slutsats är att bostadsbristen inte enbart går att bygga bort. Istället är det ett ökat utbud av bostäder i det befintliga beståndet som är den stora utmaningen.

  • Do we see the same movement impairments? : the inter-rater reliability of movement tests for experienced and novice physiotherapists

    • Study design: Inter-rater reliability study. Background: Physiotherapists (PTs) use clinical tests including movement tests to identify faulty movement patterns. Aims: To investigate the inter-rater-reliability of active movement tests in the cervical spine, shoulder joint and scapulo-thoracic joint, and to describe the reasons for judgment of a positive test. Methodology: Four PTs, two experienced and two recently educated (novice), rated performance of five movement tests for 36 participants. Twenty-one of the participants were patients under treatment because of neck and/or shoulder problems, while 15 participants declared no problem from this region of the body. All tests were video recorded and the ratings were done by observing the video recordings. First, the PTs judged the tests as negative (the movement being ideally performed) or positive (the movement not being ideally performed). Then, the PTs described why the movements that they judged positive were not being ideally performed, using a predefined protocol, which represented different movement quality aspects. The inter-rater reliability was calculated for each test using Kappa statistics between the two experienced and the two novice PTs, respectively, and between each of the experienced and each of the novice PTs. Major findings: The experienced PTs had a higher inter-rater reliability than the novice PTs. The reasons for considering a movement test being positive differed highly between the (novice) PTs. Principal conclusion: This study supports previous studies concluding that the observation of active movement tests is reliable when assessed by experienced PTs. Novice PTs might benefit from further supervision.

  • Improved Methods for Characterization of Protein Dynamics by NMR spectroscopy and Studies of the EphB2 Kinase Domain

    • Proteins are essential for all known forms of life and in many lethal diseases protein failure is the cause of the disease. To understand proteins and the processes they are involved in, it is valuable to know their structures as well as their dynamics and interactions. The structures may not be directly inspected because proteins are too small to be visible in a light microscope, which is why indirect methods such as nuclear magnetic resonance (NMR) spectroscopy have to be utilized. This method provides atomic information about the protein and, in contrast to other methods with similar resolution, the measurements are performed in solution resulting in more physiological conditions, enabling analysis of dynamics. Important dynamical processes are the ones on the millisecond timeframe, which may contribute to interactions of proteins and their catalysis of chemical reactions, both of significant value for the function of the proteins.To better understand proteins, not only do we need to study them, but also develop the methods we are using. This thesis presents four papers about improved NMR techniques as well as a fifth where the kinase domain of ephrinB receptor 2 (EphB2) has been studied regarding the importance of millisecond dynamics and interactions for the activation process. The first paper presents the software COMPASS, which combines statistics and the calculation power of a computer with the flexibility and experience of the user to facilitate and speed up the process of assigning NMR signals to the atoms in the protein. The computer program PINT has been developed for easier and faster evaluation of NMR experiments, such as those that evaluate protein dynamics. It is especially helpful for NMR signals that are difficult to distinguish, so called overlapped peaks, and the soft- ware also converts the detected signals to the indirectly measured physical quantities, such as relaxation rate constants, principal for dynamics. Next are two new versions of the Carr-Purcell-Maiboom-Gill (CPMG) dispersion pulse sequences, designed to measure millisecond dynamics in a way so that the signals are more separated than in standard experiments, to reduce problems with overlaps. To speed up the collection time of the data set, a subset is collected and the entire data set is then reconstructed, by multi-dimensional decomposition co-processing. Described in the thesis is also a way to produce suitably labeled proteins, to detect millisecond dynamics at Cα positions in proteins, using the CPMG dispersion relaxation experiment at lower protein concentrations. Lastly, the kinase domain of EphB2 is shown to be more dynamic on the millisecond time scale as well as more prone to interact with itself in the active form than in the inactive one. This is important for the receptor function of the protein, when and how it mediates signals.To conclude, this work has extended the possibilities to study protein dynamics by NMR spectroscopy and contributed to increased understanding of the activation process of EphB2 and its signaling mechanism.

    • Two different numerical models for compression moulding of Sheet Moulding Compound are discussed and compared, with one being a more traditional flow model implemented in the 3DTimon commercial software, and the other being a more advanced flow model implemented in more general fluid dynamics software. The focus will be on comparing the resulting fibre orientations.

    • Sheet Moulding Compound (SMC) based composites have a large potential in industrial contexts due to the possibility of achieving comparatively short manufacturing times. It is however necessary to be able to numerically predict both mechanical properties as well as manufacturability of parts.In this paper a fully 3D, semi-empirical model based on fluid mechanics for the compression moulding of SMC is described and discussed, in which the fibres and the resin are modelled as a single, inseparable fluid with a viscosity that depends on volume fraction of fibres, shear strain rate and temperature. This model is applied to an advanced carbon-fibre SMC with a high fibre volume fraction (35%). Simulations are run on a model of a squeeze test rig, allowing comparison to experimental results from such a rig. The flow data generated by this model is then used as input for an Advani-Tucker type of model for the evolution of the fibre orientation during the pressing process. Numerical results are also obtained from the software 3DTimon. The resulting fibre orientation distributions are then compared to experimental results that are obtained from microscopy. The experimental measurement of the orientation tensors is performed using the Method of Ellipses. A new, automated, accurate and fast method for the ellipse fitting is developed using machine learning. For the studied case, comparison between the experimental results and numerical methods indicate that 3D Timon better captures the random orientation at the outer edges of the circular disc, while 3D CFD show larger agreement in terms of the out-of-plane component. One of the advantages of the new image technique is that less work is required to obtain microscope images with a quality good enough for the analysis.

    • A numerical model for compression moulding of Sheet Moulding Compound is presented. The model is based on fluid mechanics and the SMC charge is modelled as a fluid with a viscosity that is dependent on the charge temperature, the fibre volume fraction of the material and the shear strain rate. Trends observed in the simulations regarding the shape of the flow front agree with experimental observations in previous studies. The simulations also yield that the type and magnitude of initial heating effects the initial flow to a large extent.

    • A review of the numerical modeling of the compression molding of the sheet molding compound (SMC) is presented. The focus of this review is the practical difficulties of modeling cases with high fiber content, an area in which there is relatively little documented work. In these cases, the prediction of the flows become intricate due to several reasons, mainly the complex rheology of the compound and large temperature gradients, but also the orientation of fibers and the micromechanics of the interactions between the fluid and the fibers play major roles. The details of this during moldings are discussed. Special attention is given to the impact on viscosity from the high fiber volume fraction, and the various models for this. One additional area of interest is the modeling of the fiber orientation.
